Poly(ADP-ribose)(PAR)is a highly negatively charged polymer.PAR is synthesized by poly(ADP-ribose)polymerases(PARPs)and is involved in the assembly and stabilization of macromolecular complexes.Here,the presence and putative roles of poly(ADP-ribosyl)ation(PARylation)associated to adherens junctions(AJ)and the actin cytoskeleton in epithelial and Schwann cells,is reviewed.The hypothesis generated by analogy,stating that PAR is associated to AJ in other cell types,is postulated.According to this hypothesis,PAR associated to puncta adherentia in chemical synapses would participate in plasticity,learning and memory.In turn,PAR associated to fascia adherens in cardiomyocytes,would affect heart beating.PARP inhibitors are currently under development and clinical testing.Basic research in different tissues will probably influence their clinical uses.

Post-translational modifications （PTMs） chemically and physically alter the properties of proteins, including their folding, subcellular localization, stability, activity, and consequently their function. In spite of their relevance, studies on PTMs in plants are still limited. Small Ubiquitin-like Modifier （SUMO） modification regulates several biological processes by affecting protein-protein interactions, or changing the subcellular Iocalizations of the target proteins. Here, we describe a novel proteomic approach to identify SUMO targets that combines 2-D liquid chromatography, immunodetection, and mass spectrometry （MS） analyses. We have applied this approach to identify nuclear SUMO targets in response to heat shock. Using a bacterial SUMOylation system, we validated that some of the targets identified here are, in fact, labeled with SUMO1. Interestingly, we found that GIGANTEA （GI）, a photoperiodic-pathway protein, is modified with SUMO in response to heat shock both in vitro and in vivo.

Nutrient flux,net metabolism,and N2 fixation and denitrification processes were estimated and compared in two semi-arid subtropical coastal lagoons in the Gulf of California:El Soldado(ES),where no wastewater is discharged,and El Rancho(ER),where shrimp farm effluents are regularly discharged.Biogeochemical processes were evaluated with the LOICZ model.Flushing time wasdenitrification in the summer.denitrification dominated in ER(−2.21 mmol m^(2)day^(−1))and values were higher than those estimated in ES(−0.45 mmol m^(2)day^(−1)).The comparative analysis between ES and ER evidenced the significant changes in its biogeochemical performance caused by the input of anthropogenic nutrients,and can orient the environmental management of coastal lagoons.

Progression of chronic lymphocytic leukemia(CLL)is determined by the localization of malignant cells in lymphoid tissues,where they receive growth and survival signals.CLL cells produce angiogenic factors that are regulated by internal and external stimuli and whose levels vary according to the clinical stage of the disease.Stromal cellular and molecular components in CLL niches disturb the balance of pro-and antiangiogenic molecules in CLL cells and induce an angiogenic switch.Additionally,CLL cells also influence the behavior of microenvironmental cells,inducing endothelial cell proliferation and increasing the angiogenic capacity of macrophages,neutrophils,and other cells present in CLL niches.As a result of these reciprocal functional interactions,bone marrow angiogenesis is frequently increased in CLL and has been proposed as a prognostic marker in early disease.Besides their role in regulating angiogenesis,angiogenic factors are also involved in CLL cell migration and survival,all contributing to disease progression.Angiogenic factors,particularly vascular endothelial growth factor,have therefore been attractive therapeutic targets in CLL and many clinical trials were established in the past years.However,the results of these trials reveal that anti-angiogenic therapies alone are not as efficient as expected and should rather be used in combination with other treatments.

Objective: To report the renal histological lesions in synanthropic rodents, Mus musculus and Rattus rattus, naturally infected with Leptospira spp., captured in a rural community in Yucatan, Mexico. Methods: Kidney samples of synanthropic rodents were collected from a rural community in Yucatan, Mexico. Polymerase chain reaction was used to detect Leptospira spp. infection. Tissue kidney was fixed in 10% buffered formalin, processed according to the usual techniques for paraffin inclusion, cut and stained with hematoxylin and eosin, and examined using a conventional electronic microscope. Results: A total of 187 rodents were captured. Nine individuals(4.8%) were positive for Leptospira spp. in the molecular analysis. All renal lesions observed in the histopathological study had been reported previously for Leptospira spp. infection. Conclusions: The histopathological lesions are present in the kidneys, plus the results of the polymerase chain reaction confirm that these rodents are true carriers of Leptospira spp.

Single-particle electron microscopy has now reached maturity, becoming a commonly used method in the examination of macromolecular structure. Using a small amount of purified protein, isolated molecules are observed under the electron microscope and the data collected can be averaged into a 3D reconstruction. Single-particle electron microscopy is an appropriate tool for the analysis of proteins that can only be obtained in modest quantities, like many of the large complexes currently of interest in biomedicine. Whilst the use of electron microscopy expands, new methods are being developed and improved to deal with further challenges, such as reaching higher resolutions and the combination of informa tion at different levels of structural detail. More importantly, present methodology is still not robust enough when studying certain "tricky" proteins like those displaying extensive conformational flexibility and a great deal of user expertise is required, posing a threat to the consistency of the final structure. This mini review describes a brief outline of the methods currently used in the 3D analysis of macromolecules using single-particle electron microscopy, intended for those first approaching this field. A summary of methods, techniques, software, and some recent work is presented. The spectacular improvements to the technique in recent years, its advantages and limitations compared to other structural methods, and its future developments are discussed.

