DNA mismatch repair(MMR)is one of the biological pathways that plays a critical role in DNA homeostasis,primarily for repairing base-pair mismatches and insertion/deletion loops that occur during DNA *** also takes part in other metabolic pathways and regulates cell cycle *** in MMR are associated with genomic instability,predisposition to certain types of cancers including hereditary nonpolyposiscolorectal cancer and some other sporadic cancers,resistance to certain therapeutic drugs,infertility and the health of offspring conceived by assisted reproductive technology in human.

Objective To determine whether patients with isolated teratozoospermia have increased or decreased incidence of chromosomal aneuploidies. Methods Sperm obtained from isolated teratozoospermic men （teratozoospermic group, n=18） and normal fertile men （the control, n=5） were analyzed using FISH （for chromosomes 18, X and Y）. Results A total of 58 178 spermatozoa were counted from the teratozoospermia group and 16 369 spermatozoa were counted from the control, with the hybridization rates of 97.5% and 98.3%, respectively. The major types of chromosomal aneuploidies were disomy （YY18, XX18, XY18, Y1818 and 3（1818） and diploidy （1818XX, 1818YY, 1818XY）. In the teratozoospermic group and the control, the disomy rates of 18 chromosome were 0.29 ±0.16% and 0.03 ±0.02%, the disomy rates of sex chromo- some were 0.65 ±0.24% and 0.05 ± 0.02%, the diploidy rates were 0.14 ± 0.12% and 0. 04±0.03%, respectively..411 the differences between these two groups were significant （P〈0. 05）.Conclusion Sperm of isolated teratozoospermic men have higher rates of 18, X and Y chromosomal aneuploidies than that of the fertile controls.

Factors Relating with Fertilization Failure in Conventional IVF-ET Cycles

Objective To analyze the factors relating with fertilization failure in conventional IVF cycles. Methods The total fertilization failure rate of 2 429 IVF cycles from January 2011 to May 2012 in the reproductive centre of the Women and Children Hospital of Jiangxi Province were retrospectively analyzed. Risk factors were identified by univariate and multivariate logistic regression analyses. Results The total fertilization failure rate of conventional IVF-ET rate was 5. 7% （139/ 2 429）. Thepercentage of morphologically normal sperm （11.1 ±5.8% vs 13.4 ±5.3%）, progressive motility （47.4± 10.5% vs 50.1 ±8.6%）, percentage of couple primary infertility （79.1% vs 44.3%）, percentage o f female primary infertility （69.1% vs 36.3%）, percentage of male primary infertility （74.8% vs 41.7%）, percentage of without oviduct obstruction patients （30.2% vs 13.6%） and percentage of couples primary infertility （79. 1% vs 44. 3%） of the fertilization failure patients were significantly different from those of the fertilized patients （P〈O.05）. Besides, higher total fertilization failure rate was found in couples with isolated teratozoospermia than in couples with normal percentage of morphologically normal sperm （15.0% vs 5.2%）. After that, it was found by multivariate logistic regression analyses that many factors including percentage of morphologically normal sperm, sperm concentration, couples primary infertility and female infertility years were related with fertilization failure. Conclusion Patients with low percentage of morphologically normal sperm （〈4%）, low sperm concentration and motility, male primary infertility, female primary infertility, without oviduct obstruction, long female infertility years or/and couples primary infertility are at high risk of fertilization failure. More attention should be paid to these patients for avoiding the fertilization failure.

