Design and characterization of monolaurin loaded electrospun shellac nanofibers with antimicrobial activity

The aim of this study was to elucidate the optimized fabrication factors influencing the formation and properties of shellac(SHL) nanofibers loaded with an antimicrobial monolaurin(ML). The main and interaction effects of formulation and process parameters including SHL content(35%–40% w/w), ML content(1%–3% w/w), applied voltage(9–27 kV) and flow rate(0.4–1.2 ml/h) on the characteristic of nanofibers were investigated through a total of 19 experiments based on a full factorial design with three replicated center points. As a result, the SHL content was the major parameter affecting fiber diameter. Another response result revealed that the SHL content would be also the most significant negative impact on amount of beads. An increase in the concentration of SHL leaded to a reduction in the amount of beads. From the results of characterization study, it was proved that ML might be entrapped between the chains of SHL during the electrospinning process exhibiting an excellent encapsulation. According to the response surface area, small(?488 nm) and beadless(?0.48) fibers were obtained with the SHL and ML contents of 37.5% and 1.1% w/w respectively, at the applied voltage of 18 k V and the flow rate of 0.8 ml/h. In addition, the results of the kill-kinetic studies showed that SHL nanofibers loaded with ML exhibited an excellent antibacterial activity against Staphylococcus aureus, while Escherichia coli was less affected due to the hydrophilic structure of the its outer membrane. ML also exerted an antifun-gal activity by reducing the number of Candida albicans colonies. Based on their structural and antimicrobial properties, SHL nanofibers containing ML could be potentially used as a medicated dressing for wound treatment.

Inhibition of subicular seizure-labeled c-fos+ neurons alleviates cognitive deficit in epilepsy

OBJECTIVE Cognitive deficit is a common comorbidity in temporal lobe epilepsy(TLE) and that is not well controlled by current therapeutics. Currently,how epileptic seizure affects cognitive performance remains largely unclear. The subiculum is the major output of the hippocampus, which projects to entorhinal cortex and other more distinct brain regions. Physiologically,the subiculum codes spatial working memory and navigation information including place, speed, and ***, prior studies have noted the importance of the subiculum in the beginning, spreading, and generalizing process of hippocampal seizure. How seizure-activated neurons in subiculum participate in cognitive impairment remains largely elusive. METHODS In this study, we sought to label the subicular seizure-activated c-fos+neurons with a special promoter with enhanced synaptic activity-responsive element E-SARE in the subiculum,combined with chemogenetics and designer receptors exclusively activated by designer drugs(DREADDs), Ca2+fiber photometry approaches, and behavioral tasks, to reveal the role of these neurons in cognitive impairment in epilepsy. RESULTS We found that chemogenetic inhibition of subicular seizure-tagged c-fos+neurons(mainly CaMK Ⅱ α+glutamatergic neurons) alleviates seizure generalization and improves cognitive performance in the hippocampal CA3 kindling TLE model. While inhibition of seizure-labeled c-fos+GABAergic interneuron shows no effect on seizure and cognition. As a comparison, chemogenetic inhibition of the whole subicular CaMK Ⅱ α+neuron impairs cognitive function in na?ve mice in basal condition. Notably, inhibition of subicular seizure-tagged c-fos+neurons enhances the recruitment of cognitionresponsive c-fos+neurons via increasing neural excitability during cognition tasks. CONCLUSION Our results demonstrate that subicular seizure-activated c-fos+neurons contribute to cognitive impairment in TLE, suggesting seizure-tagged c-fos+neurons as the potential

There is little information about drivers’body balance responses to combined exposure of noise and *** fill the gap,this study aims to investigate the combined effects of exposure to noise and whole-body vibration(WBV)on the body balance under simulated driving *** this purpose,30 male participants were exposed to noise level at 85 dB(A)and two vibration levels(0.87 and 1.3 m/s^(2))in five *** design of the study was repeated-measures,and it attempted to assess the effects of 40 minutes of exposure to noise and/or ***,the participants’fatigue was measured with the Borg scale(CR 10).The findings revealed there was a significant change in body sway after WBV and combined noise and WBV exposure(p0.05).The effect sizes of exposure to noise,WBV(1.3 m/s^(2)),and combined noise and WBV(1.3 m/s^(2))on body balance were 0.035,0.425,and 0.635,***,single exposure to WBV caused more fatigue than single exposure to noise(p<0.05).Combined noise and WBV exposure descriptively caused more fatigue in comparison with the influence of WBV *** study concluded that the combined effects of exposure to noise and vibration are more than the sum of ***,some synergistic effects may be observed in human body *** is essential to increase drivers’awareness and revise current health care interventions about new possible effects of combined exposures.

Synthesis and Structure Analysis of a Tripeptide Containing N-methyl Group Amino Acid

A convenient method of synthesizing a tripeptide-containing N-methyl group amino acid was developed using O-benzotriazole-N,N,N＇,N＇-tetramethyluronium-hexafluorophosphate as the condensing agent. The crystals of tripeptide had white needles belonging to the orthorhombic space group P2_12_12_1. The conformational preference for homochiral tripeptides with one N-methylated amide bond was also investigated. Crystal-structure analysis showed that homochiral tripeptides with an internal N-methylated amide bond preferred a trans-amide form, thereby giving the peptide β-fold characteristics. Intermolecular C-H···O and N-H···O hydrogen bonds linked the molecules into a one-dimensional chain and stabilized the structure.