Background:Tropical montane cloud forests are one of the most important hotspots on Earth and show presence of relict-endemic and endangered species,representing about 14%of the total tropical forest worldwide.Synchronous seed production or masting in tropical montane cloud tree species is a widespread reproductive strategy of decidu-ous and evergreen broad-leaved tree associations to decrease costs of reproduction and ensure offspring.Masting event maintains a high avian diversity,which can be modified by phenological process(seed production and non-seed production).Methods:The main aim of this study was to assess alpha and beta avian diversity and whether the composition of the trophic guild modifies among phenological processes and between two fragmented relict-endangered Mexican Beech(Fagus grandifolia subsp.mexicana)forests(Medio Monte and El Gosco)in the Mexican state of Hidalgo.In addi-tion,we evaluated beechnut production.Results:We recorded 36 bird species,11 of them included in some conservation risk status,and 5 endemic species.Alpha diversity values were dissimilar in avian richness(q=0)among phenological processes and between frag-mented beech forests.Avian communities among three phenological processes and between fragmented forests were structurally similar,dominated during immature seeds the Brown-backed Solitaire(granivores-insectivores-frugivores);during mature seeds the White-crowned Parrot(Pionus senilis,granivores-frugivores);and the Dwarf Jay(Cyanolyca nana,insectivores)was abundant during low seed quality.The complementarity index was high among phenological processes and low between forests.We found a high bird turnover value between immature seeds—mature seeds and during mature seeds—low seed quality.Furthermore,a similar pattern was recorded between the two study forests.Seed production showed a high number of undamaged beechnuts in Medio Monte,while in El Gosco beechnuts were attacked by insects.Conclusions:Our results reflect that masting phenological process and contrasting study fore

Marine mammals undergo cycles of tissue ischemia and reperfusion during the dive response.Reperfusion injury can result in oxidative tissue damage and the activation of a pro-inflammatory immune response.The risk of oxidative damage is reduced by antioxidants.Our hypothesis is that the reported higher antioxidant defenses within marine mammal tissues provide additional protection in situations that produce oxidative stress,like inflammation,in comparison to terrestrial mammal tissues.Leukocytes were isolated from the whole blood of Pacific bottlenose dolphins(Tursiops truncatus gilli)and humans(Homo sapiens)and were exposed to lipopolysaccharides(LPS,10μg/mL)in vitro to simulate a pro-inflammatory challenge.Oxidative stress indicators,including superoxide radical(O_(2)^(•−))production,activities of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GPx),glutathione reductase(GR),and glutathione S-transferase(GST),as well as oxidative protein damage,were quantified by spectrophotometry.Following 48 h under experimental conditions,bottlenose dolphin leukocytes produced 1.9 times more O_(2)^(•−)but displayed 2.0 times lower protein carbonyl concentrations compared to human leukocytes.Following 48 h under experimental conditions,bottlenose dolphin leukocytes displayed 7.9,2.0,11.1,and 3.3 times more activities of CAT,GPx,GR,and GST,respectively,compared to human leukocytes.These results suggest that,under cell culture conditions,the antioxidant defenses in bottlenose dolphin leukocytes provide additional protection against pro-inflammatory challenges in comparison to human leukocytes,likely as an adaptive advantage.

AIM To investigate if mutations in TCF7 L2 are associated with "atypical diabetes" in the Uruguayan population.METHODS Healthy, nondiabetic controls(n = 133) and patients with type 2 diabetes(n = 177) were selected from among the presenting population at level-3 referral healthcare centers in Uruguay. Patients with type 2 diabetes were subgrouped according to "atypical diabetes"(n = 92) and "classical diabetes"(n = 85). Genotyping for the rs12255372 and rs7903146 single nucleotide polymorphisms(SNPs) in the TCFTL2 gene was carried out with Taq Man? probes. Random samples were sequenced by Macrogen Ltd.(South Korea). Statistical analysis of the SNP data was carried out with the SNPStats online tool(http://***/SNPstats). The best inheritance model was chosen according to the lowest values of Akaike's information criterion and Bayesian information criterion. Differences between groups were determined by unpaired t-tests after checking the normal distribution or were converted to normalize the data. The association of SNPs was tested for matched case-control samples by using χ2 analysis and calculation of odds ratios(ORs) with 95% confidence intervals(CIs). All statistical tests were performed using SPSS v10.0 and EpiI nfo7 statistical packages. Significant statistical differences were assumed in all cases showing adjusted P del was the dominant type: rs12255372 = G/G vs G/T+T/T, OR = 0.63, 95%CI: 0.40-0.98, P < 0.05 and rs7903146 = C/C vs C/T+T/T, OR = 0.79, 95%CI: 0.41-1.55, P = 0.3. The rs12255372 SNP showed high association with the type 2 diabetes cases(OR = 1.60, 95%CI: 1.20-2.51, P < 0.05).
