The protein apoptoUc protease activating factor I （Apafl） is the central component of the apoptosome, a multiprotein complex that activates procaspase-9 after cytochrome c release from the mitochondria in the intrinsic pathway of apoptosis. We have developed a vital method that allows fluorescence-activated cell sorting of cells at different stages of the apoptotic pathway and demonstrated that upon pharmacological inhibition of Apafl, cells recover from doxorubicinor hypoxia-induced early apoptosis to normal healthy cell. Inhibiting Apafl not only prevents procaspase-9 activation but delays massive mitochondrial damage allowing cell recovery.

The Gulf of Cadiz is situated geologically at the Gibraltar Arc, the westernmost arc of the Alpine- Himalayan orogenic belt. Based on extensive previous studies that include swath bathymetry, multi-channel and very high-resolution seismic reflection, gravime- try, magnetism, heat flow probes, and underwater photography surveys, more than 500 polymetallic nodules were collected at water depths ranging from 850 to 1000 m, associated with hydrocarbon-derived carbonate chimneys, slabs, and crusts. Nodules show a wide range of sizes, densities, weights and mor- phologies. Nodules are composed of multiple milli- metre-thick layers of Fe and Mn oxyhydroxides sur- rounding a nucleus composed of Early-Middle Mio- cene plastic marls, which were expulsed from underly- ing units by fluid venting. Dialysis plays a crucial role in the purification of nanomaterials but its impact on the structural properties of carbon nanomaterials was never ***,a carbon-based nanomaterial generated electrochemically in potassium phosphate buffer,was characterized before and after dialysis against pure *** is shown that dialysis affects the size of the carbon domains,structural organization,surface functionalization,oxidation degree of carbon,and grade of ***,dialysis drives the nanomaterial organization from discrete roundish carbon domains,with sizes ranging from 70 to 160 nm,towards linear stacking structures of small nanoparticles(de)surface groups evolve into more oxidized carbon groups(e.g.,ketone and ester groups).Investigation of the as-prepared nanomaterial by electron paramagnetic resonance(EPR)revealed a resonance signal consistent with carbon-oxygen centred ***,this study brings to light the selective affinity of the carbon nanomaterial under study to capture Na^(+)ions,a property greatly enhanced by the dialysis process,and its high ability to trap oxygen,particularly before *** findings open new perspectives for the application of carbon-based nanomaterials and raise awareness of the importance of structural changes that can occur during the purification of carbon-based nanomaterials.

Molecular signaling of 5-HT<sub>1A</sub>and presence of serotonergic cells in the fetal cerebral cortex

Early appearance of the serotonergic system in the fetal brain and the various effects of serotonin (5-HT) on brain morphogenesis, have given support to a neurotrophic role of serotonin. This function of serotonin is accomplished through a system of serotonin nerve terminals in the target regions that involves various 5-HT receptors. In visual, auditory and somatosensory cortex an early and intense serotonergic innervation is particularly important. The neuronal somata of these terminals are normally located in the mesencephalon and they have not been observed in the maturing cerebral cortex, neither in the adult brain. By using immunolabeling techniques, fluorescence and confocal microscopy, we observe the presence of both, 5-HT terminals and 5-HT cells in mesencephalon (Me, E17) and in the neopallium (Np, E13-E16) cocultures. Cells immunopositive to 5-HT and to tryptophan-5-hydroxilase are also observed in the Np on day 12 of culture. These results concerning the unexpected presence of serotonergic cells in the fetal cerebral cortex are interesting and may be of importance in corticogenesis. As it happens with other elements of the serotonergic system, the presence of these phenotypically serotonergic cells in the early cerebral cortex may be transitory and probably supporting cortex maturation processes. The molecular signaling path of the 5-HT1A receptor has also been identified.

Pigmented rice varieties are rich in antioxidant and anti-inflammatory compounds(*** and proanthocyanidins).Therefore,their consumption could exert beneficial effects,particularly in people suffering from chronic diseases(e.g.,celiac disease).Pigmented rice is commonly consumed as brown rice,but technological treatments could be applied to reduce its cooking time and improve its nutritional value(vitamins and minerals).In this study,two relatively new pigmented varieties(Violet and Orange)were characterized in terms of phenolic content and antioxidant capacity and the impact of two technological treatments(e.g.,milling and parboiling)on their phytochemical composition was *** pigmented and one non-pigmented Italian varieties were included for *** technological processes affected the concentration of phenolic compounds and their relative antioxidant *** milling mainly reduced the phenolic content and the antioxidant activity,anthocyanins seem to be more affected by parboiling(reduction of 91.5%).despite the effects of technological treatments on active compounds,pigmented varieties still represent an interesting antioxidant source when compared to the non-pigmented ones.

