p38 MAPK is Crucial for Wnt1- and LiCl-Induced Epithelial Mesenchymal Transition

Idiopathic pulmonary fibrosis （IPF） is characterized by myofibroblast loci in lung parenchyma. Myofibroblasts are thought to originate from epithelial-to-mesenchymal transition （EMT）. Wntl and lithium chloride （LiCl） induce EMT in alveolar epithelial cells （AECs）, but the mechanisms are unclear. AECs were treated with Wntl and LiC1, respectively; morphological change and molecular changes of EMT, including E-cadherin, fibronectin, and vimentin, were observed. SB203580 was administrated to test the role of p38 MAPK signaling in EMT. Then AECs were treated with siRNAs targeting p38 MAPK to further test the effects of p38 MAPK, and the role was further confirmed by re-expression of p38 MAPK. At last β-catenin siRNA was used to test the role of β-catenin in the EMT process and relationship of β-catenin and p38 MAPK was concluded. Exposure of AECs to Wntl and LiC1 resulted in upregulation of vimentin and fibronectin with subsequent downregulation of E-cadherin. Wntl and LiC1 stimulated the p38 MAPK signaling pathways. Perturbing the p38 MAPK pathway either by SB203580 or through p38 MAPK siRNA blocked EMT and inhibited fibronetin synthesis, which were reversed by transfection of p38 MAPK expression plasmid. β-catenin siRNA attenuated the EMT process and decreased p38 MAPK phosphorylation, indicating that β-catenin is involved in the EMT- related changes through regulation of p38 MAPK phosphorylation. These findings suggest that p38 MAPK participates in the pathogenesis of EMT through Wnt pathway and that p38 MAPK may be a novel target for IPF therapy.

Implementation Multi Representation and Oral Communication Skills in department of Physics Education on Elementary Physics II

The ability of multi representation has been widely studied, but there has been no implementation through a model of learning. This study aimed to determine the ability of the students multi representation, relationships of multi representation capabilities and oral communication skills, as well as the application of the relations between the two capabilities through learning model Presentatif Based on Multi representation (PBM) in solving optical geometric (Elementary Physics II). A concurrent mixed method was used with qualitative-quantitative methods. Means of collecting data in the form of the pre-test and post-test are essay form, observation sheets of oral communication skills, and assessment of learning by observation sheet PBM learning models, they all have a high degree of validity category, which is 3.91, 4.22, 4.13, and 3.88, respectively. Test reliability with Alpha Cronbach technique, the reliability coefficient is 0.494. The students are in department of Physics Education Unnes as a research subject. Sequence multi representation tendency of students from high to low in sequence, representation of M, D, G, V; whereas the order of accuracy, the group representation V, D, G, M. Relationship of multi representation ability and oral communication skills are comparable/proportional. Implementation of conjunction can generate grounded theory. This study should be applied to the physics of matter, or any other university for comparison.

Acute hypobaric hypoxic brain damage is a potentially fatal high-altitude *** plays a critical role in ischemic brain injury,but its role in hypo-baric hypoxia(HH)remains *** we used an HH chamber to demonstrate that acute HH exposure impairs autophagic activity in both the early and late stages of the mouse brain,and is partially responsible for HH-induced oxidative stress,neuronal loss,and brain *** autophagic agonist rapamycin only promotes the initiation of *** proteome analysis,a screen showed that protein dynamin2(DNM2)potentially regulates autophagic *** of DNM2 significantly increased the formation of autolysosomes,thus maintaining autophagic flux in combination with ***,the enhancement of autophagic activity attenuated oxidative stress and neurological deficits after HH *** results contribute to evidence supporting the conclusion that DNM2-mediated autophagic flux represents a new therapeutic target in HH-induced brain damage.

Electrochemical hydrogen storage behaviors of as-cast and spun RE-Mg-Ni-Co-Al-based AB2-type alloys applied to Ni-MH battery

Preparation of La-Mg-Ni-Co-Al-based AB2-type alloys La0.8-xCe0.2YxMgNi3.4Co0.4Al0.1(x=0,0.05,0.10,0.15,0.20)was performed using melt spinning *** influences of spun rate and Y content on structures and electrochemical hydrogen storage characteristics were *** base phase LaMgNi4 and the lesser phase LaNis were detected by X-ray diffraction(XRD)and scanning electron microscope(SEM).The variations of spinning rate and Y content cause an obvious change in phase content,but without altering phase composition,namely,with spinning rate and Y content growing,LaMgNi4 phase content augments while LaNi5 content ***,melt spinning and the replacing La by Y refine the grains *** electrochemical tests show a favorable activation capability of the two kinds of alloys,and the maximum discharge capacities are achieved during the first *** capacity firstly increases and subsequently decreases with spinning rate rising,while cycle stability is ameliorated and discharge capacity decreases with Y addition *** is found that the amelioration of cycle stability is due to the enhancement of anti-pulverization,anti-corrosion and antioxidation abilities by both replacement of La with Y and melt ***,with the increase of Y addition and/or spinning rate,the electrochemical kinetics that contain charge transfer rate,limiting current density(IL),hydrogen diffusion coefficient(D)and the high rate discharge ability(HRD)firstly augment and then reduce.

