As the population ages,the burden of age-related diseases becomes ***,over 55 million people suffer from dementia worldwide,with Alzheimer’s disease being the most common ***,it is becoming clearer that underlying vascular pathology such as cerebral small vessel disease(cSVD)may be a more detrimental cause for dementia(Cuadrado-Godia et al.,2018).It is estimated that 10%-30%of the elderly population and 35%-90%of all dementia patients exhibit signs of *** term cSVD refers to pathology affecting the small vessels of the brain,which can lead to lacunar cerebral infarcts,enlarged perivascular spaces,and cortical hemorrhages(Cuadrado-Godia et al.,2018).CSVD is often associated with cognitive decline,gait problems,and dementia(Cuadrado-Godia et al.,2018).

This study was conducted to investigate the effects of glycerol monolaurate(GML)on lipopolysaccharide(LPS)-induced immunological stress and intestinal mucosal injury in broilers and its underlying mechanisms.A total of 144 one-d-old Arbor Acres broilers were allocated to a 22 factorial arrangement involving dietary treatment(0 or 1,200 mg/kg dietary GML)and LPS challenge(injected with saline or Escherichia coli LPS on d 16,18,and 20).Samples were collected on d *** results revealed that dietary GML augmented serum immunoglobulin A(P=0.009)and immunoglobulin G(P<0.001)levels in challenged *** GML normalized LPS-induced variations in serum interleukin-6,interferongamma,and LPS levels(P<0.05),jejunal villus height(P=0.030),and gene expression of interleukin-6,macrophage inflammatory protein-3 alpha,Toll-like receptor 4,nuclear factor kappa-B,caspase-1,tight junction proteins,adenosine monophosphate-activated protein kinase alpha 1(AMPKa1),nuclear factorerythroid 2-related factor 2(Nrf2),and superoxide dismutase-1(P<0.05).GML supplementation ameliorated LPS-induced peroxidation by reducing malondialdehyde content and increasing antioxidant enzyme activity(P<0.05).Dietary GML enhanced the abundances of Anaerostipes,Pseudoflavonifractor,and Gordonibacter and reduced the proportion of Phascolarctobacterium in challenged *** GML was positively correlated with alterations in antioxidant enzyme activities and AMPKa1,Nrf2,and zonula occludens-1 *** genera Anaerostipes,Lachnospira,Gordonibacter,Lachnospira,Marvinbryantia,Peptococcus,and Pseudoflavonifractor were linked to attenuated inflammation and improved antioxidant capacity of challenged *** conclusion,dietary GML alleviated LPS-induced immunological stress and intestinal injury of broilers by suppressing inflammation and oxidative *** GML regulated cecal microbiota and activated the AMPK/Nrf2 pathway in LPS-challenged broilers.

The N-terminal EF-hand calcium-binding proteins 1–3(NECAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one EF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NECAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NECAB2 interacts with predominantly striatal G-protein-coupled receptors, while NECAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NECAB proteins exhibit distinct subcellular localizations: NECAB1 is found in the nucleus and cytosol, NECAB2 resides in endosomes and the plasma membrane, and NECAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NECAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NECAB proteins by controlling the availability of protein–protein interaction domains situated between the EF hands and the antibiotic biosynthesis monooxygenase domain. In the pathogenesis of major depressive disorder, chronic stress-related neuroinflammation hinders favorable prognosis and antidepressant response. mitochondrial DNA may be an inflammatory trigger, after its release from stress-induced dysfunctional central nervous system mitochondria into peripheral circulation. This evidence supports the potential use of peripheral mitochondrial DNA as a neuroinflammatory biomarker for the diagnosis and treatment of major depressive disorder. Herein, we critically review the neuroinflammation theory in major depressive disorder, providing compelling evidence that mitochondrial DNA release acts as a critical biological substrate, and that it constitutes the neuroinflammatory disease pathway. After its release, mitochondrial DNA can be carried in the exosomes and transported to extracellular spaces in the central nervous system and peripheral circulation. Detectable exosomes render encaged mitochondrial DNA relatively stable. This mitochondrial DNA in peripheral circulation can thus be directly detected in clinical practice. These characteristics illustrate the potential for mitochondrial DNA to serve as an innovative clinical biomarker and molecular treatment target for major depressive disorder. This review also highlights the future potential value of clinical applications combining mitochondrial DNA with a panel of other biomarkers, to improve diagnostic precision in major depressive disorder.

Background: To study the antidiabetic effects and mechanisms of the fenugreek extracts in streptozotocin(STZ)-induced type 2 diabetic(T2 DM) mice fed a high-fat diet(HFD).Methods: We established C57 BL/6 J mice model of T2 DM using HFD-fed and STZinduced method. Then, the mice were administered with two types of fenugreek extracts(E1, flavonoid and E2, stilbene glycoside) for 4 weeks and the effects on fasting blood glucose(FBG), weight, superoxide dismutase(SOD), catalase(CAT),malondialdehyde(MDA), and pathological indexes were ***: Administration of fenugreek extracts decreased the FBG level compared with that of the model group. Comparatively, the high-dose E2 decreased the FBG more significantly than the other treatments did. Both extracts showed an obvious antioxidant effect by increasing serum SOD and CAT activities and decreasing the MDA content. Furthermore, the high-dose E1 showed a significant difference(P <.01) compared with the model group in the three investigated ***: Our study demonstrated that both the flavonoid and stilbene glycoside extracts of fenugreek improved the hyperglycemia in the T2 DM mice model. Moreover, the antidiabetic effects of both extracts might be due to their antioxidant activity in vivo.

Calcium(Ca2+) is an essential nutrient and a cellular signal in plants. The level of free Ca2+in the apoplast is ～10,000-fold higher than that in the cytoplasm at the resting state. This large Ca2+level gradient across the membrane between the interior and exterior can cause Ca2+overload and toxicity. Plants have evolved mechanisms to export Ca2+out of the cytoplasm or store it in intracellular organelles, such as endoplasmic reticulum（ER）, mitochondria, and vacuole.
