Objective This study optimizes three-dimensional(3D) culture conditions of HepG2 using response surface methodology(RSM) based on the VitroGel system to facilitate the cell model in vitro for liver tissues.Method HepG2 cell was 3D cultured on the VitroGel system.Cell viability was detected using Cell Counting Kit-8(CCK-8) assay of HepG2 lived cell numbers.The proliferation of HepG2 cell and clustering performance was measured via fluorescence staining test.Albumin concentration in the culture medium supernatant as an index of HepG2 cell biological function was measured with ELISA kit.Independent factor tests were conducted with three key factors:inoculated cell concentration,cultured time,and dilution degree of the hydrogel.The preliminary results of independent factor tests were used to determine the levels of factors for RSM.Result The selected optimal culture conditions are as follows:concentration of inoculated cells was4.44 × 10^(5)/mL,culture time was 4.86 days,and hydrogel dilution degree was 1:2.23.The result shows that under optimal conditions,the predicted optical density(OD) value of cell viability was 3.10 and measured 2.978 with a relative error of 3.94%.Conclusion This study serves as a reference for the 3D HepG2 culture and constructs liver tissues in vitro.Additionally,it provides the foundation for repeated dose high-throughput toxicity studies and other scientific research work.

AIM To investigate changes in dietary intake following a 6-mo randomised controlled trial of the self-directed, gendertailored type 2 diabetes mellitus(T2DM) Prevention Using LifeS tyle Education(PULSE) program in men. METHODS Men aged 18-65 years, with a body mass index(BMI)25-40 kg/m^2, and at high risk for developing T2DM were recruited from the Hunter Region of New South Wales,Australia. Eligible participants were randomised into one of two groups:(1) waitlist control; or(2) PULSE intervention. Dietary intake was assessed at baseline and immediately post-program using the Australian Eating Survey food frequency questionnaire and diet quality measured using the Australian Recommended Food Score(ARFS). RESULTS One hundred and one participants(n = 48, control; n= 53, intervention, mean age 52.3 ± 9.7 years, BMI of 32.6 ± 3.3 kg/m^2) commenced the study. Following the active phase, differences between groups were observed for proportion of total energy consumed from healthful(core) foods(+7.6%EI, P nutrition messages led to significant improvements in dietary intake in men at risk of T2DM.

AIM: To assess the anti-inflammatory effect of the probiotic Bifidobacterium lactis (B. lactis) in an adoptive transfer model of colitis. METHODS: Donor and recipient mice received either B. lactis or bacterial culture medium as control (deMan Rogosa Sharpe) in drinking water for one week prior to transfer of a mix of naive and regulatory T cells until sacrifice. RESULTS: All recipient mice developed signs of colonic inflammation, but a significant reduction of weight loss was observed in B. lactis-fed recipient mice compared to control mice. Moreover, a trend toward a diminution of mucosal thickness and attenuated epithelial damage was revealed. Colonic expression of pro-inflammatory and T cell markers was significantly reduced in B. lactis-fed recipient mice compared to controls. Concomitantly, forkhead box protein 3, a marker of regulatory T cells, was significantly up-regulated by B. lactis. CONCLUSION: Daily oral administration of B. lactis was able to reduce inflammatory and T cells mediators and to promote regulatory T cells specific markers in a mouse model of colitis.

