The mucin 1(MUC1)gene emerged in mammals to afford protection of barrier epithelial tissues from the external ***1 encodes a transmembrane C-terminal(MUC1-C)subunit that is activated by loss of homeostasis and induces inflammatory,proliferative,and remodeling pathways associated with wound *** a consequence,chronic activation of MUC1-C promotes lineage plasticity,epigenetic reprogramming,and *** driving cancer progression,MUC1-C is imported into the nucleus,where it induces NF-κB inflammatory signaling and the epithelial-mesenchymal transition(EMT).MUC1-C represses gene expression by activating(i)DNA methyltransferase 1(DNMT1)and DNMT3b,(ii)Polycomb Repressive Complex 1(PRC1)and PRC2,and(iii)the nucleosome remodeling and deacetylase(NuRD)***1/2-mediated gene repression is counteracted by the SWI/SNF chromatin remodeling ***1-C activates the SWI/SNF BAF and PBAF complexes in cancer stem cell(CSC)models with the induction of genome-wide differentially accessible regions and expressed ***1-C regulates chromatin accessibility of enhancer-like signatures in association with the induction of the Yamanaka pluripotency factors and recruitment of JUN and BAF,which promote increases in histone activation marks and opening of *** and other findings described in this review have uncovered a pivotal role for MUC1-C in integrating lineage plasticity and epigenetic reprogramming,which are transient in wound repair and sustained in promoting CSC progression.

Global Memory Net (GMNet) is intended to be an effective gateway to the world cultural, historical, and heritage image collections from selected academic educational and research partners in the world. Much of these unique collections of great value to education and research are not currently accessible due to distance, form, and technical barriers. This project is to find new ways to enable users to access and exploit these significant research collections via global network. As GMNet is ending its first 5-year phase in October 2005, it has contributed substantially to the community building in digital library development by ac- commodating numerous collaborators and technical staff from various parts of the world to spend 3 to 5 months as a full-member of the GMNet team in boston. They have come from different parts of China—such as Sichuan, Hainan, Shanghai and Xi’an; Croatia; and Hanoi, Vietnam. In addition to contribute to the overall system development and enhancement of system function- alities, they have brought valuable sample image collections of their own institutions/countries, and actually developed prototype collections as a part of GMNet. This paper describes the exciting and productive experience of the first of this visiting research group in developing the GMNet’s Version 2.0 PHP-based system under Prof. Chen’s overall supervision. It also describes both the system’s technical level structure—user/Web-based application/data, and complex functionalities with multi-collection, multi-lingual, multi-modal searching capabilities; system management capabilities; as well as provisions for user uploads and retrieval for our own projects. This Version 2.0 system is built on the Linux/Apache/PHP/MySQL platform. What is described in this paper is an actual case which has formed a base for further new development by others in the research group. It demonstrates fully the value of the synergistic collaboration among global partners for universal digital library

Hypoxia is a major factor shaping the immune landscape,and several cancer models have been developed to emulate hypoxic ***,to date,they still have several limitations,such as the lack of reproducibility,inadequate biophysical cues,limited immune cell infiltration,and poor oxygen(O_(2))control,leading to non-pathophysiological tumor ***,it is essential to develop better cancer models that mimic key features of the tumor extracellular matrix and recreate tumor-associated hypoxia while allowing cell infiltration and cancer-immune cell ***,hypoxia-inducing cryogels(HICs)have been engineered using hyaluronic acid(HA)to fabricate three-dimensional microtissues and model a hypoxic tumor ***,tumor cell-laden HICs have been designed to deplete O_(2) locally and induce long-standing *** promoted changes in hypoxia-responsive gene expression and phenotype,a metabolic adaptation to anaerobic glycolysis,and chemotherapy ***,HIC-supported tumor models induced dendritic cell(DC)inhibition,revealing a phenotypic change in the plasmacytoid DC(pDC)subset and an impaired conventional DC(cDC)response in ***,our HIC-based melanoma model induced CD8^(+)T cell inhibition,a condition associated with the downregulation of pro-inflammatory cytokine secretion,increased expression of immunomodulatory factors,and decreased degranulation and cytotoxic capacity of T ***,these data suggest that HICs can be used as a tool to model solid-like tumor microenvironments and has great potential to deepen our understanding of cancer-immune cell relationship in low O_(2) conditions and may pave the way for developing more effective therapies.

Treatment of children with cervical spine disorders requiring fusion is a challenging endeavor for a variety of reasons. The size of the patients, the corresponding abnormal bony anatomy, the inherent ligamentous laxity of children, and the relative rarity of the disorders all play a part in difficulty of treatment. The benefits of modern posterior cervical instrumentation in children, defined as rigid screw–rod systems, have been shown to be many including: improved arthrodesis rates, diminished times in halo-vest immobilization, and improved reduction of deformities. The anatomy of children and the corresponding pathology seen frequently is at the upper cervical spine and craniocervical junction given the relatively large head size of children and the horizontal facets at these regions predisposing them to instability or deformity. Posterior screw fixation, while challenging, allows for a rigid base to allow for fusion in these upper cervical areas which are predisposed to pseudarthrosis with non-rigid fixation. A thorough understanding of the anatomy of the cervical spine, the morphology of the cervical spine, and the available screw options is paramount for placing posterior cervical screws in children. The purpose of this review is to discuss both the anatomical and clinical descriptions re-lated to posterior screw placement in the cervical spine in children.

During the last decades,the use of nanotechnology in med icine has effectively been translated to the design of drug delivery systems,nanostructured tissues,diagnostic platforms,and novel nanomaterials against several human diseases and infectious ***-enabled vaccines have been positioned as solutions to mitigate the pandemic outbreak caused by the novel pathogen severe acute respiratory syndrome coronavirus *** fast-track the development of vaccines,unprecedented industrial and academic collaborations emerged around the world,resulting in the clinical translation of effective vaccines in less than one *** this article,we provide an overview of the path to translation from the bench to the clinic of nanotechnology-enabled messenger ribonucleic acid vaccines and examine in detail the types of delivery systems used,their mechanisms of action,obtained results during each phase of their clinical development and their regulatory approval *** also analyze how nanotechnology is impacting global health and economy during the COVID-19 pandemic and beyond.

The Casimir Effect is a physical manifestation of quantum fluctuations of the electromagnetic *** two metal plates are placed close together,typically much less than a micron,the long wavelength modes between them are frozen out,giving rise to a net attractive force between the plates,scaling as d^(−4)(or d^(−3)for a spherical-planar geometry)even when they are not electrically *** this paper,we observe the Casimir Effect in ambient conditions using a modified capacitive micro-electromechanical system(MEMS)*** a feedback-assisted pick-and-place assembly process,we are able to attach various microstructures onto the post-release MEMS,converting it from an inertial force sensor to a direct force measurement platform with pN(piconewton)*** this system we are able to directly measure the Casimir force between a silver-coated microsphere and goldcoated silicon *** device is a step towards leveraging the Casimir Effect for cheap,sensitive,room temperature quantum metrology.