Objective: To compare the presence of Mycoplasma spp. in pharyngeal swabs of patients with asthma, allergic rhinitis and controls, also to compare the reactivity to allergens in the three groups. Material and Methods: Throat swabs of 120 patients with asthma, 211 allergic rhinitis patients and 171 healthy persons were cultured in E media. Positive cultures were identified by PCR. Susceptibility of patients positive for isolation of Mycoplasma spp. to indoor, forest and undergrowth allergens were determined. Results: Mycoplasma spp. was isolated in 26.6% of samples from asthmatic patients, 36% in patients with allergic rhinitis and 2.9% in the group of healthy persons. Positive patients for Mycoplasma with asthma and allergic rhinitis showed susceptibility tomite, grass and thunder mainly. Conclusion: There is a higher incidence of Mycoplasma spp. in asthmatic and allergic rhinitis patients than healthy persons. Only patients with asthma or allergic rhinitis showed reactivity to environmental allergens.

This study compares the kinetics of adherence of both the prototype strain PG-18 and the recently clinically isolated strain P-140 of Mycoplasma fermentans to HEp-2 cells. M. fermentans may induce respiratory and genitourinary tract infections. We investigated the presence of putative adhesins from strain P-140. A surface protein of 47 kDa, labeled SP47, isolated from M. fermentans P-140, seems to be involved in acterial adherence on HEp-2 cells;this study could allow helping understand the interaction between mycoplasmas and their hosts. Anti-SP47 antibodies inhibited the formation of bacterial clusters and adherence to cultured cells, quantified by ELISA. M. fermentans P-140 was more efficient at adhering to cultured HEp-2 cells than PG-18.

Individual male rats may systematically display or not copulatory behavior when paired with receptive females. Although these phenotypes are associated with differences in brain organization and function, they might also do so at the level of the reproductive organs. We then used high performance liquid chromatography to quantify serotonin concentration and the activity of tryptophan hydroxylase in the reproductive organs of copulator and non-copulator males. Sexual behavior display was compared between groups and parameters of fertility and reproductive fitness were determined for copulator males. Copulator males had higher concentrations of serotonin in the epididymis, testicle and ventral prostate than their non-copulator counterparts, as it was found for epididymal and testicular tryptophan hydroxylase activity. However, preliminary data shows that serotonin elevation occurs in copulator males only until they have accumulated several sexual encounters, so it might be a response to genital gratification or sexual rewarding. Interestingly, only epididymal serotonin concentration correlated with reproductive fitness, offspring number, mating success and seminal plug volume in copulator males. Our results support that copulator and non-copulator male rats feature a phenotype-specific serotoninergic tone in the epididymis, testicle and ventral prostate gland. The observation documenting that epididymal serotonin concentration correlated with parameters that monitor male fertility and reproductive fitness in copulator males predicts that epididymal factors increase their chances of parenting offspring.

Breast cancer is a complex and heterogeneous disease with different clinical outcomes. The investigations of new biomolecular markers are essential to know the prognosis and improve the clinical management of patients. The SIRT-1 （sirtuin- 1） is a histone deacetylase implicated in various epigenetic critical functions for the cells and the maintenance of genomic stability. The objective of this study is to investigate the grade of expression of the SIRT-I （sirtuin-1） and the prognostic value in overall survival of women with breast cancer. Retrospective cohort of 457 women with breast cancer has been researched, undergoing treatment in Erechim-RS from 2003 to 2013 and followed until July 2015. The degree of SIRT-1 expression was investigated by immunohistochemistry in 123 patients （26.9%） of the cohort. The OS （overall survival） from specific disease and risk of death from breast cancer were estimated by Kaplan-Meier and Cox＇s proportional risks. The median age of 57.4 years cohort with OS of 79.6% in 5 years and 69.1% at 10 years, with follow-up time of 61.9 months are revealed in this work. The SIRT-1 overexpression was found in 6.5% of cases and characterized a subgroup of women with shorter survival and increased risk of death from breast cancer （HR = 2.66; 95% CI 1.03 to 6.86; p = 0.043） and adjusted by age （HR = 2.86; 95% CI 1.11 to 7.38; p = 0.030）, histology （HR = 2.79; 95% CI 1.07 to 7.28; p = 0.036）, lymph nodes （HR = 2.73; 95% CI 1.06 to 7.04; p = 0.037）, Her-2 （HR = 2.82; 95% CI 1.07 to 7.44; p = 0.036）; chemotherapy （HR = 2.90; 95% CI 1.11 to 7.60; p = 0.030） and radiotherapy （HR = 2.71; 95% CI 1.05 to 7.01; p = 0.040）. In regressive multivariate models adjusted for age, status of axillary lymph nodes, Her-2 expression and proliferation index （Ki-67）, the grade of expression of the SIRT-1 maintained association with poor prognosis.