A Data-Driven Clustering Recommendation Method for Single-Cell RNA-Sequencing Data

Recently,the emergence of single-cell RNA-sequencing(scRNA-seq)technology makes it possible to solve biological problems at the single-cell *** of the critical steps in cellular heterogeneity analysis is the cell type *** scRNA-seq clustering methods have been proposed to partition cells into *** all the methods,hierarchical clustering and spectral clustering are the most popular approaches in the downstream clustering analysis with different preprocessing strategies such as similarity learning,dropout imputation,and dimensionality *** this study,we carry out a comprehensive analysis by combining different strategies with these two categories of clustering methods on scRNA-seq datasets under different biological *** analysis results show that the methods with spectral clustering tend to perform better on datasets with continuous shapes in two-dimension,while those with hierarchical clustering achieve better results on datasets with obvious boundaries between clusters in *** by this finding,a new strategy,called QRS,is developed to quantitatively evaluate the latent representative shape of a dataset to distinguish whether it has clear boundaries or ***,a data-driven clustering recommendation method,called DDCR,is proposed to recommend hierarchical clustering or spectral clustering for scRNA-seq *** perform DDCR on two typical single cell clustering methods,SC3 and RAFSIL,and the results show that DDCR recommends a more suitable downstream clustering method for different scRNA-seq datasets and obtains more robust and accurate results.

BACKGROUND Pulmonary thromboembolism(PTE)is a serious postoperative complication that can occur after a ***,PTE is caused by the falling off of lower extremity deep vein thrombosis(LEDVT)after lower limb fracture *** and PTE after upper extremity fracture surgery are very *** is one of the most common clinical causes of sudden *** thromboembolism includes PTE and *** experienced one case of LEDVT and PTE after distal ulna and radius fracture *** purpose of our report is to raise awareness for orthopedic surgeons that PTE can occur after distal ulna and radius fracture surgery,and patients with high risk factors should be considered for prevention and treatment of thrombosis in a timely *** SUMMARY We report a 51-year-old Chinese male who had severe fractures of the left distal ulna,radius and little finger after a motorcycle *** patient underwent external fixation,open reduction and internal *** the third post-operative day,computed tomographic pulmonary angiography showed *** ultrasonography showed thrombus formation in the bilateral posterior tibial *** a period of anticoagulation therapy,on the 25th d after the PTE,computed tomographic pulmonary angiography showed that thrombus in both sides of the pulmonary artery ***,about 4 mo after the PTE,thrombosis in the deep veins of the lower limbs *** 1 year after the surgery,X-rays showed good fracture healing,and the function of the wrist joint recovered *** Though rare,PTE can occur after distal ulna and radius fracture surgery and patients with high risk factors should be considered for prevention and treatment of thrombosis in a timely manner.

Occurrence and ecotoxicological risk assessment of pesticides in sediments of the Rosetta branch,Nile River, Egypt

This study aimed to(1) monitor the occurrence and spatiotemporal variations of 100 pesticides in sediments collected monthly from July 2018 to June 2019 from sampling sites in El-Rahawy, Sabal, and Tala, along the Rosetta branch of the Nile River, Egypt, and(2) perform an ecological risk assessment for aquatic organisms upon exposure to the detected sediment pesticides based on the risk quotient(RQ) method. Out of the 100 pesticides monitored, 16 pesticides belonging to seven chemical families were detected, and 55% of the sediment samples were contaminated with one or more pesticide residues. The mean concentration(mg/kg dry weight(dw)) and detection frequency(%) of the four most frequently detected pesticides in the sediment samples were as follows: chlorpyrifos(0.18 mg/kg dw and 34%), p,p-DDE(0.018 mg/kg dw and 30%), cypermethrin(0.03 mg/kg dw and 14%), and deltamethrin(0.026 mg/kg dw and 13%). The spatial distribution exhibited that El-Rahawy had the highest pesticide load(2.86 mg/kg dw) among the studied sites, whereas the temporal variations revealed that the highest total pesticide concentrations were detected in winter season(1.73 mg/kg dw). Meanwhile, 12 pesticides showed high RQs(>1), posing a potential ecological risk to aquatic species that live and feed on such sediments.

Masquelet technique using an allogeneic cortical bone graft for a large bone defect:A case report

BACKGROUND The induced-membrane technique was initially described by Masquelet as an effective treatment for large bone defects,especially those caused by ***,we report a case of chronic osteomyelitis of the radius associated with a 9 cm bone defect,which was filled with a large allogeneic cortical bone graft from a bone *** bony union was achieved after 14 months of *** studies have used autogenous bone as the primary bone source for the Masquelet technique;in our case,the exclusive use of allografts is as successful as the use of autologous bone *** the advent of bone banks,it is possible to obtain an unlimited amount of allograft,and the Masquelet technique may be further improved based on this new way of bone *** SUMMARY In this study,we reported a case of repair of a long bone defect in a 40-year-old male patient,which was characterized by the utilization of allograft cortical bone combined with the Masquelet technique for the treatment of the patient's long bone defect in the *** patient's results of functional recovery of the forearm were surprising,which further deepens the scope of application of Masquelet technique and helps to strengthen the efficacy of Masquelet technique in the treatment of long bones *** Allograft cortical bone combined with the Masquelet technique provides a new method of treatment to large bone defect.
