Development of a droplet digital polymerase chain reaction assay for the sensitive detection of total and integrated HIV-1 DNA

Background:Total human immunodeficiency virus(HIV)DNA and integrated HIV DNA are widely used markers of HIV *** digital polymerase chain reaction(ddPCR)can be used for absolute quantification without needing a standard ***,we developed duplex ddPCR assays to detect and quantify total HIV DNA and integrated HIV ***:The limit of detection,dynamic ranges,sensitivity,and reproducibility were evaluated by plasmid constructs containing both the HIV long terminal repeat(LTR)and human CD3 gene(for total HIV DNA)and ACH-2 cells(for integrated HIV DNA).Forty-two cases on stable suppressive antiretroviral therapy(ART)were assayed in total HIV DNA and integrated HIV *** coefficient analysis was performed on the data related to DNA copies and cluster of differentiation 4 positive(CD4^(+))T-cell counts,CD8^(+)T-cell counts and CD4/CD8 T-cell ratio,*** assay linear dynamic range and lower limit of detection(LLOD)were also ***:The assay could detect the presence of HIV-1 copies 100%at concentrations of 6.3 copies/reaction,and the estimated LLOD of the ddPCR assay was 4.4 HIV DNA copies/reaction(95%confidence intervals[CI]:3.6-6.5 copies/reaction)with linearity over a 5-log_(10)-unit range in total HIV DNA *** the integrated HIV DNA assay,the LLOD was 8.0 copies/reaction(95%CI:5.8-16.6 copies/reaction)with linearity over a 3-log 10-unit *** HIV DNA in CD4^(+)T cells was positively associated with integrated HIV DNA(r=0.76,P<0.0001).Meanwhile,both total HIV DNA and integrated HIV DNA in CD4^(+)T cells were inversely correlated with the ratio of CD4/CD8 but positively correlated with the CD8^(+)T-cell ***:This ddPCR assay can quantify total HIV DNA and integrated HIV DNA efficiently with robustness and *** can be readily adapted for measuring HIV DNA with non-B clades,and it could be beneficial for testing in clinical trials.

Importance:Systemic lupus erythematosus(SLE)is a diffuse connective tissue disease with complex clinical manifestations and prolonged *** early diagnosis and condition monitoring of SLE are crucial to disease ***:To assess the diagnostic value of long noncoding RNA(lncRNA)nuclear enriched abundant transcript 1(NEAT1)in childhood-onset SLE(cSLE).Methods:Fifty-seven children diagnosed with SLE,40 children diagnosed with juvenile idiopathic arthritis(JIA),and 40 healthy children were *** blood samples from each patient were collected.A quantitative polymerase chain reaction was used to confirm the expression of lncNEAT1_1 and lncNEAT1_2 in peripheral *** among parameters were analyzed using the Mann-Whitney U test or independent sample ***:The expression of both lncNEAT1_1 and lncNEAT1_2 in patients with cSLE were significantly higher than that of healthy control and patients with *** operating characteristic curves revealed an area under the curve(AUC)of 0.633(95%confidence interval[CI],0.524-0.742;P=0.024)for lncNEAT1_*** AUC of lncNEAT1_2 was 0.812(95%CI,0.727-0.897;P<0.0001)to discriminate individuals with cSLE from health control and children with JIA with a sensitivity of 0.622 and a specificity of ***,lncNEAT1_2 expression was higher in patients with cSLE presenting with fever,lupus nephritis,elevated erythrocyte sedimentation rate,active disease activity,and decreased C3 level,compared with those without these ***,no similar correlation was observed for lncNEAT1_***:The expression of lncNEAT1_2 was significantly elevated in children with SLE,especially those with fever,renal involvement,and low C3 *** findings suggest that lncNEAT1_2 may represent a potential biomarker for cSLE.

Skin wounds are characterized by injury to the skin due to trauma,tearing,cuts,or *** such injuries are common to all human groups,they may at times represent a serious socioeconomic ***,increasing numbers of studies have focused on the role of mesenchymal stem cell(MSC)-derived extracellular vesicles(EVs)in skin wound *** a cell-free therapy,MSC-derived EVs have shown significant application potential in the field of wound repair as a more stable and safer option than conventional cell *** based on MSC-derived EVs can significantly promote the repair of damaged substructures,including the regeneration of vessels,nerves,and hair *** addition,MSC-derived EVs can inhibit scar formation by affecting angiogenesis-related and antifibrotic pathways in promoting macrophage polarization,wound angiogenesis,cell proliferation,and cell migration,and by inhibiting excessive extracellular matrix ***,these structures can serve as a scaffold for components used in wound repair,and they can be developed into bioengineered EVs to support trauma *** the formulation of standardized culture,isolation,purification,and drug delivery strategies,exploration of the detailed mechanism of EVs will allow them to be used as clinical treatments for wound *** conclusion,MSCderived EV-based therapies have important application prospects in wound *** we provide a comprehensive overview of their current status,application potential,and associated drawbacks.

The application of metalDorganic frameworks(MOFs) is hindered by several crucial factors, including low photocatalytic efficiency, poor stability and challenges in recycling. In this study, MIL-53(Fe) was grown in situ on a charcoal sponge(CS) with a large surface area,excellent mechanical properties, and high photocatalytic efficiency using a solvothermal method, thus addressing the above-mentioned shortcomings of MOF ***, CS/MIL-53(Fe) has an amazing light-harvesting capacity in the visible range, leading to a significant improvement in the utilization of light. The results of the experiments indicate that CS/MIL-53(Fe) displays a higher photocatalytic activity(87% Cr(VI)) when exposed to visible light than CS(58%) or MIL-53(Fe)(27%),attributing to the increased separation of photogenerated carriers. e-is the main reactive radical and dominates the photocatalytic reduction process, rooting from that e-can reduce Cr(VI) to low-toxicity Cr(III). This study presents a novel method for constructing photocatalysts with large surface areas, superior mechanical characteristics, and enhanced photocatalytic performance. Additionally, it achieves the resource utilization of agricultural and forestry waste.
