Treg Immunomodulation Contributes to the Anti-atherosclerotic Effects of Huxin Formula in ApoE-/- Mice

Objective: To explore the effects of Huxin formula（HXF） in curtailing atherosclerosis and its underlying mechanism. Methods: According to random number table method, 24 specific pathogen free male ApoE-/-mice were randomly divided into model group, HXF low-dose（HXF-L） group（8.4 g/kg daily）, HXF high-dose（HXF-H） group（16.8 g/kg daily）, and pravastatin（8 mg/kg daily） group in Experiment Ⅰ（n=6 per group）. C57BL/6J mice served as the control group（n=6）. ApoE-/-mice in HXF-L, HXF-H, pravastatin groups were fed a Western diet and administered continuously by gavage for 12 weeks, while C57BL/6J mice in the control group were fed conventional lab mouse chow for 12 weeks. Further, Tregs were depleted by weekly intraperitoneal injection of purified anti-mouse CD25 antibody（PC61, 250 μg per mouse） for 4 weeks in Experiment Ⅱ（n=6 per group）. Oil Red O and Masson staining were used to evaluate the plaque area and aortic root fibrosis. The CD4+CD25+Foxp3+Treg counts in the lymph nodes and spleen cells were detected using flow cytometric analysis. The transforming growth factor-β1（TGF-β1）, interleukin（IL）-10, and IL-6 serum levels were examined by MILLIPLEX?MAP technology. Quantitative real-time reverse transcription PCR（qRT-PCR） and Western blot were utilized to assess the expression of TGF-β mRNA and protein in the aorta. The expression of CD4+T lymphocytes, macrophages and smooth muscle cells in the aortic root were detected by immunofluorescence staining. Results: HXF reduced plaque area in ApoE-/-mice（P0.05）. Also, HXF upregulated the expression of TGF-β mRNA and protein in the aorta（P<0.05）. Additionally, HXF attenuated CD4+T lymphocytes, macrophages and smooth muscle cells in aortic root plaque（P<0.01）. Minimally invasive surgery(MIS)has significantly progressed and emerged as the preferred approach for various gastrointestinal procedures,including gastrectomy,cholecystectomy,and various colorectal surgeries(1).However,the widespread acceptance of robotic surgery in liver resection has encountered obstacles,attributed to factors such as the lack of appropriate surgical instruments,the intricate learning curve,and safety ***,the utilization of MIS in liver resection is linked to decreased blood loss,reduced post-operative pain,shorter hospital stays,and lower morbidity rates compared to both open and laparoscopic liver resection procedures(2).

Exploring luminescence quenching on lanthanide-doped nanoparticles through changing the spatial distribution of sensitizer and activator

Luminescence quench is common in overdoped upconversion *** methods have been proposed to counteract the adverse effects of concentration quenching on luminescence,but in upconversion nanoparticles that are highly doped with both sensitizers and activators,the factors that contribute to the diminishing of the emission cannot be summarized by a single ***,a core-shell design is used to spatially separate the sensitizer(Yb^(3+))and activator(Er^(3+))and to modulate the emission by changes in the distribution position as well as the concentration of the dopant ions in order to probe the factors affecting the *** the sensitizer ions are located in the core,the luminescence intensity of the nanoparticles is significantly weaker than that of the other distribution,which implies that the effect of sensitizer and activator on luminescence in the highly doped state has a different and more complex *** intensity of the emission is more affected by Yb^(3+)than Er^(3+),which includes not only the self-quenching of Yb^(3+),but also the dominance in the Yb^(3+)-Er^(3+)*** this finding may provide new ideas for revealing the reasons for the diminished luminescence of highly doped upconversion nanoparticles and thus for enhancing luminescence.

Artemisia argyi extract subfraction exerts an antifungal effect against dermatophytes by disrupting mitochondrial morphology and function

Objective:To explore the potential mechanism of lysionotin in treating ***:First,target prediction based on Bernoulli Naïve Bayes profiling and pathway enrichment was used to predict the biological activity of *** binding between 5-lipoxygenase(5-LO)and lysionotin was detected by surface plasmon resonance(SPR)and molecular docking,and the inhibitory effects of lysionotin on 5-LO and proliferation of glioma were determined using enzyme inhibition assay in vitro and cell viability analysis,***,the pharmaceutical effect of lysionotin was explored by cell survival rate analysis and liquid chromatography with tandem mass spectrometry(LC-MS/MS).The protein expression,intracellular calcium ion concentration and cytoskeleton detection were revealed by Western blot,flow cytometry and fluorescence labeling,***:Target prediction and pathway enrichment revealed that lysionotin inhibited 5-LO,a key enzyme involved in the arachidonic acid metabolism pathway,to inhibit the proliferation of *** docking results demonstrated that 5-LO can be binding to lysionotin through hydrogen bonds,forming bonds with His600,Gln557,Asn554,and *** analysis further confirmed the interaction between 5-LO and ***,enzyme inhibition assay in vitro and cell survival rate analysis revealed that 50%inhibition concentration of lysionotin and the median effective concentration of lysionotin were 90 and 16.58µmol/L,respectively,and the results of LC-MS/MS showed that lysionotin inhibited the production of 5S-hydroperoxy-eicosatetraenoic acid(P<0.05),and moreover,the LC-MS/MS results indicated that lysionotin can enter glioma cells well(P<0.01)and inhibit their *** blot analysis demonstrated that lysionotin can inhibit the expression of 5-LO(P<0.05)and downstream leukotriene B4 receptor(P<0.01).In addition,the results showed that lysionotin affected intracellular calcium ion concentration by inh

Recurrent cyclops lesion after primary resection of fibroreactive nodule following anterior cruciate ligament reconstruction

In this case report featured in World Journal of Orthopedics,Kelmer et al describe a rare finding of a 28-year-old female patient who presented with a recurrent fibroreactive nodule 7 months following the resection of a primary cyclops lesion,suggesting recurrent cyclops *** patient had undergone an initial anterior cruciate ligament reconstruction for a non-contact right knee injury and reported successful *** years later,the patient sustained a repeat right knee injury followed by a positive McMurray test and acute pain with terminal *** synovectomy confirmed magnetic resonance imaging(MRI)finding of a cyclops lesion,which was surgically *** months postoperatively,the patient reported stiffness and difficulty with terminal *** MRI indicated a recurrent cyclops lesion,which was surgically *** resection of the second lesion,the patient underwent physical therapy and achieved full range of motion,maintaining complete recovery 19 months *** cyclops lesions have rarely been reported in the literature,and this article is novel in its report of recurrent cyclops syndrome following a bone-patellar tendon-bone *** presentation of this unusual finding exposes a need for further investigation of cyclops lesion pathology,which will aid its prevention and treatment.