Background: Floorball training offers a motivating and socially stimulating team activity for older adults, and 12 weeks of floorball training twice a week among men aged 65—76 years have been shown to have positive effects on a number of physiological parameters important for health. However, the effect of long-term participation in floorball training among male elderly has not been investigated. The aim of the present study was to examine the effect of 26-month self-organized regular participation in floorball training on cardiovascular fitness, body composition, blood lipids, glucose control, and physical function among recreationally active men aged 66—78 years.Methods: After completing a 12-week randomized and controlled intervention with floorball and petanque training in the autumn 2014 or spring2015, 15 subjects chose to participate in floorball training(floorball group, FG), whereas 16 subjects resumed their usual lifestyle(control group,CG). FG took part in self-organized floorball training 1.7 sessions of 40 min/week, and CG continued their normal recreationally active lifestyle during a 26-month follow-up period. At baseline and after the follow-up period subjects were tested for cardiovascular fitness, glucose control(resting blood samples), body composition dual-energy X-ray absorptiometry(DXA-scanning), and functional capacity.Results: In FG, the decline in maximal oxygen uptake(VO_(2max)) during the follow-up period was lower(242 ± 379 mL/min, p = 0.01), blood glycosylated hemoglobin(HbA1c) increased less(—1.6 ± 2.9 mmol/L, p = 0.02), and leg bone mineral density increased more(0.03 ± 0.05 g/cm^2,p = 0.02) than those in CG. The effects on body mass, total lean body mass, fat mass, blood lipids, and physical function were similar in FG and CG.Conclusion: Approximately twice weekly floorball sessions with 40 min/session over 26-month appear to reduce age-related decline in cardiovascular fitness and glucose control and improve leg bone mineral density, suggesting that long-term participation in floo

In vitro digestion studies demonstrate large potential to gain more and quicker insights into the underlying mechanisms of feed additives,allowing the optimization of feed design.Unfortunately,current in vitro digestion models relevant for broiler chickens lack sufficient description in terms of protocols and standardisation used.Furthermore,no distinction is made between the different life phases of these animals(starter,grower,and finisher).Hence,our research aimed to establish adapted in vitro digestion conditions,corresponding to the 3 life phases in broilers,with specific focus on lipid digestion.The effect of 3 different bile salt concentrations of 2,10,and 20 mM,and 3 different lipase activities of 5,20,and 100 U/mL,on in vitro lipid digestion kinetics were evaluated using a full factorial design.These values were selected to represent starter,grower,and finisher birds,respectively.Our findings showed that the extent of lipid digestion was mainly influenced by lipase activity.The rate of lipid digestion was affected by an interplay between bile salt concentration and lipase activity,due to possible lipase inhibition at certain bile salt concentrations.Overall,this work resulted in 3 in vitro lipid digestion models representative for starter,grower,and finisher birds.In conclusion,this research showed the impact of adapted in vitro digestion conditions on lipid digestion kinetics and thus the need for these conditions relevant for each life phase of broilers.

The principle determinant of melanin derived hair colour and patterning in mammals is genetic, but environmental factors are now thought to play a role. It has been shown that the concentration of melanins in cat hair is influenced by the amino acid composition of their diets. Also, puppies were found to require tyrosine(Tyr) intake significantly greater than that recommended for normal growth and development in order to optimize melanin expression in their coats. Much of the work to date has been conducted in growing animals. Less is known about the relationship between nutrition and hair melanin deposition in healthy adult animals. In this study, we fed 2 groups of adult black Labrador retrievers(12 dogs/group) different concentrations of Phe + Tyr(5.6 vs. 3.5 g/Mcal) for 24 weeks and used spectrophotometric measurements every 8 weeks to detect any associated changes in the dogs' hair colour.The higher intake dogs showed reduced dilution of their black coat pigment compared with the lower intake dogs. Specifically, following 16 weeks at the higher intake, the dogs showed less yellow pigmentation to their coats(P = 0.0032), and after 24 weeks at the higher intake, the dogs showed less red(P health.

Edible mushrooms as a potent therapeutics of subclinical thyroid dysfunction among adults,especially in obese individuals:a prospective cohort study

