Genes and proteins expressed at different life cycle stages in the model protist Euplotes vannus revealed by both transcriptomic and proteomic approaches

Sexual reproduction first appeared in unicellular protists and has continued to be an essential biological process in almost all *** protists, which contain both germline and somatic genomes within a single cell, have evolved a special form of sexual reproduction called conjugation that involves mitosis, meiosis, fertilization, nuclear differentiation, genome rearrangement, and the development of unique cellular structures. The molecular basis and mechanisms of conjugation vary dramatically among ciliates, and many details of the process and its regulation are still largely unknown. In order to better comprehend these processes and mechanisms from an evolutionary perspective, this study provides the first comprehensive overview of the transcriptome and proteome profiles during the entire life cycle of the newly-established marine model ciliate Euplotes vannus. Transcriptome analyses from 14 life cycle stages(three vegetative stages and 11 sexual stages) revealed over 26,000 genes that are specifically expressed at different stages, many of which are related to DNA replication,transcription, translation, mitosis, meiosis, nuclear differentiation, and/or genome rearrangement. Quantitative proteomic analyses identified 338 proteins with homologs associated with conjugation and/or somatic nuclear development in other ciliates, including dicer-like proteins, Hsp90 proteins, RNA polymerase Ⅱ and transcription elongation factors, ribosomal-associated proteins, and ubiquitin-related proteins. Four of these homologs belong to the PIWI family, each with different expression patterns identified and confirmed by RT-qPCR,which may function in small RNA-mediated genome rearrangement. Proteins involved in the nonhomologous end-joining pathway are induced early during meiosis and accumulate in the developing new somatic nucleus, where more than 80% of the germline sequences are eliminated from the somatic genome. Insecticide resistance status and biochemical mechanisms involved in Aedes mosquitoes: A scoping review

Mosquitoes belonging to the genus Aedes pose a significant threat to human health on a global scenario due to their role in transmission of dengue,chikungunya,zika,and yellow *** absence of specific medications and vaccines against these diseases,disease prevention relies on vector ***,in today’s world,vector control is facing major challenges due to the onset of insecticide resistance in *** are four main mechanisms of insecticide resistance,namely,behavioral resistance,reduced penetration/cuticular resistance,metabolic detoxification,and target site resistance;however,the latter two mechanisms have been studied widely in Aedes *** resistance in Aedes mosquitoes is widespread throughout the *** review compiles the degree of insecticide resistance/susceptibility prevailing among different field populations of Aedes mosquitoes *** addition,the review has detailed the mechanisms providing the resistance phenomenon observed in nature in Aedes mosquitoes.

Target-site mediated insecticide resistance in major mosquito(Diptera:Culicidae)vectors:A systematic review

The use of insecticides remain the most effective vector control approach for mosquito borne diseases like malaria,dengue fever,chikungunya,and Zika virus disease;however,their increasing resistance has complicated the *** in kdr,ace-1/ace-2,rdl,and nAChR,are involved in target-site mediated resistance and prevent the binding of pyrethroids and dichlorodiph enyltrichloroethane,organophosphorus pesticide,carbamates and cyclodienes,***,we review the current knowledge on target-site mediated insecticidal resistance mechanisms in major mosquito vectors and the techniques used to identify these *** results showed that kdr mutations are frequently reported in mosquito species,indicating the overuse of pyrethroid in mosquito *** on ace mutations is very limited,primarily detected in Anopheles and Culex,with extremely low detection rate in Aedes,despite the wide application of organophosphates in their ***,reports of rdl mutations and changes in nAChR are rare,which provides an opportunity to exploit these target sites,particularly in the event of disease *** various detection methods such as RFLP,TaqMan,Multiplex PCR and FRET,allele-specific PCR is mostly employed,especially in detecting kdr ***,allele-specific PCR cannot detect novel *** a result,there is an urgent need to develop a scalable,cost-effective tool that can be widely applied especially as these mutations are genetic markers for early detection of insecticide *** monitoring methods remain critical to manage insecticide resistance and effective control of mosquito-borne diseases.

Isolation and identification of bioactive antibacterial components in leaf extracts of Vangueria spinosa(Rubiaceae)

Objective:The column chromatographic fraction of ethyl acetate(EA1,EA2,EA3,EA4 and EA5) leaf extracts of Vangueria spinosa(***) were screened for antibacterial activity and phytochemical ***:EA3 fraction was isolated and identified by column chromatography,thin layer chromatography,spectral data analysis and phytochemical screening were used for ***:EA3 fraction was significantly active at 4 to 64 mg/L against Staphylococcus aureus,Escherichia coli,Klebsiella pneumoniae and Pseudomonas aeruginosa with minimum inhibitory concentration of 1.5625 to 3.1250 mg/*** active fraction(EA3) revealed the presence of flavonoid with retention factor value(R_f) of *** active antibacterial agent in the most potent fraction(EA3) was isolated and identified as flavonoid(-)-epicatechin-3-O-β-glucopyranoside by thin layer chromatography(TLC) and phytochemical ***1 and EA2 show inhibitory activity at 4 to 64 mg/L against Staphylococcus aureus only where as fraction EA4 and EA5 do not shows any inhibitory activity within that range of concentration against any ***:The results support the ethnomedicinal use of leaf of *** for the treatment of bacterial diseases.

