Dyssynchronous heart failure models in canines:New insights into electrocardiographic,echocardiographic and histological features

Background:We investigated the similarities and differences between two experimental approaches using tachy-pacing technology to induce desynchronized heart failure in ***:A total of eight dogs were included in the experiment,four were tachypaced in right ventricle apex (RVAP) and 4 were paced in right atrium after the ablation of left bundle branch to achieve left bundle branch block (RAP+LBBB).Three weeks of follow-up were conducted to observe the changes in cardiac function and myocardial staining was performed at the end of the ***:Both experimental approaches successfully established heart failure with reduced ejection fraction models,with similar trends in declining cardiac *** RAP+LBBB group exhibited a prolonged overall ventricular activation time, delayed left ventricular activation,and lesser impact on the right *** RVAP approach led to a reduction in overall right ventricular compliance and right ve ntricular *** RAP+LBBB group exhibited significant reductions in left heart compliance (LVGLS,%:RAP+LBBB-12.60±0.12 to-5.93±1.25;RVAP-13.28±0.62to-8.05±0.63, p=0.023;LASct,%:RAP+LBBB-15.75±6.85 to-1.50±1.00;RVAP-15.75±2.87 to-10.05±6.16,p=0.035).Histological examination revealed more pronounced fibrosis in the left ventricular wall and left atrium in the RAP+LBBBgroup while the RVAP group showed more prominent fibrosis in the right ventricular ***:Both approaches establish HFrEF models with comparable *** RVAP group shows impaired right ventricular function,while the RAP+LBBB group exhibits more severe decreased compliance and fibrosis in left ventricle.

Cardiac rehabilitation,a comprehensive exercise-based lifestyle and medical management,is effective in decreasing morbidity and improving life quality in patients with coronary heart *** function,an irreplaceable indicator in coronary heart disease progression,is measured by various methods in traditional cardiac rehabilitation pathways,including medicinal treatment,aerobic training,and smoking ***,studies on the effect of some emerging cardiac rehabilitation programs on endothelial function are *** article briefly reviewed the endothelium-beneficial effects of different cardiac rehabilitation pathways,including exercise training,lifestyle modification and psychological intervention in patients with coronary heart disease,and related experimental models,and summarized both uncovered and potential cellular and molecular mechanisms of the beneficial roles of various cardiac rehabilitation pathways on endothelial *** exercise training and some lifestyle interventions,the enhanced bioavailability of nitric oxide,increased circulating endothelial progenitor cells(EPCs),and decreased oxidative stress are major contributors to preventing endothelial dysfunction in coronary heart ***,the preservation of endothelial-dependent hyperpolarizing factors and inflammatory suppression play *** the one hand,to develop more endothelium-protective rehabilitation methods in coronary heart disease,adequately designed and sized randomized multicenter clinical trials should be advanced using standardized cardiac rehabilitation programs and existing assessment *** the other hand,additional studies using suitable experimental models are warranted to elucidate the relationship between some new interventions and endothelial protection in both macro-and microvasculature.

Screening optimal target populations with symptomatic bradyarrhythmia for pharmacotherapy:a discriminant analysis pilot study

OBJECTIVE: To screen the target population with symptomatic bradyarrhythmia for pharmacotherapy. METHODS: This study use database from clinical trial that use Yuanjiang decoction(元姜方), a traditional Chinese medicinal prescription. Eligible participants were recruited and treated with Yuanjiang decoction(composed of 6 Chinese herbal medicines), 200 m L twice daily. Cross-contingency analysis, logistic regression analysis, interaction analysis, discriminant analysis and 10-fold cross validation methods were used to establish discriminant model. RESULTS: This study concluded that the clinical treatment of bradyarrhythmia has a clear effect. Low minimum heart rate, high maximum heart rate are risk factors that affect the efficacy. Patients with only one comorbid disease did not significantly affect the efficacy, but patient with two or more diseases of coronary heart disease, hypertension, paroxysmal atrial fibrillation, premature ventricular contraction and premature atrial contraction at the same time did not have a good effect. Using the discriminant analysis method to establish a efficacy prediction model, y = 0.07X1 + 0.16X2-0.65X3-1.12X4-0.71X5-0.75X6-0.91X7(X1 = 24 h mean heart rate, X2 = minimum heart rate, X3 = coronary heart disease, X4 = paroxysmal atrial fibrillation, X5 = premature ventricular contraction, X6= sinus block, X7 = atrioventricular block). CONCLUSION: Our model based on the clinical features of patients with bradyarrhythmia. Should be useful aid for predicting pharmacotherapy response and could screen the optimal pharmacotherapy target.

