Unusual Acute Onset of Symptomatic Langerhans Cell Histiocytosis of the Skull: Case Report and Literature Review

Introduction: Langerhans cell histiocytosis (LCH) is a group of idiopathic disorders characterized by the proliferation of specialized bone marrow-derived Langerhans cells (LCs) and mature eosinophils, resulting in solitary or few, indolent and chronic, lesions of bone or other organs called eosinophilic granulomas. Calvarial LCH is quite rare and an underappreciated differential etiology of skull lesions. We present a most unusual case of a young child with hyperacutely symptomatic langerhans histiocytosis of the skull. Method: A 7-year-old male presented with a history of increasing (progressive) frontal headaches of 8 days duration, unaccompanied by associated nausea, vomiting, or diplopia. His only additional complaint was a hard bump on his forehead. MRI and CT done in the ER identified a right fronto-parietal lesion with associated skull erosion. nuclear medicine and SPECT studies confirmed an erosive skull lesion without significant metabolic activity. A right frontal craniectomy and excision was performed. Results: A soft, rubbery well-circumcised mass coming from the diploic layer of the skull with involvement of bone was identified. The mass had eroded both the outer and inner table of the skull, and the involved area of the right frontal bone was resected. Intra-operative histo-pathologic analysis of the lesion revealed Langerhans cell histiocytosis without involvement of the dura. The patient experienced no neurological worsening as a result of the resection. He was discharged home in stable condition. Conclusion: LCH lesions of the skull are common findings, however, this focal hyperacute symptomatic presentation is most rare and should not deter us from anticipating an erosive bony tumor and planning timely surgical management.

We have examined primary tumor sections from melanoma patients by immunohistochmistry (IHC) for the presence of the odontogenic ameloblast-associated protein (ODAM). Within these patient tissues we have observed a correlation of nuclear ODAM staining in the primary tumors with sentinel lymph node (SLN) metastasis. Surgically, SLN invasion in melanoma is considered an important indicator of more aggressive, invasive melanoma and to date there are limited biomarkers which strongly correlate with metastatic disease. The observation that ODAM staining in melanoma associates with SLN invasion may have important prognostic implications which could assist in the management of melanoma. Notably, ODAM expression may correlate with pathway-signaling we have previously reported to be affected by ectopic ODAM expression in cultured melanoma and breast cancer cell lines.

Ultrastructural Aspects of Apoptosis in Systemic Sclerosis Inflammatory Myopathy

Muscle biopsies from two female patients with systemic sclerosis (SS) and an inflammatory myopathy were studied ultrastructurally in relation to the possible presence of apoptosis in skeletal muscle fibers. Undergoing apoptosis showed characteristic morphological features of this process, including chromatin aggregation as well as nuclear and sarcoplasmic partition into membrane bound-vacuoles (apoptotic bodies) which contained autophagosomes, mitochondria, isolated myofilaments and nuclear material. Vacuoles exhibited different diameters and were covered by single membranes, appearing beneath basement membrane. Apoptosis occurred in some fiber segments as in necrosis or included whole atrophied fibers. These results indicate that apoptosis coexists with necrosis in the inflammatory myopathy of SS.

Splicing of precursor messenger RNA(pre-m RNA), involving removal of the non-coding introns and ligation of neighboring exons, is a defining feature for all *** splicing contributes to numerous debilitating diseases. Pre-m RNA splicing is accomplished by a dynamic supramolecular complex known as the spliceosome. A complete cycle of pre-m RNA splicing requires at least six distinct states of the spliceosome: the pre-catalytic complex(B complex), the activated complex(Bact complex), the catalytically competent complex(B* complex), the catalytic step I complex(C complex), the post-catalytic complex(P complex), and the intron lariat complex(ILS complex). In this presentation, I describe our systematic investigationinto thestructures of the yeast spliceosome using single-particle cryo-electron microscopy.A series of atomic structures identify the spliceosome as a protein-directed metalloribozyme.A portion of the U2 small nuclear RNA(sn RNA) undergoes a concerted set of dazzling molecular choreography to sequentially deliver the two exon-intron junctions to the active site for the trans-esterification reactions. Remarkably, the U6 sn RNA, which harbors the catalytic nucleotides, and U5 sn RNA, which anchors the RNA elements of the spliceosome, remain largely static throughout the splicing cycle.

Ganoderma lucidum polysaccharide inhibits LPS-induced inflammatory injury to mammary epithelial cells

This study sought to investigate whether Ganoderma lucidum polysaccharide(GLP)has a protective effect on lipopolysaccharide(LPS)-induced inflammatory injury to mammary epithelial HC-11 cells and to characterize the mechanism *** viability was assessed using the cell counting kit 8(CCK-8)method,tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and IL-1βlevels were measured by enzyme linked immunosorbent assay(ELISA),and IκBa,p65 NF-κB and STAT3 mRNA were determined using quantitative reverse transcription PCR(qRT-PCR),p65 and STAT3 protein expression were determined using Western blotting,*** was shown to inhibit LPS-induced TNF-α,IL-6,and IL-1βproduction(Pnuclear factor kappa-B(NF-κB)and signal transducers and activators of transcription(STAT)signaling by preventing IκBαdegradation and p65 and STAT3 *** results in lower LPS-induced TNF-α,IL-6,and IL-1βproduction and prevents inflammatory cell injury.

