As the elderly population expands,the pursuit of therapeutics to reduce morbidity and extend lifespan has become increasingly *** an FDA-approved drug for chronic cholestatic liver diseases,tauroursodeoxycholic acid(TUDCA),a natural bile acid,offers additional health benefits beyond liver ***,we show that TUDCA extends the lifespan and healthspan of ***,oral supplementation of TUDCA improves fitness in old mice,including clinically relevant phenotypes,exercise capacity and cognitive ***,TUDCA treatment drives broad transcriptional changes correlated with anti-aging ***,we discover that TUDCA targets the chaperone HSP90 to promote its protein refolding *** collaboration further alleviates aging-induced endoplasmic reticulum(ER)stress and facilitates protein homeostasis,thus offering resistance to *** summary,our findings uncover new molecular links between an endogenous metabolite and protein homeostasis,and propose a novel anti-aging strategy that could improve both lifespan and healthspan.

OBJECTIVE: To propose an automatic acupuncture robot system for performing acupuncture operations. METHODS: The acupuncture robot system consists of three components: automatic acupoint localization, acupuncture manipulations, and De Qi sensation detection. The OptiTrack motion capture system is used to locate acupoints, which are then translated into coordinates in the robot control system. A flexible collaborative robot with an intelligent gripper is then used to perform acupuncture manipulations with high precision. In addition, a De Qi sensation detection system is proposed to evaluate the effect of acupuncture. To verify the stability of the designed acupuncture robot, acupoints' coordinates localized by the acupuncture robot are compared with the Gold Standard labeled by a professional acupuncturist using significant level tests. RESULTS: Through repeated experiments for eight acupoints, the acupuncture robot achieved a positioning error within 3.3 mm, which is within the allowable range of needle extraction and acupoint insertion. During needle insertion, the robot arm followed the prescribed trajectory with a mean deviation distance of 0.02 mm and a deviation angle of less than 0.15°. The results of the lifting thrusting operation in the Xingzhen process show that the mean acupuncture depth error of the designed acupuncture robot is approximately 2 mm, which is within the recommended depth range for the Xingzhen operation. In addition, the average detection accuracy of the De Qi keywords is 94.52%, which meets the requirements of acupuncture effect testing for different dialects. CONCLUSION: The proposed acupuncture robot system streamlines the acupuncture process, increases efficiency, and reduces practitioner fatigue, while also allowing for the quantification of acupuncture manipulations and evaluation of therapeutic effects. The considerable hazard posed by periprosthetic joint infections underlines the urgent need for the rapid advancement of in-situ drug delivery systems within joint materials. However, the pursuit of sustained antibacterial efficacy remains a formidable challenge. In this context, we proposed a novel strategy that leverages swelling and erosion mechanisms to facilitate drug release of drug-loaded ultrahigh molecular weight polyethylene(UHMWPE), thereby ensuring its long-lasting antibacterial *** oxide(PEO), a hydrophilic polymer with fast hydrating ability and high swelling capacity, was incorporated in UHMWPE alongside the antibacterial tea polyphenol(epigallocatechin gallate,EGCG as representative). The swelling of PEO enhanced water infiltration into the matrix, while the erosion of PEO balanced the release of the encapsulated EGCG, resulting in a steady release. The behavior was supported by the EGCG release profiles and the corresponding fitted release kinetic *** demonstrated by segmented antibacterial assessments, the antibacterial efficiency was enhanced 2to 3 times in the PEO/EGCG/UHMWPE composite compared to that of EGCG/UHMWPE. Additionally, the PEO/EGCG/UHMWPE composite exhibited favorable biocompatibility and mechanical performance, making it a potential candidate for the development of drug-releasing joint implants to combat prosthetic bacterial infections.

Trimethylation of histone H3K4(H3K4me3)is widely distributed at numerous actively transcribed protein-coding genes throughout the ***,the interplay between H3K4me3 and other chromatin modifications in plants remains poorly *** this study,we show that the Arabidopsis thaliana ALFIN-LIKE(AL)proteins contain a C-terminal PHD finger capable of binding to H3K4me3 and a PHD-associated AL(PAL)domain that interacts with components of the Polycomb repressive complex 1,thereby facilitating H2A ubiquitination(H2Aub)at H3K4me3-enriched genes throughout the ***,we demonstrate that loss of function of SDG2,encoding a key histone H3K4 methyltransferase,leads to a reduction in H3K4me3 level,which subsequently causes a genome-wide decrease in H2Aub,revealing a strong association between H3K4me3 and ***,we discover that the PAL domain of AL proteins interacts with various other chromatin-related proteins or complexes,including those involved in regulating H2A.Z deposition,H3K27me3 demethylation,histone deacetylation,and chromatin *** genome-wide analysis suggests that the AL proteins play a crucial role in coordinating H3K4me3 with multiple other chromatin modifications across the genome.

Early diagnosis and accurate boundary delineation are the key steps of tumor precision *** tumor cells(CTCs)detection of liquid biopsy can provide abundant information for early diagnosis of *** detection specificity and good enrichment features are two key factors for CTCs accurate identification in peripheral blood *** this purpose,iron oxide(IO)-based surface-enhanced Raman scattering(SERS)bioprobes with good biocompatibility,high detection sensitivity,remarkable detection specificity,and good enrichment efficiency,were developed for detecting different types of *** SERS bioprobes combined with programmed death ligand-1(PD-L1)antibody are regarded as an effective way to boost the targeting ability and detection specificity,benefiting for accurately capturing and identifying rare *** types of CTCs with different PD-L1 expression were accurately distinguished among white blood cells via high-resolution SERS mapping images and stable Raman ***,CTCs blood samples obtained from the triple negative breast cancer patients were also successfully recognized compared to that of health people,indicating IO@AR@PDA-a PD-L1 SERS bioprobe possessed great potential for CTCs detection in liquid ***,IO-based bioprobe exhibited excellent dual-modal imaging abilities of high-resolution SERS imaging mode and microimaging magnetic resonance imaging *** two highly complementary imaging modes endowed IO-based bioprobes unrivalled capacity in tumor boundary differentiation,supporting tumor accurate resection and precise *** our best knowledge,this is the first time that biocompatible IO-based SERS bioprobes without noble metal element were reported not only for CTCs accurate detection,but also for precise tumor boundary delineation,showing great advantages in tumor diagnosis and treatment.