Objective: To observe the effects of electroacupuncture(EA) on reproductive outcomes in women with Shen(Kidndy) deficiency syndrome after in vitro fertilization-embryo transfer(IVF-ET),and explore the underlying molecular ***: Sixty-six infertile patients with Shen de?ciency syndrome undergoing IVF-ET were divided into EA or control groups according to a random table,33 cases in each *** undergoing IVF,patients in the EA and control groups received EA therapy and placebo needle puncture,respectively,for 3 menstrual *** de?ciency syndrome scores were *** outcome measures included the number of retrieved oocytes and fertilization,high-quality embryo and clinical pregnancy *** ?uid was collected on the day of oocyte retrieval,and granulosa cell expression of phosphatidylinositide3-kinases(PI3 K),serine-threonine kinase(Akt) and forkhead box O3(Foxo3 a) m RNA were measured by reverse transcribed and quantitative real-time polymerase chain ***: Syndrome scores for pre-versus post-treatments decreased significantly(16.53±1.75 to 8.67±1.61) in the EA group(P<0.05),but showed no signi?cant change in the control group(17.18±1.58 to 14.74±1.58).A signi?cant difference in score change was found between the EA and control groups(P<0.05).High-quality embryo and clinical pregnancy rates were both increased in the EA group compared with the control group [69.15%(195/282) vs.60.27%(176/292) and 66.67%(22/33) vs.42.42%(14/33),respectively,P<0.05].The fertilization rate was equivalent in EA and control *** difference was found in the number of retrieved oocytes between the two *** cell expression levels of PI3 K and Akt m RNA were signi?cantly increased in the EA group compared with the control group,while the expression of Foxo3 a was reduced(all P<0.05).Conclusions: For infertile patients with Shen de?ciency syndrome undergoing IVF,EA for tonifying Shen as an adjunct treatment may alleviate clini

Background Polycystic ovary syndrome （PCOS） is considered to be the most common endocrine disorder in women of reproductive *** involvement of an abnormal menstrual cycle in the etiology of PCOS remains *** aimed to analyze the characteristics of abnormal menstrual cycles and their association with PCOS in community and hospital patient *** Women with PCOS identified from 2111 permanent female residents in the community of Beijing and 506 outpatients obtained from the reproductive clinic of Peking University Third Hospital were recruited for this study,comprising the PCOS community group and the PCOS hospital group, *** group was further divided into four subgroups according to the length of menstrual cycles：〈21 days; 21-34 days; 35-60 days; and 〉60 *** in each group were interviewed using a questionnaire to assess factors including age, age of menarche, menstrual cycle history, related family history, and modified Ferriman-Gallwey （mF-G） *** women received transvaginal ultrasound scan and had fasting blood samples taken for endocrine evaluation.A two-tailed P value of 〈0.05 was considered *** In the community population, the prevalence of abnormal menstrual cycle was 27.19% （574/2111）.The prevalence of PCOS in the community was 6.11% （129/2111） according to Rotterdam *** the community group,the most common menstrual cycle length was 35-60 days, whereas for the hospital group, it was 〉60 *** both the community and hospital groups, the most common phenotype of PCOS was that of Oligo/amenorrhea＋PCO＋hyperandrogenism （HA）（O＋P＋H）（P=0.000）.With increasing cycle length of 35-60 days to 〉60 days, the percentages of Oligo/amenorrhea＋PCO （O＋P） and O＋P＋H were found to significantly decrease in the community group and significantly increase in the hospital group （P=0.000 for each）.In the hospital group, as the menstrual cycle length increased from 35-60 days to 〉60 days, the rate of spontaneous

Revised Super-long Down-regulation Protocol Improves the Outcome of Infertile Patients with Repeated Implantation Failure in IVF/ICSI-ET

Objective To investigate the outcome of revised super-long down-regulation protocol （RSDP） for in vitro fertilization / intracytoplasmic sperm injection-embryo transfer （1VF/1CS1-ET） in the special infertile patients with repeated implantation failure （RIF）. Methods Patients with RIF were divided into RSDP group and routine long downregulation protocol （RLDP） group. In RSDP group, gonadotropin releasing hormone agonist （GnRHa） was injected intramuscularly by 2.5 mg in mid-luteal phase for the ftrst time and 1.25 mg after 28 d; gonadotropin （Gn） was started 14 d later after the second GnRHa dose. IVF/ICSI-ET was performed according to the routine procedure. The clinical outcomes of RSDP group were compared with those of RLDP group. Results In RSDP group, the number of retrieved oocytes and valid embryos was significantly lower （P〈0.05）; there were no significant differences about fertilization rate （P〉0. 05）; both good-quality embryo rate and implantation rate were significantly increased （P〈0. 005）; clinical pregnancy rate was obviously improved （P〈0. 05）, as compared with RLDP *** RSDP can improve the IVF outcomes significantly in RIF patients.