The aim of the present work is to develop rifampicin loaded phospholipid lipospheres containing sulfphobutyl etherβ-cyclodextrin and Vitamin C for inhalation to test their potential for deep lung *** findings of the solid state characterization revealed the amorphous nature of the *** exhibited a better flowability,an aerodynamic diameter in the range of 1.76 to 3.99μ***,the fine particle fraction and emitted dose was found in the range of 68.84–83.73% and 80–93%,***,lipospheres exhibited enhanced/equivalent efficacy in vitro in H37Rv ***,the results show the potential of lipospheres for pulmonary delivery of rifampicin.

Gene expression profiles of hepatic cell-type specific marker genes in progression of liver fibrosis

AIM: To determine the gene expression profile data for the whole liver during development of dimethylni-trosamine (DMN)-induced hepatic ***: Marker genes were identified for different types of hepatic cells, including hepatic stellate cells (HSCs), Kupffer cells (including other inflammatory cells), and hepatocytes, using independent temporal DNA microarray data obtained from isolated hepatic cells. RESULTS: The cell-type analysis of gene expression gave several key results and led to formation of three hypotheses: (1) changes in the expression of HSC-specific marker genes during fibrosis were similar to gene expression data in in vitro cultured HSCs, suggesting a major role of the self-activating characteristics of HSCs in formation of fibrosis; (2) expression of mast cell-specific marker genes reached a peak during liver fibrosis, suggesting a possible role of mast cells in formation of fibrosis; and (3) abnormal expression of hepatocyte-specific marker genes was found across several metabolic pathways during fibrosis, including sulfur-containing amino acid metabolism, fatty acid metabolism, and drug metabolism, suggesting a mechanistic relationship between these abnormalities and symptoms of liver fibrosis. CONCLUSION: Analysis of marker genes for specific hepatic cell types can identify the key aspects of fibro-genesis. Sequential activation of inflammatory cells and the self-supporting properties of HSCs play an important role in development of fibrosis.

In clinical practice,we noticed that triple negative breast cancer(TNBC)patients had higher shear-wave elasticity(SWE)stiffness than non-TNBC patients and a higherα-SMA expression was found in TNBC tissues than the non-TNBC ***,SWE stiffness also shows a clear correlation to neoadjuvant response *** elaborate this phenomenon,TNBC cell membrane-modified polylactide acid-glycolic acid(PLGA)nanoparticle was fabricated to specifically deliver artesunate to regulate SWE stiffness through inhibiting CAFs functional *** tested in MDA-MB-231 and E0771 orthotopic tumor models,CAFs functional status inhibited by 231M-ARS@PLGA nanoparticles(231M-AP NPs)had reduced the SWE stiffness as well as attenuated hypoxia of tumor as tumor soil loosening agent which amplified the antitumor effects of paclitaxel and PD1 ***-cell sequencing indicated that the two main CAFs(extracellular matrix and wound healing CAFs)that produces extracellular matrix could influence the tumor SWE stiffness as well as the antitumor effect of ***,biomimetic nanoparticles inhibited CAFs function could attenuate tumor hypoxia by increasing proportion of inflammatory blood vessels and oxygen transport ***,our finding is fundamental for understanding the role of CAFs on affecting SWE stiffness and drugs antitumor effects,which can be further implied in the potential clinical theranostic predicting in neoadjuvant therapy efficacy through non-invasive analyzing of SWE imaging.

Oxidative Degradation of 2,4,6-Trichlorophenolin the Presence of Air Ions

2,4,6-trichlorophenol (TCP) was firstly treated with air ions generated by electric discharge, and high dechlorination (53%) was observed after 60 min treatment, indicating that air ions are an efficient dechlorination means and might have a future in the oxidative removal of chlorinated phenols. In addition, a stepwise degradation of TCP, beginning with the formation of a major product 2,6-dichloro-1,4-benzenediol via substitution, is proposed through a detailed analysis of gas chromatography/mass spectrometry.

Surface modified solid lipid microparticles based on homolipids and Softisan 142:preliminary characterization

ObjectiveTo preliminarily investigate three different lipid matrices consisting of two natural homolipids from Capra hircus (goat fat) and Bovine Spp. (tallow fat) and one semi-synthetic lipid (Softisan? 142) separately structured with Phospholipon? 90G (P90G) as potential delivery systems for poorly water soluble drugs.MethodsThe structured lipid matrices were characterized by differential scanning calorimetry (DSC) and employed to prepare solid lipid microparticles (SLMs) by the melt homogenization method using gradient concentrations of polysorbate 80 and at different emulsification times of 2, 5 and 10 min using a Silverson mixer. The SLMs were analyzed for morphology and particle size, thermal properties, stability studies and determination of injectability.ResultsThe results showed that SLM production was optimum at 5 % of lipid matrices, 1.5 % of polysorbate 80 and emulsification time of 5 min. Increase in polysorbate 80 concentrations decreased the particle size of the SLMs. The SLMs were well formed, spherical, smooth and non-porous with particle sizes in the ranges of (13.90 ± 2.10) μm - (0.09 ± 0.01) μm for SLMs produced from the structured - tallow fat; (13.40 ± 1.30) μm - (0.10 ± 0.01) μm for the structured - goat fat and (13.40±2.00) μm - (2.10± 1.00)μm for the structured Softisan? 142 lipid matrices. DSC traces showed that Softisan? 142 was the most crystalline of all three bulk matrices due to its high enthalpy (?7.962 mW/mg) while tallow fat was the least (?5.067 mW/mg) but addition of P90G to the matrices lowered their enthalpies mostly in the structured goat fat matrices. The SLMs when stored at 4-6 ° were most stable and syringeable with 27 G needle.ConclusionsThis suggests that structured goat fat matrices with the enthalpy of ?2.813 mW/mg will mostly favour drug loading of some poorly soluble drugs more than tallow fat (?4.892 mW/mg) and Softisan? 142 (?5.501 mW/mg).