Transcriptional co-activators: emerging roles in signaling pathways and potential therapeutic targets for diseases

Specific cell states in metazoans are established by the symphony of gene expression programs that necessitate intricate synergic interactions between transcription factors and the *** of these regulatory molecules is associated with cell state transitions,which in turn is accountable for diverse maladies,including developmental disorders,metabolic disorders,and most significantly,cancer.A decade back most transcription factors,the key enablers of disease development,were historically viewed as‘undruggable’;however,in the intervening years,a wealth of literature validated that they can be targeted indirectly through transcriptional co-activators,their confederates in various physiological and molecular *** co-activators,along with transcription factors,have the ability to initiate and modulate transcription of diverse genes necessary for normal physiological functions,whereby,deregulation of such interactions may foster tissue-specific disease ***,it is essential to analyze how these co-activators modulate specific multilateral processes in coordination with other *** proposed review attempts to elaborate an in-depth account of the transcription co-activators,their involvement in transcription regulation,and context-specific contributions to pathophysiological *** review also addresses an issue that has not been dealt with in a comprehensive manner and hopes to direct attention towards future research that will encompass patient-friendly therapeutic strategies,where drugs targeting co-activators will have enhanced benefits and reduced side *** insights into currently available therapeutic interventions and the associated constraints will eventually reveal multitudes of advanced therapeutic targets aiming for disease amelioration and good patient prognosis.

Mosquito larvicidal effect of orthophosporic acid and lactic acid individually or their combined form on Aedes aegypti

Objective:To observe the effect of two common organic acids on the larvae of Aedes aegypti (***)(L),the natural vector of dengue fever/dengue hemorrhage fever,chikugunya and allergic skin reaction especially in ***:Two common organic acids(lactic acid and orthophosporic acid of gradually increasing concentration) were used against laboratory reared third instars larvae of *** in order to observe the rate of mortality after 8.16 and 24 h of post exposure respectively in ***:Larval mortality rates recorded were in the following sequences:orthophosphoric acid and lactic acid at 1:1 combination >orthophosphoric acid>lactic ***:These two organic acids may be used perfectly in combination (1:1) along with other conventional vector control methods to reduce the *** population, especially in those areas where surveillance and supervisory mechanism are poor or insufficient.

Efficacy of Limonia acidissima L.(Rutaceae) leaf extract on larval miniatures of Culex quinquefasciatus Say 1823

Objective:To investigate the role of leaf extract of Limonia acidissima L.(Rutaceae) as a biocontrol agent against the larval form of Culex quinquefasciatus,and characterization of bioactive component responsible for larvicidal ***:Larval mortality of mosquito species was observed after 24,48 and 72 hours of exposure to different concentrations of aqueous extract,solvent extract and subsequently bioaclive *** bioaclive compound was subjected to IR and GC-MS ***:Mortality rale al 3%concentration of crude extract were highest(90%) amongsl all concentrations tested and subsequently highest(95%) mortality was achieved in chloroform:methanol extract at 100 ppm *** and GC-MS analysis of bioaclive compound revealed the presence of steroid compound which may act as larvicide. Conclusions:The chloroform:methanol extract of mature leaves of Limonia acidissima was found to exhibit considerable mosquito larvicidal activity against Culex quinquefasciatus.

Molecular evidence on the occurrence of co-infection with Pichia guilliermondii and Wuchereria bancrofti in two filarial endemic districts of India

Background:Lymphatic filariasis(LF),a vector-borne parasitic disease,is endemic in several parts of India and mostly affects the poor or those with a *** disease results in huge numbers of morbidities,disabilities,and deaths every *** of co-infection with other pathogens makes the condition more *** co-infection is becoming a growing area of research,it is yet to emerge as a frontier research topic in filarial research *** study reports the occurrence of a fungal infection in a large number of patients suffering from bancroftian filariasis in two districts of West bengal,***:Nocturnal blood samples from filarial patients containing parasites and fungus were initially co-cultured,and further the fungus was isolated and *** identification of the isolate was carried out by PCR-based selective amplification and sequencing of highly-conserved D1/D2 region of 26S rDNA,whereas pathogenicity was determined by amplification of the RPS0 gene.A phylogenetic tree was constructed to study the relationship between the isolate and common pathogenic *** isolate was studied for antibiotic sensitivity,whereas morphological characterization was performed by microscopic ***:The isolate was identified as Pichia guilliermondii and this fungus was found to exist in co-infection with Wuchereria bancrofti in filarial *** fungus showed resistance to azole antifungals,griseofulvin,and,amphotericin B,whereas significant susceptibility was evident in cases of nystatin and cycloheximide.A total of 197 out of 222 patients showed this ***:This study revealed,for the first time,that *** exists as a co-infection in microfilaraemic individuals living in a filarial endemic *** findings are important and have relevance to human health,especially for filarial patients.
