Correlation of contrast-enhanced ultrasonography parameters of ovarian cancer with angiogenesis and cancer cell growth

Objective:To study the correlation of contrast-enhanced ultrasonography parameters of ovarian cancer with angiogenesis and cancer cell ***:Patients with ovarian tumor who underwent surgical resection in the Second People's Hospital of Liangshan Yi Autonomous Prefecture between February 2015 and June 2017 were selected as the research subjects and divided into the ovarian cancer group with ovarian cancer and the control group with benign tumor according to the postoperative pathological results of tumor. Contrast-enhanced ultrasonography was performed before operation to determine the time to peak TTP and enhanced intensity EI;the tumor lesion was collected after operation to determine the protein levels of angiogenesis molecules, cell proliferation molecules and cell invasion ***: The EI of ovarian cancer lesion tissue of ovarian cancer group was significantly higher than that of benign tumor lesion tissue of control group whereas the TTP was significantly lower than that of benign tumor lesion tissue of control group;VEGFR1, Tie-2, CGB5, NFATc1, CyclinD1, FUNDC1, IGFBP2, IGFBP3 and ZEB1 protein levels in ovarian cancer lesion tissue were significantly higher than those in benign tumor lesion tissue, positively correlated with EI level and negatively correlated with TTP level;TNFSF15, Fibulin-5, ELF5, p27Kip1, p21, FN14 and E-cadherin protein levels were significantly lower than those in benign tumor lesion tissue, negatively correlated with EI level and positively correlated with TTP ***:The contrast-enhanced ultrasonography parameters of ovarian cancer can reflect the degree of angiogenesis and the viability of the invasive growth of cancer cells within lesions.

The Hexokinase inhibitor 2-Deoxy-D-glucose(2-DG) is a synthetic glucose analogue that inhibits glycolysis and glycosylation and blocks cell *** is also a classical induce of endoplasmic reticulum(ER) *** is undergoing clinical trials as an anticancer *** studies have shown that sensitivity of breast cancer cells to TNF-related apoptosis-inducing ligand (TRAIL) -induced apoptosis is largely correlated with the expression levels of TRAIL death receptors on the cell ***,fresh breast cancer isolates and breast cancer tissue sections express generally low levels of death receptors for *** clinical potential of TRAIL in the treatment of breast cancer may therefore be limited unless given with agents that increase the cell surface expression of TRAIL death *** this study we report that whether 2-DG and TRAIL enhanced sensitivity of the cells to apoptosis -induced by *** addition, which protein play the decisive role in this signaling *** result indicate that 2-DG did not kill breast cancer cells,but enhanced TRAIL-induced apoptosis in breast cancer *** 2-DG sensitizes breast cancer cells to TRAIL-induced apoptosis by down-regulation of RIP1 and up-regulation of *** suggested that 2-DG can become a promising agent to increase the therapeutic response to TRAIL in breast cancer.

Assessment of released acrosin activity as a measurement of the sperm acrosome reaction

Aim： To develop a method for assessing sperm function by measuring released acrosin activity during the acrosome reaction （AR）. Methods： Human semen samples were obtained from 24 healthy donors with proven fertility after 3-7 days of sexual abstinence. After collection, samples were liquefied for 30 min at room temperature. Standard semen parameters were evaluated according to World Health Organization （WHO） criteria. Calcium ionophore A23187 and progesterone （P4） were used to stimulate the sperm to undergo AR. After treatment, sperm were incubated with the supravital dye Hoechst33258, fixed in a glutaraldehyde-phosphate-buffered saline solution, and the acrosomal status was determined by fluorescence microscopy with fluorescein isothiocyanate-labeled Pisum sativum agglutinin （FITC-PSA）. The percentage of sperm undergoing AR （AR%） was compared to sperm acrosin activities as assessed by spectrocolorimetry. The correlation between AR% and acrosin activity was determined by statistical analysis. Results： The AR% and released acrosin activity were both markedly increased with A23187 and P4 stimulation. Sperm motility and viability were significantly higher after stimulation with P4 versus stimulation with A23187 （P 〈 0.001）. There was a significant positive correlation between released acrosin activity and AR% determined by FITC-PSA staining （r= 0.916, P 〈 0.001）. Conclusion： Spectrocolorimetric measurement of released acrosin activity might serve as a reasonable alternative method to evaluate AR. （Asian J Androl 2008 Mar, 10： 236-242）

Propofol injection combined with bone marrow mesenchymal stem cell transplantation better improves electrophysiological function in the hindlimb of rats with spinal cord injury than monotherapy

The repair effects of bone marrow mesenchymal stem cell transplantation on nervous system damage are not satisfactory. Propofol has been shown to protect against spinal cord injury. Therefore, this study sought to explore the therapeutic effects of their combination on spinal cord injury. Rat models of spinal cord injury were established using the weight drop method. Rats were subjected to bone marrow mesenchymal stem cell transplantationvia tail vein injection and/or propofol injectionvia tail vein using an infusion pump. Four weeks after cell transplan-tation and/or propofol treatment, the cavity within the spinal cord was reduced. The numbers of PKH-26-positive cells and horseradish peroxidase-positive nerve ifbers apparently increased in the spinal cord. Latencies of somatosensory evoked potentials and motor evoked potentials in the hindlimb were noticeably shortened, amplitude was increased and hindlimb motor function was obviously improved. Moreover, the combined effects were better than cell transplantation or propofol injection alone. The above data suggest that the combination of propofol injection and bone marrow mesenchymal stem cell transplantation can effectively improve hindlimb electro-physiological function, promote the recovery of motor funtion, and play a neuroprotective role in spinal cord injury in rats.

