Background Culex pipiens pallens andCulex pipiens quinquefasciatus are the dominant species ofCulex mosquitoes in China and important disease vectors. Long-term use of insecticides can cause mutations in the voltage-gated sodium channel (vgsc) gene of mosquitoes, but little is known about the current status and evolutionary origins ofvgsc gene in different geographic populations. Therefore, this study aimed to determine the current status ofvgsc genes inCx. p. pallens andCx. p. quinquefasciatus in China and to investigate the evolutionary inheritance of neighboring downstream introns of thevgsc gene to determine the impact of insecticides on long-term *** Sampling was conducted from July to September 2021 in representative habitats of 22 provincial-level administrative divisions in China. Genomic DNA was extracted from 1308 mosquitoes, the IIS6 fragment of thevgsc gene on the nerve cell membrane was amplified using polymerase chain reaction, and the sequence was used to evaluate allele frequency and knockdown resistance (kdr) frequency. MEGA 11 was used to construct neighborjoining (NJ) tree. PopART was used to build a TCS *** There were 6 alleles and 6 genotypes at the L1014 locus, which included the wild-type alleles TTA/L and CTA/L and the mutant alleles TTT/F, TTC/F, TCT/S and TCA/S. The geographic populations with akdr frequency less than 20.00% were mainly concentrated in the regions north of 38° N, and the geographic populations with akdr frequency greater than 80.00% were concentrated in the regions south of 30° *** frequency increased with decreasing latitude. The infection of host plants by many different viruses causes reactive oxygen species(Ros)accumulation and yellowing symptoms,but the mechanisms through which plant viruses counteract RoS-mediated immunity to facilitate infection and symptom development have not been fully *** plant viruses are transmitted by insect vectors in the field,but the molecular mechanisms underlying virus-host-insect interactions are *** this study,we investigated the interactions among wheat,barley yellow dwarf virus(BYDV),and its aphid vector and found that the BYDV movement protein(MP)interacts with both wheat catalases(CATs)and the 26S proteasomeubiquitin receptor non-ATPase regulatorysubunit2homolog(PSMD2)to facilitate the 26S proteasome-mediateddegradation of CATs,promotingviral infection,disease symptom development,and aphid *** of the BYDV MP gene in wheat enhanced the degradation of CATs,which leading to increased accumulation of ROS and thereby enhanced viral ***,transgenic wheat lines overexpressing BYDV MP showed significantly reduced proliferation of wingless aphids and an increased number of winged *** with this observation,silencing of CAT genes also enhanced viral accumulation and reduced the proliferation of wingless aphids but increased the occurrence of winged *** contrast,transgenic wheat plants overexpressing TaCAT1 exhibited the opposite changes and showed increases in grain size and weight upon infection with *** assays demonstrated that BYDV MP interacts with PSMD2 and promotes 26S proteasome-mediated degradation of TaCAT1 likely in a ubiquitination-independent ***,our study reveals a molecular mechanism by which a plant virus manipulates the Ros production system of host plants to facilitate viral infection and transmission,shedding new light on the sophisticated interactions among viruses,host plants,and insect vectors.

Rodent pests not only cause severe agricultural loss but also spread zoonotic pathogens to human ***-agulant rodenticides are widely used to decrease the population densities of rodents but often lead to the spillover of ectoparasites becausefleas and ticks may gather on surviving ***,it is necessary to killfleas and ticks before culling rodents to minimize the risk of pathogen *** this study,we used a mixture of ivermectin(an antiparasitic drug)and bromadiolone(an anticoagulant rodenticide)to control both rodent andflea/tick *** found that in a laboratory test,0.01%ivermectin bait was not lethal for greater long-tailed hamsters after 7 days of treatment,while 0.1%ivermectin bait was lethal for approximately 33%of treated *** afield test,bait containing 0.001%,0.005%,0.01%,and 0.05%ivermectin decreased the number offleas per vole of Brandt’s voles to 0.42,0.22,0.12,and 0.2,respectively,compared with 0.77 in the control group,indicating that 0.01%ivermectin bait performed best in *** another laboratory test,bait containing a 0.01%ivermectin and 0.005%bromadiolone mixture caused the death of all voles within 6–14 days after the intake of the *** thefield test,the bait containing 0.01%ivermectin and 0.005%bromadiolone reduced the average number offleas per vole to 0.35,which was significantly lower than the 0.77 of the control *** results indicate that a 0.01%ivermectin and 0.005%bromadiolone mixture could be used to control both rodents andfleas to minimize the spillover risk of disease transmission when using traditional rodenticides.

The small brown planthopper(SBPH,Laodelphax striatellus)is a significant rice pest,responsible for transmitting rice stripe virus(RSV)in a persistent and propagative *** is one of the most detrimental rice viruses,causing rice stripe disease,which results in considerable loss of rice grain *** RNA interference and gene knockout techniques have enabled gene downregulation in SBPH,no system currently exists for the overexpression of endogenous or exogenous ***,the development of a protein expression system for SBPH is imperative to serve as a technical foundation for pest control and gene function *** study aimed to construct an expression vector using the promoter of the constitutive-expressed tubulin gene of SBPH,and promoter of human cytomegalovirus(CMV).Fluorescence experiments demonstrated that both tubulin and CMV promoter could drive green fluorescent protein(GFP)expression in SBPH,and could also facilitate the expression of a nucleocapsid protein(NP)-GFP fusion protein containing viral NP with comparable *** expression vector optimization,we have identified that the 3 tandem CMV promoters display a significantly higher promoter activity compared with both the 2 tandem CMV promoters and the single CMV *** addition,the incorporation of Star polycation nanoparticles significantly enhanced the expression efficiency in *** results provide a promising technical platform for investigating gene functions in SBPH.