The evaluation of toxicity related to polychlorinated dibenzo-p-dioxins and furans(PCDD/Fs)and dioxin-like polychlorinated biphenyls(DL-PCBs)is crucial for a comprehensive risk assessment in real-world exposure *** study employed a controlled feeding experiment to investigate the metabolic effects of dioxin-like compounds(DLCs)on laying hens via feed *** enriched with two concentrations(1.17 and 5.13 pg toxic equivalents(TEQ)/g dry weight(dw))were administered over 14 days,followed by 28 days of clean *** analyses of blood samples revealed significant metabolic variations between PCDD/Fs and DL-PCBs exposed groups and controls,reflecting the induced metabolic *** changes were observed in sphingosine,palmitoleic acid,linoleate,linolenic acid,taurocholic acid,indole acrylic acid,and dibutyl phthalate levels,implying possible connections between PCDD/Fs and DL-PCBs toxic effects and energy-neuronal imbalances,along with lipid accumulation and anomalous amino acid metabolism,impacting taurine ***,we identified three differential endogenous metabolites—L-tryptophan,indole-3-acetaldehyde,and indole acrylic acid—as potential ligands for the aryl hydrocarbon receptor(AhR),suggesting their role inmediating PCDD/Fs and DL-PCBs *** comprehensive investigation provides novel insights into the metabolic alterations induced by PCDD/Fs and DL-PCBs in laying hens,thereby enhancing our ability to assess risks associated with their exposure in human populations.

Genetic variants and mRNA expression levels of KLF4 and KLF5 with hypertension:A combination of case-control study and cohort study

Hypertension(HT)is a major risk factor for cardiovascular ***üppel-like factors(KLFs)are important transcription factors in *** have reported that KLF4 and KLF5 are correlated with several cardiovascular diseases,but population-based studies on associations between HT and KLF4 or KLF5 have rarely been ***,the current study investigated the associations of genetic variants and m RNA expression levels of KLF4 and KLF5 with HT,as well as the effects of antihypertensive drugs on the expression levels of these *** associations of one single-nucleotide polymorphism(SNP)in KLF4 and three SNPs in KLF5with HT were analyzed using a combination of case-control and cohort *** study populations were selected from a community-based cohort in four regions of Jiangsu *** risks of HT were estimated through logistic and Cox regression *** addition,m RNA expression levels of KLF4 and KLF5 were detected in 246 controls and 385 HT cases selected from the aforementioned *** the HT cases,263were not taking antihypertensive drugs[AHD(-)]and 122 were taking antihypertensive drugs[AHD(+)].In the case-control study,SNP rs9573096(C>T)in KLF5 was significantly associated with an increased risk of HT in the additive model(adjusted odds ratio[OR],1.106;95%confidence interval[CI],1.009 to 1.212).In the cohort study of the normotensive population,rs9573096 in KLF5 was also significantly associated with an increased risk of HT in the additive model(adjusted hazards ratio[HR],1.199;95%CI,1.070 to 1.344).KLF4 and KLF5m RNA expression levels were significantly higher in the AHD(-)group than in the control group(P<0.05),but lower in the AHD(+)group than in the AHD(-)group(P<0.05).The current study demonstrated the associations of KLF4 and KLF5 genetic variants with hypertension,as well as the association of the indicative variations in m RNA expression levels of KLF4 and KLF5 with the risk of hypertension and antihypertensive