Roxadustat for treatment of anemia in a cancer patient with endstage renal disease:A case report

BACKGROUNDMost cancer patients are accompanied by anemia, which will be more seriouswhen combined with end-stage renal disease (ESRD). At present, cancer-relatedanemia and renal anemia treatments mainly include erythropoiesis-stimulatingagents (ESAs), iron supplementation, and blood transfusion, but their effects areoften poor with several safety concerns. We have used roxadustat to treat anemiain a cancer patient with ESRD and achieved a successful outcome for the *** SUMMARYA 64-year-old man was diagnosed with right renal cancer (clear cell renal cellcarcinoma). He did not receive surgery or radiotherapy before admission. He wastreated with oral soltan (sunitinib malate) on April 18, 2017. During oral chemotherapy,he had numerous complications, including anemia, hypertension,thyroid hypofunction, skin pigment loss, and renal function deterioration. At last,he progressed to ESRD and began hemodialysis treatment. We initially treated thepatient with high-dose ESAs, iron supplementation, adequate dialysis, and evenblood transfusion, but his anemia did not improve. Roxadustat is a newlydeveloped drug for renal anemia treatment, but not for cancer-related anemia, letalone to treat anemia in cancer patients with ESRD. We prescribed oral roxadustatto the patient. After a period, his hemoglobin gradually increased. He did nothave obvious discomfort symptoms, and his tumor did not progress *** roxadustat could achieve good results in treating anemia in cancer patients with ESRD.

BACKGROUND： Prenatal stress has been shown to inhibit cell proliferation in the dentate gyrus and hippocampus, reduce hippocampal volume, and cause neuronal loss and oxidative damage in the hippocampus of offspring rats, but the sexual difference of the effects on offsprings is seldom referred to. OBJECTIVE： To observe the effect of prenatal stress to adult pregnant rats on expression of extracellular signal-regulated kinases （ERK） in hippocampus of the offspring rats of different genders. DESIGN ： A randomized and control animal experiment.SETTING： Department of Physiology and Pathophysiology, School of Medicine, Xi＇an Jiaotong University. MATERIALS ： The experiments were carried out in the Key Laboratory of Environment and Gene Related Diseases （Xi＇an Jiaotong University）, Ministry of Education between October 2005 and March 2006. Fifteen female and five male adult Sprague-Dawley rats were adopted. Female rats weighing 230-250 g and male rats weighing 280-350 g were used. METHODS： The virgin female rats were placed overnight with adult male rats （3：1） for mating. A total of twelve pregnant rats were randomly assigned to prenatal stress group （PNS group, n=6） and control group （n=6）. The pregnant rats of the PNS group were exposed to restraint stress on days 14-20 of pregnancy three times a day, 45 minutes for each time . The restraint device was a transparent plastic tube （6.8 cm in diameter） with air holes for breathing and closed end. The length could be adjusted to accommodate the size of the animals. To prevent habituation of animals to the daily procedure, restraint periods were randomly shifted within certain time periods （8：00-11：00, 11：00-14：00, and 16：00-19：00）. After birth, offsprings of all groups were culled to 8-10 litters in each group and housed in the same animal room, and kept together with their biologic mothers. High-efficiency quaternary polymer solar cells enabled with binary fullerene additives to reduce nonfullerene acceptor optical band gap and improve carriers transport

Characteristic analysis of skin keratinocytes in patients with type 2 diabetes based on the single-cell levels

Background: Keratinocytes play an important role in wound healing;however, less is known about skin keratinocytes in patients with type 2 diabetes mellitus (T2DM). Therefore, this study aimed to search for the transcriptional characteristics of keratinocytes at the single-cell level from T2DM patients, and to provide experimental data for identifying the pathological mechanisms of keratinocytes under pathological ***: We performed single-cell RNA sequencing on the skin tissue from two T2DM patients and one patient without diabetes-induced trauma using the BD Rhapsody^(TM) Single-cell Analysis System. With the help of bioinformatics R-based single-cell analysis software, we analyzed the results of single-cell sequencing to identify the single-cell subsets and transcriptional characteristics of keratinocytes at the single-cell level, including Kyoto Encyclopedia of Genes and Genomes and Gene Ontology ***: In this study, we found specific highly expressed signature keratinocyte-related genes. We analyzed the transcriptome of keratinocytes from experimental and control groups and screened a total of 356 differential genes, which were subject to bioinformatics analysis. Enriched pathways included oxidative phosphorylation, antigen processing and presentation, prion and Huntingtons’ diseases, bacterial invasion of epithelial cells, thermogenesis, vasopressin-regulated water reabsorption, and protein processing in the endoplasmic ***: This study revealed the characteristics of keratinocytes at the single-cell level and screened a group of differentially expressed genes related to T2DM-associated keratinocytes, oxidative phosphorylation, cytokine receptor interactions, prion diseases, and other signaling pathways.
