Background Commercial diets are frequently formulated to meet or exceed nutrient levels including those of lim-iting essential amino acids(AA)covering potential individual variations within the ***,the provision of dietary excess of AA,such as Lys,may lead to reduced appetite and growth in *** mechanisms modulat-ing these responses have not been extensively *** study evaluated the effect of Lys dietary excesses on performance and satiety biomarkers in post weaning *** Twenty-four pigs aged 21 d and weighing 6.81±0.12kg(mean±SEM)were individually housed and offered 1 of 4 dietary treatments for 3weeks:a diet containing a standardized ileal digestible Lys reaching 100%(T0),120%(T1),150%(T2)or 200%(T3)of the NRC(2012)*** the end of the experiment,blood samples from the cephalic vein of the T0 and T3 groups were obtained for AA *** addition,primary intestinal cultures from T0 pigs were used,following their humane killing,to evaluate the effect of Lys on gut hormone secretion and AA sensors gene expression under ex vivo *** Feed intake was linearly reduced(P<0.001)and the weight gain to feed ratio reduced(P<0.10)with increased dietary levels of Lys during the third-and first-week post weaning,*** con-centration(P<0.05)and the metabotropic glutamate receptor 1 and the solute carrier family 7 member 2(P<0.10)gene expression was enhanced in proximal jejunum tissues incubated with Lys at 20mmol/L when compared to the control(Lys 0mmol/L).Plasma Lys and Glu(P<0.05)concentration increased in the T3 compared to T0 *** contrast,plasma levels of His,Val,Thr,Leu(P<0.05)and Gln(P<0.10)were lower in T3 than T0 *** The present results confirm that excess dietary Lys inhibits hunger in ***,the results provide evidence of pre-and post-absorptive mechanisms modulating these *** dietary excesses should be nar-rowed,when possible,to avoid negative effects of

This paper studies the optimization problem of heterogeneous networks under a timevarying *** agent only accesses to one local objective function,which is *** improved algorithm with noisy measurement of local objective functions' sub-gradients and additive noises among information exchanging between each pair of agents is designed to minimize the sum of objective functions of all *** weaken the effect of these noises,two step sizes are introduced in the control *** graph theory,stochastic analysis and martingale convergence theory,it is proved that if the sub-gradients are uniformly bounded,the sequence of digraphs is balanced and the union graph of all digraphs is joint strongly connected,then the designed control protocol can force all agents to find the global optimal point almost *** last,the authors give some numerical examples to verify the effectiveness of the stochastic sub-gradient algorithms.

Here,we aimed to study the changes in proteome of golden pompano fillets during post-mortem *** mass tags(TMT)-labeled quantitative proteomic strategy was applied to investigate the relationships between protein changes and quality characteristics of modified atmosphere packaging(MAP)fillets during superchilling(-3°C)*** electron microscopy was used to show that the muscle histology microstructure of fillets was damaged to varying degrees,and low-field nuclear magnetic resonance was used to find that the immobilized water and free water in the muscle of fillets changed *** sulfhydryl content,TCA-soluble peptides and Ca2+-ATPase activity also showed that the fillet protein had a deterioration by oxidation and *** Fresh(FS),MAP,and air packaging(AP)groups were *** of 150 proteins were identified as differential abundant proteins(DAPs)in MAP/FS,while 209 DAPs were in AP/FS *** KEGG pathway analysis indicated that most DAPs were involved in binding proteins and protein *** analysis found that 52 DAPs were correlated with quality *** them,8 highly correlated DAPs are expected to be used as potential quality markers for protein oxidation and water-holding *** results provide a further understanding of the muscle deterioration mechanism of packaging golden pompano fillets during superchilling.

Popular fermented golden pomfret(Trachinotus ovatus)is prepared via spontaneous fermentation;however,the mechanisms underlying the regulation of its flavor development remain *** study shows the roles of the complex microbiota and the dynamic changes in microbial community and flavor compounds during fish ***-molecule real-time sequencing and molecular networking analysis revealed the correlations among different microbial genera and the relationships between microbial taxa and volatile *** underlying flavor development were also elucidated via KEGG based functional ***,Shewanella,and Staphylococcus were the dominant microbial ***-nine volatile compounds were detected in the fermented fish samples,with thirteen identified as characteristic volatile compounds(ROAV>1).Volatile profiles resulted from the interactions among the microorganisms and derived enzymes,with the main metabolic pathways being amino acid biosynthesis/metabolism,carbon metabolism,and glycolysis/*** study demonstrated the approaches for distinguishing key microbiota associated with volatile compounds and monitoring the industrial production of high-quality fermented fish products.

Pre-mating nitenpyram exposure in male mice leads to depression-like behavior in offspring by affecting tryptophan metabolism in gut microbiota

Several studies have confirmed that the health status of the paternal affects the health of the offspring,however,it remains unknown whether paternal exposure to pesticides affect the offspring ***,we used untargeted metabolomics and 16S rRNA sequencing technology,combined with tail suspension test and RT-qPCR to explore the effects of paternal exposure to nitenpyram on the neurotoxicity of *** results found that the paternal exposure to nitenpyram led to the offspring’s depressive-like behaviors,accompanied by the reduction of tryptophan content and the disorder of microbial abundance in the gut of the ***,we determined the expression of tryptophan metabolism-related genes tryptophanase(tnaA)and tryptophan hydroxylase 1(TpH1)in gut bacteria and colonic *** found that tryptophan is metabolized to indoles rather than being absorbed into colonocytes,which coursed the reduce of tryptophan availability after nitenpyram *** conclusion,our study deepens our understanding of the intergenerational toxic effects of pesticides.

