Precision medicine in pantothenate kinase-associated neurodegeneration

Neurodegeneration with brain iron accumulation is a broad term that describes a heterogeneous group of progressive and invalidating neurologic disorders in which iron deposits in certain brain areas,mainly the basal *** predominant clinical symptoms include spasticity,progressive dystonia,Parkinson's disease-like symptoms,neuropsychiatric alterations,and retinal *** the neurodegeneration with brain iron accumulation disorders,the most frequent subtype is pantothenate kinase-associated neurodegeneration(PKAN) caused by defects in the gene encoding the enzyme pantothenate kinase 2(PANK2)which catalyzed the first reaction of the coenzyme A biosynthesis *** there is no effective treatment to prevent the inexorable course of these *** aim of this review is to open up a discussion on the utility of using cellular models derived from patients as a valuable tool for the development of precision medicine in ***,we have described that dermal fibroblasts obtained from PKAN patients can manifest the main pathological changes of the disease such as intracellular iron accumulation accompanied by large amounts of lipofuscin granules,mitochondrial dysfunction and a pronounced increase of markers of oxidative *** addition,PKAN fibroblasts showed a morphological senescence-like ***,pantothenate supplementation,the substrate of the PANK2 enzyme,corrected all pathophysiological alterations in responder PKAN fibroblasts with low/residual PANK2 enzyme ***,pantothenate treatment had no favourable effect on PKAN fibroblasts harbouring mutations associated with the expression of a truncated/incomplete *** correction of pathological alterations by pantothenate in individual mutations was also verified in induced neurons obtained by direct reprograming of PKAN *** observations indicate that pantothenate supplementation can increase/stabilize the expression levels of PANK2 in s

Biogenic hydroxyapatites from mammalian bones naturally contain traces of ions, like Mg, which play a vital role in the bone remodeling process. In this way, synthetic hydroxyapatites should include this kind of mineral. In this work, hydroxyapatite added with Mg was synthesized by wet precipitation using（NH;）H;PO;, Ca（NO;）;.4 H;O,and Mg（NO;）;.6 H;O（0, 0.032, 0.061, and 0.123 M）. Inductively coupled plasma and X-ray diffraction evidenced the Mg inclusion in the hydroxyapatite lattice in the same levels reported for natural hydroxyapatites. The calcination at 600℃ was performed to remove the reaction by-products, but it also gave rise to physicochemical changes as the coalescence and crystals recrystallization. It produced, in turn, an improvement in the crystalline quality, according to Raman analysis. despite those physicochemical changes, all the samples remained nanometric according to scanning electron microscopy imaging.

Effect of BaF2 addition on luminescence properties of Er3+/Yb3+ co-doped phosphate glasses

Er3+/Yb3+co-doped phosphate glasses（P2O5-Al2O3-BaO-BaF2-K2O-Er2O3-Yb2O3） with varying BaF2content,were prepared by a conventional melt quenching technique and their spectroscopic properties were examined through the Raman, absorption, emission and decay measurements. Raman spectra(350-1400 cm-1) of the Er3+/Yb3+co-doped phosphate glasses with varying BaF2content, were recorded upon laser excitation at 785 nm. Near infrared luminescence spectra were measured in the1400-1600 nm region under 970 nm diode laser excitation and characteristic band was observed at1533 nm corresponding to4Ⅰ13/2→4Ⅰ15/2transition of Er3+ion. The decay curves for the4Ⅰ13/2level of Er3+ion, were measured and the lifetime is found to decrease from 7.94 to 7.70 ms when BaF2content increases from 0 to 8 mol% and then increases up to 7.83 ms with further increase in BaF2content（12 mol%）. The emission *** and figure of merit for the4Ⅰ13/2→4Ⅰ15/2transition of Er3+ion were evaluated and compared to the other host matrices. The upconversion luminescence was measured and intense red emission was observed for all the studied samples.

Nano to micro size transition of hydroxyapatite in porcine bone during heat treatment with low heating rates

This work focuses on the changes of raw hydroxyapatite obtained from porcine bones during heat treatment with heating rates of 2.5 and 5.0 °C/min between 600 °C and 1100 °C. High-Resolution transmission electron microscopy(HR-TEM) images showed that raw hydroxyapatite was formed by high-ordered nanometric platelet crystals with a length of around 18 nm and width around 7 nm. High-Resolution scanning electron microscopy(HR-SEM) images evidenced the coalescence phenomena during heating giving. It was found that the growth of the crystals and the transition from nano to a micro-size occurred at around 700 °C. And the changes in the full width at half maximum(FWHM) for the characteristic(002) plane of the X-ray diffraction patterns and the FWHM of the characteristic Raman band at 960 cm-1 for ν1 PO43-exhibited the transition from nano to a micro size, that was confirmed by HR-SEM images and the thermogravimetric(TG) analysis. The porosity of the BIOHAp changes from intra nanometric to intercrystalline.

