Transient receptor potential vanilloid 1 involved in the analgesic effects of total flavonoids extracted from Longxuejie(Resina Dracaenae Cochinchinensis)

OBJECTIVE: To evaluate the analgesic effects of total flavonoids of Longxuejie(Resina Dracaenae Cochinchinensis)(TFDB) and explore the possible analgesic mechanism associated with transient receptor potential vanilloid 1(TRPV1).METHODS: Whole-cell patch clamp technique was used to observe the effects of TFDB on capsaicin-induced TRPV1 currents. Rat experiments in vivo were used to observe the analgesic effects of TFDB. Western blot and immunofluorescence experiments were used to test the change of TRPV1 expression in DRG neurons induced by TFDB.RESULTS: Results showed that TFDB inhibited capsaicin-induced TRPV1 receptor currents in acutely isolated dorsal root ganglion(DRG) neurons of rats and the half inhibitory concentration was(16.7 ± 1.6) mg/L.TFDB(2-20 mg/kg) showed analgesic activity in the phase Ⅱ of formalin test and(0.02-2 mg per paw)reduced capsaicin-induced licking times of rats. TFDB(20 mg/kg) was fully efficacious on complete Freund's adjuvant(CFA)-induced inflammatory thermal hyperalgesia and capsaicin could weaken the analgesic effects. The level of TRPV1 expressions of DRG neurons was also decreased in TFDB-treated CFA-inflammatory pain rats.CONCLUSION: All these results indicated that the analgesic effect of TFDB may contribute to their modulations on both function and expression of TRPV1 channels in DRG neurons.

Carbon Emission Effects Driven by Evolution of Chinese Dietary Structure from 1987 to 2020

Exploring carbon emission effects based on the evolution of residents’ dietary structure to achieve the carbon neutrality goal and mitigate climate change is an important task.This study took China as the research object(data excluding Hong Kong,Macao and Taiwan) and used the carbon emission coefficient method to quantitatively measure the food carbon emissions from 1987–2020,then analyzed the carbon emission effects under the evolution of dietary structure.The results showed that during the study period,the Chinese dietary structure gradually changed to a high-carbon consumption pattern.The dietary structure of urban residents developed to a balanced one,while that of rural residents developed to a high-quality one.During the study period,the per capita food carbon emissions and total food consumption of Chinese showed an increasing trend.The per capita food carbon emissions of residents in urban and rural showed an overall upward trend.The total food carbon emissions in urban increased significantly,while that in rural increased first and then decreased.The influence of beef and mutton on carbon emissions is the highest in dietary structure.Compared with the balanced dietary pattern,the food carbon emissions of Chinese residents had not yet reached the peak,but were evolving to a high-carbon consumption pattern.

Construction of algal-bacterial consortia using green microalgae Chlorella vulgaris and As(Ⅲ)-oxidizing bacteria:As tolerance and metabolomic profiling

Bioremediation became a promising technology to resolve arsenic(As)contamination in aquatic environment.Since monoculture such as microalgae or bacteria was sensitive to environmental disturbance and vulnerable to contamination,green microalgae Chlorella vulgaris and arsenite(As(Ⅲ))-oxidizing bacteria Pseudomonas sp.SMS11 were co-cultured to construct algal-bacterial consortia in the current study.The effects of algae-bacteria(A:B)ratio and exposure As(Ⅲ)concentration on algal growth,As speciation and metabolomic profile were investigated.Algal growth arrested when treated with 100 mg/L As(Ⅲ)without the co-cultured bacteria.By contrast,co-cultured with strain SMS11 significantly enhanced As tolerance in C.vulgaris especially with A:B ratio of 1:10.All the As(Ⅲ)in culture media of the consortia were oxidized into As(Ⅴ)on day 7.Methylation of As was observed on day 14.Over 1% and 0.5% of total As were converted into dimethylarsinic acid(DMA)after 21days cultivation when the initial concentrations of As(Ⅲ)were 1 and 10 mg/L,respectively.Metabolomic analysis was further performed to reveal the response of consortia metabolites to external As(Ⅲ).The enriched metabolomic pathways were associated with carbohydrate,amino acid and energy metabolisms.Tricarboxylic acid cycle and glyoxylate and dicarboxylate metabolism were upregulated under As stress due to their biological functions on alleviating oxidative stress and protecting cells.Both carbohydrate and amino acid metabolisms provided precursors and potential substrates for energy production and cell protection under abiotic stress.Alterations of the pathways relevant to carbohydrate or amino acid metabolism were triggered by energy requirement.