Grape seed proanthocyanidin protects liver against ischemia/reperfusion injury by attenuating endoplasmic reticulum stress

AIM: To explore the effect of grape seed proanthocyanidin(GSP) in liver ischemia/reperfusion(IR) injury and alleviation of endoplasmic reticulum ***: Male Sprague-Dawley rats(220-250 g) were divided into three groups, namely, sham, IR, and GSP groups(n = 8 each). A liver IR(70%) model was established and reperfused for 6 h. Prior to reperfusion, the GSP group was administered with GSP(100 mg/kg) for 15 d, and liver histology was then investigated. Serum aminotransferase and inflammatory mediators coupled with superoxide dismutase and methane dicarboxylic aldehyde were detected. Western blot was conducted to analyze the expression of glucoseregulated protein 78, CCAAT/enhancer-binding protein homologous protein, activating transcription factor-4, inositol-requiring enzyme-1, procaspase-12, and nuclear factor-κb. Apoptotic cells were detected by TUNEL ***: The serum aminotransferase, apoptotic cells, and Suzuki scores decreased in the GSP group compared with the IR group(P s nuclear factor-κb compared with the IR ***: GSP possesses antioxidative, anti-inflammatory, and antiapoptotic effects by relieving endoplasmic reticulum stress through regulation of related signaling pathways to protect the liver against IR injury.

The recent advances in functional genomics have discovered that a large number of long non-coding RNAs(lncRNAs)are pervasively transcribed from the human *** evidence further indicates that lncRNAs are important for gene expression during cell differentiation and development through various mechanisms such as nuclear organization,post-transcription regulation,alternative splicing,and epigenetic ***,aberrant expression of lncRNAs can cause abnormality in those cellular functions and lead to various pathological *** of such fatal consequences is cancer metastasis which is responsible for more than 90%of cancer-related deaths.A good understanding of how lncRNAs regulate different genetic and epigenetic changes during different stages of cancer metastasis is important not only for general cancer biology but also for identification of novel biomarkers and therapeutic targets for treatment of metastatic cancer.A significant progress has been made regarding the role of lncRNAs in cancer for past several *** this study,we first discuss general functions of lncRNAs and then highlight recent findings of how lncRNAs impact cancer metastasis,and finally we provide our perspectives on clinical implications of lncRNAs.

The pervasive transcription of the human genome has spurred efforts to identify and functionally characterize non-coding RNAs that are longer than 200 nts(lncRNAs),but very few molecular mechanisms have been revealed due to an insufficient understanding of lncRNA *** chemical and enzymatic probing techniques such as SHAPE have been widely utilized for lncRNA secondary structural characterization,three dimensional structural characterization of lncRNAs which usually has increased flexibility and large size is challenging to traditional techniques like X-ray crystallography(XRC),nuclear magnetic resonance(NMR) and cryo-electron microscopy(cryo-EM).Recent progress in small angle x-ray scattering(SAXS) has made it a powerful tool in characterizing the structure,dynamics and interaction of biomacromolecules including large RNAs in ***,we show that small angle x-ray scattering in combination with computation can be utilized to study the topological structures,dynamics and interactions of the HIV-1 rev response element(233 nt),the T-box riboswitch core(183 nt) and the long non-coding subgenomic flavivrus rnas(400～500 nt),thereby providing structural insights into their functions and information on accessible conformational space in *** study also highlights a general SAXS-based strategy for structural studies of other functional lncRNAs in solution.

RNA is distributed asymmetrically in eukaryotic cells, which underlies the basis of organismal development and protein local translation. The subcellular localization of RNA plays important roles in biological process, including gene expression regulation, cell structure supporting and stress response. With the development of technologies, including image-based fluorescence in situ hybridization, physical separation of subcellular components and subcellular proximity labeling, our knowledge of subcellular transcriptome has greatly advanced in the last few decades. APEX-seq[1] makes it possible to direct label adjacent transcriptome. However, the low labeling efficiency of APEX-seq leads to poor sensitivity and high demand of sample amount. Here, we introduced an approach to identify the subcellular transcriptome efficiently under introducing the nucleoside analogues with highly activated thione, 4-thiouridine and 6-thioguanosine[2], into APEX-seq labeling. Using this method, we investigated the local transcriptome of mitochondrial matrix and nuclear lamina. In mitochondial matrix, the experiment enriched all 13 mRNAs and the 2 rRNAs encoded by mtDNA. In neclear lamina, this technology highly enriched many mRNAs tend to code nuclear associated proteins. This discovery may indicate these enriched RNAs could participate in the regulation of gene expression suppression, which needs more verification.
