Gasdermins(GSDMs)are proteins cleaved by caspase(casP)to trigger *** teleosts,pyroptosis is mediated by gasdermin E(GSDME).The Pufferfish,Takifugu rubripes,possesses two GSDME orthologs:named TrGSDMEa and *** is cleaved by casP3/7 to liberate the N-terminal(NT)domain that can trigger pyroptosis in mammalian ***,the biological function of TrGSDMEa in pufferfish is unknown,and TrGSDMEb is poorly *** found that TrGSDMEb was cleaved by casP1/3/6/7/8,but the resulting NT domain,despite its similarity to TrGSDMEa-NT domain in sequence and structure,failed to induce *** and TrGSDMEb exhibited similar expression patterns in pufferfish under normal physiological conditions but were up-and downregulated,respectively,in expression during Vibrio harveyi and Edwardsiella tarda *** infection induced the activation of TrGSDMEa and casP3/7 in pufferfish cells,resulting in pyroptosis accompanied with IL-1βproduction and *** of TrGSDMEa-mediated pyroptosis via TrcasP3/7 reduced the death of pufferfish cells and augmented bacterial dissemination in fish ***-oriented mutagenesis identified 16 conserved residues in teleost GSDMEa that were required for the pore formation or auto-inhibition of *** study illustrates the role of GSDMEa-mediated pyroptosis in teleost defense against bacterial pathogens and provides new insights into the structure-based function of vertebrate GSDME.

Long non-coding RNAs(lncRNAs)function as key modulators in mammalian immunity,particularly due to their involvement in lncRNA-mediated competitive endogenous RNA(ceRNA)*** their recognized significance in mammals,research on lncRNAs in lower vertebrates remains *** the present study,we characterized the first immune-related lncRNA(pol-lnc78)in the teleost Japanese flounder(Paralichthys olivaceus).Results indicated that pol-lnc78 acted as a ceRNA for pol-miR-n199-3p to target the sterile alpha and armadillo motif-containing protein(SARM),the fifth discovered member of the Toll/interleukin 1(IL-1)receptor(TIR)adaptor *** ceRNA network regulated the antibacterial responses of flounder via the Toll-like receptor(TLR)signaling ***,SARM acted as a negative regulator and exacerbated bacterial infection by inhibiting the expression of inflammatory cytokines IL-1βand tumor necrosis factor-α(TNF-α).Pol-miR-n199-3p reduced SARM expression by specifically interacting with the 3’untranslated region(UTR),thereby promoting SARM-dependent inflammatory cytokine expression and protecting the host against bacterial ***,pol-lnc78 sponged pol-miR-n199-3p to ameliorate the inhibition of SARM *** infection,the negative regulators pol-lnc78 and SARM were significantly down-regulated,while pol-miR-n199-3p was significantly up-regulated,thus favoring host antibacterial *** findings provide novel insights into the mechanisms underlying fish immunity and open new horizons to better understand ceRNA crosstalk in lower vertebrates.

An assessment of the CMIP6 performance in simulating Arctic sea ice volume flux via Fram Strait

Numerical models serve as an essential tool to investigate the causes and effects of Arctic sea ice *** the simulation capabilities of the most recent CMIP6 models in sea ice volume flux provides references for model applications and ***,reliable long-term simulation results of the ice volume fux contribute to a deeper understanding of the sea ice response to global climate *** this study,the sea ice volume flux through six Arctic gateways over the past four decades(1979-2014)were estimated in combination of satellite observations of sea ice concentration(SIC)and sea ice motion(SIM)as well as the Pan-Arctic Ice-Ocean Modeling and Assimilation System(PIOMAS)reanalysis sea ice thickness(SIT)*** simulation capability of 17 CMIP6 historical models for the volume flux through Fram Strait were quantitatively *** ice volume flux simulated from the ensemble mean of 17 CMIP6 models demonstrates better performance than that from the individual model,yet IPSL-CM6A-LR and EC-Earth3-Veg-LR outperform the ensemble mean in the annual volume flux,with Taylor scores of 0.86 and 0.50,***6 models display relatively robust capability in simulating the seasonal variations of volume *** them,CESM2-WACCM performs the best,with a correlation coefficient of 0.96 and a Taylor score of ***,NESM3 demonstrates the largest devi-ation from the observation/reanalysis data,with the lowest Taylor score of *** variability of sea ice volume flux is primarily influenced by SIM and SIT,followed by *** extreme large sea ice export through Fram Strait is linked to the occurrence of anomalously low air temperatures,which in turn promote increased SIC and SIT in the corresponding ***,the intensified activity of Arctic cyclones and Arctic dipole anomaly could boost the southward sea ice velocity through Fram Strait,which further enhance the sea ice outflow.

