The objective of the paper is to provide a general view for automatic cup to disc ratio(cdr)assessment in fundus *** for the cause of blindness,glaucoma ranks as the second in ocular *** loss caused by glaucoma cannot be reversed,but the loss may be avoided if screened in the early stage of ***,early screening of glaucoma is very requisite to preserve vision and maintain quality of *** nerve head(ONH)assessment is a useful and practical technique among current glaucoma screening *** cdr as one of the clinical indicators for ONH assessment,has been well-used by clinicians and professionals for the analysis and diagnosis of *** key for automatic calculation of vertical cdr in fundus images is the segmentation of optic cup(OC)and optic disc(OD).We take a brief description of methodologies about the OC and disc optic segmentation and comprehensively presented these methods as two aspects:hand-craft feature and deep learning *** window regression,super-pixel level,image reconstruction,super-pixel level low-rank representation(LRR),deep learning methodologies for segmentation of OD and OC have been *** is hoped that this paper can provide guidance and bring inspiration to other *** mentioned method has its advantages and *** method should be selected or explored according to the actual *** automatic glaucoma screening,cdr is just the reflection for a small part of the disc,while utilizing comprehensive factors or multimodal images is the promising future direction to furthermore enhance the performance.

The antibody repertoire of Bos taurus is characterized by a subset of variable heavy(VH)chain regions with ultralong third complementarity determining regions(cdr3)which,compared to other species,can provide a potent response to challenging antigens like HIV *** unusual cdr3 can range to over seventy highly diverse amino acids in length and form uniqueβ-ribbon‘stalk’and disulfide bonded‘knob’structures,far from the typical antigen binding *** genetic components and processes for forming these unusual cattle antibody VH cdr3 are not well *** we analyze sequences of Bos taurus antibody VH domains and find that the subset with ultralong cdr3 exclusively uses a single variable gene,IGHV1-7(VHBUL)rearranged to the longest diversity gene,*** eight nucleotide duplication at the 3′end of IGHV1-7 encodes a longer V-region producing an extended Fβ-strand that contributes to the stalk in a rearranged cdr3.A low amino acid variability was observed in cdr1 and cdr2,suggesting that antigen binding for this subset most likely only depends on the *** a novel,potentially AID mediated,deletional diversification mechanism of the *** VH ultralong cdr3 knob was discovered,in which interior codons of the IGHD8-2 region are removed while maintaining integral structural components of the knob and descending strand of the stalk in *** deletions serve to further diversify cysteine positions,and thus disulfide bonded ***,both germline and somatic genetic factors and processes appear to be involved in diversification of this structurally unusual cattle VH ultralong cdr3 repertoire.

TO THE EDITOR:Characterization of the tumor-infiltrating B cell immunoglobulin(Ig)repertoire is critical to understanding B cell immunity in tumors and developing monoclonal antibody ***,the generation of specific antibodies for cancer therapy1,2 is a major endeavor,involving a lengthy process of antigen identification,immunization,hybridoma production and,in most cases,antibody *** a radical departure from the conventional approach,we hereby describe a rapid and potentially en masse identification of cancer-specific antibodies directly from human cancer tissues by de novo assembly from transcriptome and genome *** integrated computational framework was developed and successfully tested for antibody discovery by mining 1945 solid tumor RNA-sequencing-based samples for abundant Ig cdr3 sequences among the TCGA database of glioblastoma multiforme(GBM),lower grade glioma(LGG),lung adenocarcinoma(LUAD),lung squamous carcinoma(LUSC),pancreatic adenocarcinoma(PAAD),and skin cutaneous melanoma(SKCM).

【目的】筛选能表达鸡传染性支气管炎病毒(IBV)单链抗体(scFv)的原核阳性克隆,并分析scFv抗原结合位点的氨基酸变化。【方法】采用RT-PCR,从IBV疫苗免疫的鸡脾脏RNA中扩增出抗体编码基因的重链可变区(VH)和轻链可变区(VL)基因。利用重组链延伸反应(SOE-PCR),将linker与VH和VL基因相连构建鸡scFv基因,并将其克隆到原核表达载体pOPE101-XP中,构建重组质粒并转入大肠杆菌表达,间接ELISA筛选原核表达的抗IBVscFv的阳性克隆,对其测序后进行Clustal.W多序列比较,分析scFv骨架区(FR)和互补决定区(cdr)氨基酸序列差异。【结果】筛选到表达IBVscFv的阳性克隆ZL.10和ZL.80;将它们的氨基酸序列与鸡胚系Gemline进行多序列比较,结果显示,ZL.10、ZL.80在cdr中易变异氨基酸占各区氨基酸的比率均在40%以上,其中VH的cdr3达90%,VL的cdr3达60%。此外,ZL.10、ZL.80在VH的cdr3区分别增加5个和7个氨基酸。FR氨基酸变异率较少,且大多均在12%以下,而FR4未发生突变。【结论】通过体外筛选原核表达产物的方法,获得了表达IBVscFv的阳性克隆ZL.10和ZL.80,其VH、VL氨基酸序列中的变异主要发生在各自的cdr,二者VH的cdr3氨基酸序列差异很大,提示这2个scFv阳性克隆针对的是IBV不同的抗原位点。