3D hand pose and shape estimation from monocular RGB via efficient 2D cues

Estimating 3D hand shape from a single-view RGB image is important for many applications.However,the diversity of hand shapes and postures,depth ambiguity,and occlusion may result in pose errors and noisy hand meshes.Making full use of 2D cues such as 2D pose can effectively improve the quality of 3D human hand shape estimation.In this paper,we use 2D joint heatmaps to obtain spatial details for robust pose estimation.We also introduce a depth-independent 2D mesh to avoid depth ambiguity in mesh regression for efficient hand-image alignment.Our method has four cascaded stages:2D cue extraction,pose feature encoding,initial reconstruction,and reconstruction refinement.Specifically,we first encode the image to determine semantic features during 2D cue extraction;this is also used to predict hand joints and for segmentation.Then,during the pose feature encoding stage,we use a hand joints encoder to learn spatial information from the joint heatmaps.Next,a coarse 3D hand mesh and 2D mesh are obtained in the initial reconstruction step;a mesh squeeze-and-excitation block is used to fuse different hand features to enhance perception of 3D hand structures.Finally,a global mesh refinement stage learns non-local relations between vertices of the hand mesh from the predicted 2D mesh,to predict an offset hand mesh to fine-tune the reconstruction results.Quantitative and qualitative results on the FreiHAND benchmark dataset demonstrate that our approach achieves state-of-the-art performance.

Topology Optimization of Metamaterial Microstructures for Negative Poisson’s Ratio under Large Deformation Using a Gradient-Free Method

Negative Poisson’s ratio(NPR)metamaterials are attractive for their unique mechanical behaviors and potential applications in deformation control and energy absorption.However,when subjected to significant stretching,NPR metamaterials designed under small strain assumption may experience a rapid degradation in NPR performance.To address this issue,this study aims to design metamaterials maintaining a targeted NPR under large deformation by taking advantage of the geometry nonlinearity mechanism.A representative periodic unit cell is modeled considering geometry nonlinearity,and its topology is designed using a gradient-free method.The unit cell microstructural topologies are described with the material-field series-expansion(MFSE)method.The MFSE method assumes spatial correlation of the material distribution,which greatly reduces the number of required design variables.To conveniently design metamaterials with desired NPR under large deformation,we propose a two-stage gradient-free metamaterial topology optimization method,which fully takes advantage of the dimension reduction benefits of the MFSE method and the Kriging surrogate model technique.Initially,we use homogenization to find a preliminary NPR design under a small deformation assumption.In the second stage,we begin with this preliminary design and minimize deviations in NPR from a targeted value under large deformation.Using this strategy and solution technique,we successfully obtain a group of NPR metamaterials that can sustain different desired NPRs in the range of[−0.8,−0.1]under uniaxial stretching up to 20% strain.Furthermore,typical microstructure designs are fabricated and tested through experiments.The experimental results show good consistency with our numerical results,demonstrating the effectiveness of the present gradientfree NPR metamaterial design strategy.

Due to low immobilized ligand density,limited binding capacity,and severe interference from serum proteins,developing ideal peptide-based biomaterials for precise recognition and in vivo analysis of biopharmaceuticals remains a huge challenge.In this study,mimotope peptide modified pompon mum-like biomimetic magnetic microparticles(MMPs,3.8μm)that mimic the specific functionalities of CD20 on malignant B cells were developed for the first time.Benefit from the numerous ligand binding sites(Ni^(2+))on the pompon mum-like MMPs,these novel materials achieved≥10 times higher peptide ligand densities(>2300 mg/g)and antibody binding capacities(1380 mg/g)compared to previous reported biomaterials.Leveraging the high specificity of the mimotope peptide,rituximab can be precisely recognized and enriched from cell culture media or serum samples.We also established an LC-MS/MS method using the MMPs for tracking rituximab biotransformation in patient serum.Intriguingly,deamidation of Asn55 and Asn33,as well as oxidation of Met81 and Met34 were observed at the key complementarity determining regions of rituximab,which could potentially influence antibody function and require careful monitoring.Overall,these versatile biomimetic MMPs demonstrate superior recognition and enrichment capabilities for target antibodies,offering interesting possibilities for biotransformation analysis of biopharmaceuticals in patient serum.

