Parkinson’s disease (PD) is one of the most common neurodegenerative diseases. The loss of dopaminergic (DAergic) neurons in the substantia nigra and the decrease of dopamine (DA) levels accelerate the process of PD. L-Ergothioneine (EGT) is a natural antioxidant derived from microorganisms， especially in edible mushrooms. EGT can penetrate blood-brain barrier and its levels are significantly decreased in the plasma of PD patients. Therefore， we speculated that EGT could ameliorate PD， and determined its effect on PD development by using 1-methyl-4-phenyl-1，2，3，6-tetrahydropyridine (MPTP)-induced PD mouse models and neurotoxin 1-methyl-4-phenylpyridinium (MPP+)-induced cell models. Our results show that EGT alleviated MPTP-induced behavioral dysfunction in mice. Mechanistically， we innovatively revealed that EGT is a key regulator of DJ-1. EGT restored DA levels by activating the DJ-1-Nuclear receptor-related factor 1(Nurr1) axis. Furthermore， it reduced reactive oxygen species (ROS) levels by regulating the DJ-1-Nuclear factor erythroid 2-related factor 2 (Nrf2) pathway， which inhibited oxidative stress-induced DAergic neuronal apoptosis. combined treatment with DJ-1-siRNA transfection revealed that blocking DJ-1 reversed EGT upregulated Nurr1 and Nrf2 expression in the nucleus， which significantly decreased the benefits of EGT. Taken together， our study suggests that EGT can ameliorate PD and be considered as a strategy for PD treatment.

Regional variations in acute coronary syndrome(AcS) management and outcomes have been an enormous public health issue. However, studies have yet to explore how to reduce the variations. The National chest Pain center Program(NcPcP) is the first nationwide, hospital-based, comprehensive, continuous quality improvement program for improving the quality of care in patients with AcS in china. We evaluated the association of NcPcP and regional variations in AcS healthcare using generalized linear mixed models and interaction analysis. Patients in the Western region had longer onset-to-first medical contact(FMc) time and time stay in non-percutaneous coronary intervention(PcI) hospitals, lower rates of PcI for ST-elevation myocardial infarction(STEMI) patients, and higher rates of medication *** in central regions had relatively lower in-hospital mortality and in-hospital heart failure *** in the door-to-balloon time(Dto B) and in-hospital mortality between Western and Eastern regions were less after accreditation(β =-8.82, 95% confidence interval(cI)-14.61 to-3.03;OR = 0.79, 95%cI 0.70 to 0.91). Similar results were found in differences in Dto B time, primary PcI rate for STEMI between central and Eastern regions. The differences in PcI for higher-risk non-ST-segment elevation acute coronary syndrome(NSTE-AcS) patients among different regions had been ***, the differences in medication use between Eastern and Western regions were higher after accreditation. Regional variations remained high in this large cohort of patients with AcS from hospitals participating in the NcPcP in china. More comprehensive interventions and hospital internal system optimizations are needed to further reduce regional variations in the management and outcomes of patients with AcS.

The cOVID-19 pandemic caused by SARS-coV-2 virus is an ongoing global health *** cases of cOVID-19 and the rare cases of cOVID-19 vaccine-induced-thrombotic-thrombocytopenia(VITT)are both associated with thrombosis and thrombocytopenia;however,the underlying mechanisms remain inadequately *** infection and vaccination utilize the spike protein receptor-binding domain(RBD)of *** found that intravenous injection of recombinant RBD caused significant platelet clearance in *** investigation revealed the RBD could bind platelets,cause platelet activation,and potentiate platelet aggregation,which was exacerbated in the Delta and Kappa *** RBD–platelet interaction was partially dependent on theβ3 integrin as binding was significantly reduced inβ3−/−***,RBD binding to human and mouse platelets was significantly reduced with relatedαIIbβ3 antagonists and mutation of the RGD(arginine-glycine-aspartate)integrin binding motif to RGE(arginine-glycine-glutamate).We developed anti-RBD polyclonal and several monoclonal antibodies(mAbs)and identified 4F2 and 4H12 for their potent dual inhibition of RBD-induced platelet activation,aggregation,and clearance in vivo,and SARS-coV-2 infection and replication in Vero E6 *** data show that the RBD can bind platelets partially thoughαIIbβ3 and induce platelet activation and clearance,which may contribute to thrombosis and thrombocytopenia observed in cOVID-19 and *** newly developed mAbs 4F2 and 4H12 have potential not only for diagnosis of SARS-coV-2 virus antigen but also importantly for therapy against cOVID-19.

Social determinants of health(SDOH)affect quality of *** investigated SDOH impacts on self-perceived resilience among people with adult congenital heart disease(AcHD).Secondary analysis of data from two com-plementary studies:a survey study conducted May 2021–June 2022 and a qualitative study conducted June 2020–August *** was assessed through cD-RISc10 score(range 0–40,higher scores reﬂect greater self-perceived resilience)and interview *** and SDOH(education,employment,living situa-tion,monetary stability,ﬁnancial dependency,area deprivation index)data were collected by healthcare record review and *** used linear regression with robust standard errors to analyze survey data and performed a thematic analysis of interview *** participants(N=127)mean age was 42±14 years;51%were female,87%*** was moderate(75%)or complex(25%);41%functional class c or ***(mean 30±7)varied by monetary stability:compared to people with difﬁculty paying bills,resilience was 15.0 points higher(95%cI:6.9–23.1,pcI:5.9–22.4,p=0.001)for those reporting just enough *** participants’(N=25)mean age was 32 years(range 22–44);52%were female,72%*** was moderate(56%)or complex(44%);76%functional class c or *** discussed factors affecting resilience aligned with each of the major SDOH,prominently,economic stability and healthcare access and *** stability may be important for supporting self-perceived resi-lience in *** knowledge can inform the development of resilience interventions for this population.