In this study,the structural characters,antioxidant activities and bile acid-binding ability of sea buckthorn polysaccharides(HRPs)obtained by the commonly used hot water(HRP-W),pressurized hot water(HRP-H),ultrasonic(HRP-U),acid(HRP-C)and alkali(HRP-A)assisted extraction methods were *** results demonstrated that extraction methods had significant effects on extraction yield,monosaccharide composition,molecular weight,particle size,triple-helical structure,and surface morphology of HRPs except for the major linkage *** analysis showed that HRP-U with filamentous reticular microstructure exhibited better thermal *** HRP-A with the lowest molecular weight and highest arabinose content possessed the best antioxidant ***,the rheological analysis indicated that HRPs with higher galacturonic acid content and molecular weight showed higher viscosity and stronger crosslinking network(HRP-C,HRP-W and HRP-U),which exhibited stronger bile acid binding *** present findings provide scientific evidence in the preparation technology of sea buckthorn polysaccharides with good antioxidant and bile acid binding capacity which are related to the structure affected by the extraction methods.

Dendrobium officinale polysaccharide attenuates type 2 diabetes in mice model by modulating gut microbiota and alleviating intestinal mucosal barrier damage

The purpose of this study was to investigate the hypoglycemic effect and mechanism of Dendrobium officinale polysaccharide (DOP) on type 2 diabetes mellitus (T2DM) mice established by high-fat diet and streptozotocin. The results showed that DOP improved glycolipid metabolism and serum inflammation levels， and inhibited intestinal-derived lipopolysaccharide (LPS) translocation， suggesting that inhibiting LPS-mediated intestinal barrier damage may be a key target for DOP to alleviate T2DM. Interestingly， the study found that DOP reduced intestinal inflammation and oxidative stress levels， significantly up-regulated the mRNA expression of tight junction proteins Claudin-1， Occludin and Zonula occluden-1 (ZO-1)， and ameliorated intestinal epithelial damage. In addition， DOP strongly inhibited the intestinal pathogenic bacteria and LPS-producing bacteria Helicobacter， Enterococcus and Desulfovibrio with a reduction rate of 95%， 73% and 9%， respectively， and promoted the proliferation of anti-inflammatory bacteria Bifidobacterium and Lactobacillus by 139% and 8%， respectively. Taken together， the hypoglycemic effect of DOP was related to the protection of intestinal mucosal barrier， and its underlying mechanism lied in its excellent anti-inflammatory and gut microbiota-modulatory effects， providing a theoretical basis for developing DOP as a novel prebiotic in functional food for diabetes.

H7N9 subtype avian influenza virus poses a great challenge for poultry *** disease virus(NDV)-vectored H7N9 avian influenza vaccines(NDV_(vec)H7N9)are effective in disease control because they are protective and allow mass *** note,these vaccines elicit undetectable H7N9-specific hemagglutination-inhibition(HI)but high IgG antibodies in ***,the molecular basis and protective mechanism underlying this particular antibody immunity remain ***,immunization with an NDV_(vec)H7N9 induced low anti-H7N9 HI and virus neutralization titers but high levels of hemagglutinin(HA)-binding IgG antibodies in *** residues(S150,G151 and S152)in HA of H7N9 virus were identified as the dominant epitopes recognized by the NDV_(vec)H7N9 immune *** transferred NDV_(vec)H7N9 immune serum conferred complete protection against H7N9 virus infection in *** NDV_(vec)H7N9 immune serum can mediate a potent lysis of HA-expressing and H7N9 virus-infected cells and significantly suppress H7N9 virus *** activities of the serum were significantly impaired after heat-inactivation or treatment with complement inhibitor,suggesting the engagement of the complement ***,mutations in the 150-SGS-152 sites in HA resulted in significant reductions in cell lysis and virus neutralization mediated by the NDV_(vec)H7N9 immune serum,indicating the requirement of antibody-antigen binding for complement ***,antibodies induced by the NDV_(vec)H7N9 can activate antibody-dependent complement-mediated lysis of H7N9 virus-infected cells and complement-mediated neutralization of H7N9 *** findings unveiled a novel role of the complement in protection conferred by the NDV_(vec)H7N9,highlighting a potential benefit of engaging the complement system in H7N9 vaccine design.

Lipidomics and whole-genome sequencing identify lipid profiles in fermented golden pompano(Trachinotus ovatus) inoculated with Bacillus subtilis: Influence of indigenous strains on lipid changes

Inoculated fermentation enables rapid fermentation of aquatic products. No studies have been conducted on the lipid profiles of inoculated fermented golden pompano (Trachinotus ovatus). In this study， a lipase-producing Bacillus subtilis with salt tolerance was screened from traditionally fermented golden pompano (TF) and used as a starter culture. Whole-genome sequencing analysis revealed it carries four CRISPR structures and two genes encoding triacylglycerol lipase. Untargeted lipidomics identified lipid molecules (833) in six major classes from B. subtilis SCSMX-2 fermented golden pompano (IF). Twenty-eight lipid molecules were upregulated in IF， including phosphatidylcholines (PCs)， triacylglycerols (TAGs)， and lysophosphatidylcholine. B. subtilis supplementation enhanced the production of polyenyl phosphatidylcholines and medium- and long-chain triacylglycerols. The IF rich in linoleic， docosahexaenoic acids， and eicosapentaenoic acid were primarily distributed in the sn-2 position of DHA， PC and PE. This research revealed the lipid profiles of IF， providing theoretical basis for the application of B. subtilis in the fermented fish industry.