Meroplankton play a crucial role in both benthic and pelagic *** quantitative research on estimating the quantities of meroplankton groups is both underrepresented and *** investigate and evaluate the influence of varying mesh sizes(505 and 160μm)on the sampling efficiency of meroplankton,we conducted an examination using two commonly used plankton nets during the spring season in the Southern Yellow Sea(SYS).Our study revealed a total of 12 meroplankton groups,with 9 groups identified in the 505-μm mesh nets and 11 groups in the 160-μm mesh *** results demonstrated the superior collection efficiency of the 160-μm net compared to the 505-μm net across the majority of meroplankton ***,we focused on exploring the abundance,distribution patterns,and realized niches of meroplankton collected by the two mesh size nets,and observed that the distribution of meroplankton closely resembled the distribution of possible benthic adults in the *** analysis of the six dominant groups collected in the 160-μm mesh nets revealed that seawater temperature and salinity emerged as the key environmental factors driving variations in meroplankton abundance within the *** study also found that a smaller mesh size net does not necessarily capture meroplankton more comprehensively.A comprehensive understanding of the ecological characteristics of meroplankton requires the combination of two types of nets for *** research significantly advances our understanding of the quantification,abundance,and distribution of meroplankton,serving as a valuable contribution to the broader landscape of detailed quantitative meroplankton studies.

Eddies are major elements of ocean dynamics that affect ocean *** their effects on plankton distribution may help understand the dynamics of harmful phytoplankton *** studies on the effects of eddies in the northern Arabian Sea have primarily focused on the zooplankton community,and few have observed zooplankton dynamics during winter blooms of Noctiluca *** investigated zooplankton community structure and the related environmental variability during a *** bloom that was affected by an eddy in February *** sampling stations were deployed at eddy core and eddy edge distinguished in salinity,temperature,and *** show that *** bloomed at the eddy core with high-velocity currents induced by warm eddies that moved from eddy core to eddy *** a result,blooms significantly changed the zooplankton community ***-bloom stations had higher zooplankton diversity than bloom *** at non-bloom stations were dominated by either tunicates or copepods,such as Thalia democratica and Pleuromamma *** addition to the influence of *** blooms,the velocity of eddy currents was a crucial factor on the similarities in the zooplankton community composition between eddy edge and eddy ***,the lower abiotic factors in bloom area contribute to the structuring of the zooplankton community during *** blooms.

Comparative transcriptomic analysis of ovary and testis reveals sex-related genes in roughskin sculpin Trachidermus fasciatus

The mechanisms underlying sex determination and differentiation have long intrigued researchers in the fields of development and evolutionary *** roughskin sculpin(Trachidermus fasciatus Heckel),displaying sexual dimorphism,provides an ideal model for studying the ***,both genetic and genomic information concerning sex determination and differentiation,such as gonadal transcriptome data in roughskin sculpin,are ***,we present the first gonadal transcriptomes of roughskin sculpin and identify sex-related *** identified 8531 differentially expressed genes(DEGs),among them 4065 were upregulated in the ovary and 4466 upregulated in the *** sex-related gene ontology(GO)terms were enriched in ovary-biased genes,including“binding of sperm to zona pellucida”,“egg coat formation”,“positive regulation of acrosome reaction”,“cell division”,and“cell cycle”,while the GO terms such as“spermatogenesis”,“sperm axoneme assembly”,“cilium assembly”,“cilium movement”,and“cilium movement involved in cell motility”were enriched in testis-biased ***,six KEGG pathways were significantly enriched in the ovary,whereas only one was enriched in the *** these DEGs,40 sex-related genes were identified,which including 26 testis-biased genes(such as Dmrtb 1,Gsdf,Sox 9 b,Wnt 4 b,Tcp 11 l 2,and Efhb),and 14 ovary-biased genes(such as Cyp 19 a 1 a,Foxh 1,Foxr 1,Gdf 3,Hsd 17 b 12,and Igf 2 bp 3).This gonadal transcript dataset would broaden our understanding of sex determination and differentiation mechanisms in roughskin sculpin,expand the genomic database,support future studies on sex-related gene functions,and facilitate molecular biology research into roughskin sculpin.

Genome-wide SNP markers provided insights into the reproductive strategy and genetic diversity of the green tide causative species Ulva prolifera in China

Ulva prolifera is the causative species of the annually occurring large-scale green tides in China since *** specific biological features on reproductivity strategies,as well as intra-species genetic diversity,are still largely unknown,especially at the genome level,despite their importance in understanding the formation and outbreak of massive green *** the present study,the restriction site-associated DNA genotyping approach(2b-RAD)was adopted to identify the genome-wide single-nucleotide polymorphisms(SNPs)of 54 individual thalli including samples collected from Subei Shoal in 2019 and Qingdao coast from 2019 to *** genotype results revealed that most of the thalli in 2019 and 2020 were haploid gametophytes,while only half of the thalli were gametophytes in 2021,indicating flexibility in the reproductive strategies for the formation of the green tides among different years and the dominance of asexual and vegetative reproductive mode for the floating ***,population analysis was conducted,and it revealed a very low genetic diversity among samples from Subei Shoal and the Qingdao coast in the same year and a higher divergence among samples in different *** results showed the efficiency of 2b-RAD in the exploration of SNPs in *** and provided the first genome-wide scale evidence for the origin of the large-scale green tides on the Qingdao *** study improved our understanding of the reproductive strategy and genetic diversity of the green tide causative species and will help further reveal the biological causes of the green tide in China.