Objective: Growing evidence has shown that ultra-processed foods were significantly associated with increased risks of obesity and other noncommunicable diseases. Front-of-package(FOP) labels were recommended by many countries to promote healthy choice of pre-packaged foods, among which warning labelling, increasingly adopted in Latin Countries, was proved most effective in reducing the choice of unhealthy foods. The aim of this study is to evaluate the healthiness of pre-packaged foods in China using two warning labelling models, so as to provide policy reference for the development of FOP in China. Methods This study used nutrients and ingredients data of pre-packaged foods in China collected from supermarkets and retail stores during 2017 to 2020 using FoodSwitch China applications through designated collecting in particular areas and crowdsourcing across the country. The data were classified into 18 groups according to the food categorization system developed by the Food Monitoring Group at The George Institute Global health as part of The International Network for Food and Obesity/non-communicable diseases Research, Monitoring and Action Support(INFORMAS). A total of 14 eligible groups of foods data were included for analysis. The 4 excluded food groups were 1) Vitamins and supplements;2) Alcohol;3) Special foods;4) Foods unable to be categorized. Foods were classified into 4 processed levels according to NOVA classification system(1, unprocessed or minimally processed;2, processed culinary ingredients;3, processed and 4, ultra-processed). Two warning labelling models including the Pan American health Organization Nutrient Profile Model(PAHO NP model) and Chilean nutrient profile models(Chilean NP model) were employed to assess the nutritional quality of collected foods under the eligibility of processed and ultra-processed foods. Background:Mushrooms are a good source of many nutrients which are potentially beneficial for chronic diseases.We speculated that due to its abundant nutrients edible mushrooms might have a beneficial effect on the prevention of subclinical thyroid dysfunction(SCTD).Therefore,we designed a large-scale cohort study to examine whether mushrooms consumption is a protective factor for SCTD in adults.Methods:This prospective cohort study investigated 6631 participants(mean age:(45.0±10.2)years;55.1%men).Edible mushrooms consumption was measured at baseline using a validated food frequency questionnaire.SCTD was defined as abnormal serum thyroid-stimulating hormone levels and normal free thyroxine.Cox proportional hazards regression models were used to examine the association of edible mushrooms consumption with incident SCTD.Results:During follow-up period,a total of 262 new cases of SCTD were identified,the incidence rate of subclinical hypothyroidism was 8.9/1000 person-years and subclinical hyperthyroidism was 7.2/1000 person-years.After adjusting potential confounding factors,the multivariable hazard ratios(95%confidence intervals)for subclinical hypothyroidism were 1.00(reference)for almost never,0.53(0.29,0.97)for 1-3 times/week and 0.30(0.10,0.87)for≥4 times/week(P for trend=0.02).It also showed edible mushrooms consumption was inversely associated with subclinical hypothyroidism in obese individuals but not non-obese individuals,the final hazard ratios(95%confidence intervals)were 0.14(0.03,0.73)(P for trend<0.01).Conclusions:This population-based prospective cohort study has firstly demonstrated that higher edible mushrooms consumption was significantly associated with lower incidence of subclinical hypothyroidism among general adult population,especially in obese individuals.

Background:Physical inactivity and insomnia symptoms are independently associated with increased risk of depression and anxiety;however,few studies jointly examine these risk factors.This study aimed to prospectively examine the joint association of physical activity(PA)and insomnia symptoms with onset of poor mental health in adults.Methods:Participants from the 2013 to 2018 annnual waves of the Household Income and Labour Dynamics in Australia panel study who had good mental health(Mental health Inventory-5>54)in 2013,and who completed at least 1 follow-up survey(2014-2018),were included(n=10,977).Poor mental health(Mental health Inventory-5≤54)was assessed annually.Baseline(2013)PA was classified as high/moderate/low,and insomnia symptoms(i.e.,trouble sleeping)were classified as no insomnia symptoms/insomnia symptoms,with 6 mutually exclusive PAinsomnia symptom groups derived.Associations of PA-insomnia symptom groups with onset of poor mental health were examined using discrete-time proportional-hazards logit-hazard models.Results:There were 2322 new cases of poor mental health(21.2%).Relative to the high PA/no insomnia symptoms group,there were higher odds(odds ratio and 95%confidence interval(95%CI))of poor mental health among the high PA/insomnia symptoms(OR=1.87,95%CI:1.57-2.23),moderate PA/insomnia symptoms(OR=1.93,95%CI:1.61-2.31),low PA/insomnia symptoms(OR=2.33,95%CI:1.96-2.78),and low PA/no insomnia symptoms(OR=1.14,95%CI:1.01-1.29)groups.Any level of PA combined with insomnia symptoms was associated with increased odds of poor mental health,with the odds increasing as PA decreased.Conclusion:The se findings highlight the potential benefit of interventions targeting both PA and insomnia symptoms for promoting mental health.