The protein apoptoUc protease activating factor I （Apafl） is the central component of the apoptosome, a multiprotein complex that activates procaspase-9 after cytochrome c release from the mitochondria in the intrinsic pathway of apoptosis. We have developed a vital method that allows fluorescence-activated cell sorting of cells at different stages of the apoptotic pathway and demonstrated that upon pharmacological inhibition of Apafl, cells recover from doxorubicinor hypoxia-induced early apoptosis to normal healthy cell. Inhibiting Apafl not only prevents procaspase-9 activation but delays massive mitochondrial damage allowing cell recovery.

Synthesis and Thermal Behavior of Metallic Cobalt Micro and Nanostructures

In this contribution, a comparative study of metallic cobalt micro and nanoparticles obtained in solution by four different chemical routes is reported. Classic routes such as borohydride reduction in aqueous media and the so-called polyol methodology were used to obtain the cobalt nanostructures to be studied. Using CTAB as surfactant, cobalt hollow nanostructures were obtained. The use of strong reducing agents, like sodium borohydride, favors the formation of quasi-monodispersed nanoparticles of about 2 nm size but accompanied with impurities; for hydrazine(a mild reducer), nanoparticles of larger size are obtained which organize in spherical microagglomerates. Valuable information on the particles thermal stability and on nature of the species anchored at their surface was obtained from thermogravimetric curves. The samples to be studied were characterized from UV-vis, IR, X-ray diffraction, and electron microscopy images(scanning and transmission).

Phylogenetic endemism of the orchids of Megamexico reveals complementary areas for conservation

Orchid diversity provides a unique opportunity to further our understanding of biotic and abiotic factors linked to patterns of richness,endemism,and phylogenetic endemism in many ***,orchid diversity is consistently threatened by illegal trade and habitat ***,we identified areas critical for orchid conservation in the biogeographic province of *** this purpose,we evaluated orchid endemism,phylogenetic diversity,and phylogenetic endemism within Megamexico and characterized orchid life *** results indicate that the majority of the regions with the highest estimates of endemism and phylogenetic endemism are in southern Mexico and northern Central America,mostly located on the Pacific side of *** the most important orchid lineages,several belong to epiphytic lineages such as Pleurothallidinae,Laeliinae and *** also found that species from diverse and distantly related lineages converge in montane forests where suitable substrates for epiphytes ***,the southernmost areas of phylogenetic diversity and endemism of Megamexico are in unprotected ***,we conclude that the most critical areas for orchid conservation in Megamexico are located in southern Mexico and northern Central *** recommend that these areas should be given priority by the Mexican system of natural protected areas as complementary conservation areas.

Auxin Is Required for Valve Margin Patterning in Arabidopsis After All

Fruits are important as a nutritional source, but biologically more important are their seed protection and dispersal funcions. Dry fruits can be dehiscent and the process of opening of ripe fruits, or pod shattering, causes the seeds to be dispersed （Ferrandiz, 2002）. This process occurs in many crop species, for instance of members of the Brassicaceae family. Arabidopsis thaliana, the model plant species in the laboratory, belongs to this family and also presents pod shattering （Figure 1）. The Arabidopsis fruit is a silique, and before fertilization it is called a gynoecium or pistil. The gynoecium is formed by two carpels that are fused, and the replum is present at their margins and is connected to the septum that divides the silique inside. In between each valve-replum border, the valve margin or dehiscence zone is formed. When the fruit matures, a separation layer and a lignified cell layer are formed in the dehiscence zone. In addition, a lignified layer is formed in the valves, and when the mature fruit starts to desiccate, tension is created, which in association with the effect of middle lamella degrading enzymes in the separation layer, results in shattering of the fruit （Ferrandiz, 2002）. Research has led to the proposal of a model for the gene regulatory network that explains valve margin formation in Arabidopsis （e.g., Ch＆vez Montes et al., 2015）. Mutations in various members of this gene regulatory network result in indehiscent fruits, i.e., the fruit does not open or shatter. One of these genes is the bHLH transcription factor INdeHISCENT （IND）, which is necessary for the formation of the separation layer and the lignified cell layers （Liljegren et al., 2004）. IND has also been related to auxin, more precisely, to auxin distribution, important for gynoecium and fruit patterning （Sorefan et al., 2009; Girin et al., 2011）.