Enhanced Natural Convection for Accelerating Melting of Phase Change Material in Cellular Structure through Inserting Fin

The design of thermal conductivity enhancers(TCE) is quite critical to overcoming the disadvantage of the poor thermal conductivity of phase change materials(PCM).The main contribution of this study is firstly to discuss how to actively enhance natural convection of the melted PCM in cellular structure by the fin formed in the structure under the condition of the same metal mass,apart from simultaneously improving heat conduction,which can boost the heat transfer performance.Also,a tailored hybrid fin-lattice structure(HFS) as TCE is designed and fabricated by additive manufacturing(AM).A two-equation numerical method is applied to study the heat transfer of the PCM,and its feasibility is validated with the experimental data.The numerical results indicate that enhanced natural convection and improved heat conduction can be obtained simultaneously when a well-designed fin is embedded into a lattice structure.The enhanced natural convection results in the improved melting rate and the decreased wall temperature;e.g.,the complete melting time and the wall temperature are reduced by 11.6% and 19.7%,respectively,because of the fin for metal aluminum.Moreover,the parameters of HFS including the porosity,pore density,and fin dimension have a great impact on the heat transfer.The enhancement effect of the fin for HFS on the melting rate of the PCM increases as the thermal conductivity of the base material decreases.For example,when the fin is introduced into the lattice structure,the complete melting time is reduced by 24.1% for metal titanium.In summary,this study enables us to obtain a good understanding of the mechanism of the heat transfer and provides necessary experimental data for the structural design of HFS fabricated by AM.

Topology Optimization Method for Microscale Structures Described with Integral Nonlocal Theory

The integration of additive manufacturing and topology optimization makes it possible to fabricate complex configurations,especially for microscale structures,which can guarantee the realization of high-performance structural designs.However,topology results often contain microstructures(several multicellular scales)similar to the characteristic length of local macrostructures,leading to errors in structural performance analysis based on classical theories.Therefore,it is necessary to consider the size effect in topology optimization.In this paper,we establish a novel topology optimization model utilizing the integral nonlocal theory to account for the size effect.The approach consists of an integral constitutive model that incorporates a kernel function,enabling the description of stress at a specific point in relation to strain in a distant field.Topology optimization structures based on nonlocal theory are presented for some benchmark examples,and the results are compared with those based on classical medium theory.The material layout exhibits significant differences between the two approaches,highlighting the necessity of topology optimization based on nonlocal theory and the effectiveness of the proposed method.

Improving error-correcting capability in DNA digital storage via soft-decision decoding

Error-correcting codes(ECCs) employed in the state-of-the-art DNA digital storage(DDS) systems suffer from a trade-off between error-correcting capability and the proportion of redundancy. To address this issue, in this study, we introduce soft-decision decoding approach into DDS by proposing a DNA-specific error prediction model and a series of novel strategies. We demonstrate the effectiveness of our approach through a proof-of-concept DDS system based on Reed-Solomon(RS) code, named as Derrick. Derrick shows significant improvement in error-correcting capability without involving additional redundancy in both in vitro and in silico experiments, using various sequencing technologies such as Illumina, Pac Bio and Oxford Nanopore Technology(ONT). Notably, in vitro experiments using ONT sequencing at a depth of 7× reveal that Derrick, compared with the traditional hard-decision decoding strategy, doubles the error-correcting capability of RS code, decreases the proportion of matrices with decoding-failure by 229-fold, and amplifies the potential maximum storage volume by impressive 32 388-fold. Also, Derrick surpasses ‘state-of-the-art' DDS systems by comprehensively considering the information density and the minimum sequencing depth required for complete information recovery. Crucially, the soft-decision decoding strategy and key steps of Derrick are generalizable to other ECCs' decoding algorithms.

Orthogonal Combinatorial Raman Codes Enable Rapid High-Throughput-Out Library Screening of Cell-Targeting Ligands

High-throughput assays play an important role in the fields of drug discovery,genetic analysis,and clinical diagnostics.Although super-capacity coding strategies may facilitate labeling and detecting large numbers of targets in a single assay,practically,the constructed large-capacity codes have to be decoded with complicated procedures or are lack of survivability under the required reaction conditions.This challenge results in either inaccurate or insufficient decoding outputs.Here,we identified chemical-resistant Raman compounds to build a combinatorial coding system for the high-throughput screening of cell-targeting ligands from a focused 8-mer cyclic peptide library.The accurate in situ decoding results proved the signal,synthetic,and functional orthogonality for this Raman coding strategy.The orthogonal Raman codes allowed for a rapid identification of 63 positive hits at one time,evidencing a high-throughput-out capability in the screening process.We anticipate this orthogonal Raman coding strategy being generalized to enable efficient high-throughput-out screening of more useful ligands for cell targeting and drug discovery.
