Emerging evidence shows that dietary oligosaccharides are important prebiotics that can improve intestinal flora,while dietary polyphenols can act directly on intestinal ***,information about their synergistic effects on gut health is still *** this study,alginate oligosaccharide(AOS)and cyanidin-3-O-glucoside(C3G)were selected as a common marine plant oligosaccharide and terrestrial plant polyphenol,respectively,to study their effects on intestinal *** results show that,in comparison to their individual applications,the combination of AOS and C3G(mass ratio,3:1)displayed a stronger ability to up-regulate the expression of tight junction proteins,while enhanced intestinal epithelial barrier was also observed and levels of mucin-2 andβ-defensins were simultaneously increased in the intestinal ***,the secretion of immunoglobulin A and immune-related cytokines were approximately doubled by the AOS+C3G *** addition,the AOS+C3G mixture was found to be more conducive to the positive transformation of intestinal flora,which stimulated the growth of beneficial bacteria Akkermansia,Lachnospiraceae and Feacalibaculum while inhibiting the growth of harmful bacteria Helicobacter and *** data generated herein thus suggests that dietary oligosaccharides and dietary polyphenols may be more beneficial to intestinal health when applied in combination than their individual effects alone.

Antioxidative and hepatoprotective activities of a novel polysaccharide (LSAP) from Lepista sordida mycelia

A novel alkali-soluble polysaccharide from Lepista sordida(LSAP)mycelia with antioxidative and hepatoprotective activities was *** weight-average molecular weight and number-average molecular weight of LSAP were 1.442×10^(3) and 6.05×10^(2) kDa,*** was consisted of glucose(57.9%),xylose(31.8%),and small amounts of rhamnose,arabinose,galactose,glucuronic acid,and galacturonic acid(1.2%–3.1%).The FT-IR and 2D NMR confi rmed that LSAP was composed of Xylp,Araf,4-O-Me-α-D-GlcpA,(1→4)-linkedβ-D-Glcp,and(1→4)-α-D-GalA,andβ-glycosidic linkages between these sugar *** displayed notable effects on 1,1-dephenyl-2-picryhydrazyl(DPPH)radical scavenging,hydrogen peroxide scavenging,lipid peroxidation inhibitory ability,reducing power and Fe^(2+)chelating *** biological effects were further verifi ed by suppressing CCl_(4)-induced oxidative liver damage in mice at doses of 100 and 200 mg/*** of LSAP in mice prior to CCl_(4) signifi cantly prevented the CCl_(4)-induced elevation in serum alanine aminotransferase,aspartate aminotransferase,and hepatic *** treated with LSAP demonstrated to increase activities in superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in the *** also found out that LSAP prevented CCl_(4)-induced oxidative liver histological *** may exert hepatoprotective effects against CCl_(4)-induced damage through antioxidant mechanisms in model mice.

Dear Editor,Hepatoblastoma(HB)is a predominant hepatic neoplasm that develops in children from 0 to 4 years of age at the rate of 2.16 per 1,000,*** originates from abnormal differentiation of hepatocyte precursors(hepatoblasts)during embryogenesis(Sumazin et al.,2017).Approximately 20%of children with HB have metastasis in lung at diagnosis,which indicates poor prognosis(Angelico et al.,2019).While surgery in combination of chemotherapy and/or metastasectomy is the most popular therapy,relapse happens in a significant portion of HB patients(Zhang et al.,2021).Therefore,novel and less aggressive therapies targeting the pathogenesis of HB should be explored to prolong patients’s disease-free survival as well as to improve their quality of life.

Knee osteoarthritis is a chronic disease caused by the deterioration of the knee joint due to various factors such as aging,trauma,and obesity,and the nonrenewable nature of the injured cartilage makes the treatment of osteoarthritis ***,we present a three-dimensional(3D)printed porous multilayer scaffold based on cold-water fish skin gelatin for osteoarticular cartilage *** make the scaffold,cold-water fish skin gelatin was combined with sodium alginate to increase viscosity,printability,and mechanical strength,and the hybrid hydrogel was printed according to a pre-designed specific structure using 3D printing ***,the printed scaffolds underwent a double-crosslinking process to enhance their mechanical strength even *** scaffolds mimic the structure of the original cartilage network in a way that allows chondrocytes to adhere,proliferate,and communicate with each other,transport nutrients,and prevent further damage to the *** importantly,we found that cold-water fish gelatin scaffolds were nonimmunogenic,nontoxic,and *** also implanted the scaffold into defective rat cartilage for 12 weeks and achieved satisfactory repair results in this animal ***,cold-water fish skin gelatin scaffolds may have broad application potential in regenerative medicine.

Alternative therapies are necessary for the treatment of malaria due to emerging drug ***,many promising antimalarial compounds have poor water solubility and suffer from the lack of suitable delivery systems,which seriously limits their *** address this problem,we synthesized a series of azacarbazoles that were evaluated for antimalarial activity against D10(chloroquine-sensitive)and W2(chloroquine-resistant)strains of *** most active compound,9H-3-azacarbazole(3),was encapsulated in a novel o/w nanoemulsion consisting of ethyl esters of polyunsaturated fatty acids n-3 and n-6 obtained from flax oil as the oil phase,Smix(Tween 80 and Transcutol HP)and *** formulation was further analyzed using transmission electron microscopy,dynamic light scattering and in vitro and in vivo *** was shown that droplets of the 3-loaded nanosystem were spherical,with satisfactory stability,without cytotoxicity towards fibroblasts and intestinal cell lines at concentrations corresponding to twice the IC50 for ***,the nanoemulsion with this type of oil phase was internalized by Caco-2 ***,pharmacokinetics demonstrated rapid absorption of compound 3(tmax=5.0 min)after intragastric administration of 3-encapsulated nanoemulsion at a dose of 0.02 mg/kg in mice,with penetration of compound 3 to deep *** 3-encapsulated nanoemulsion was found to be 2.8 and 4.2 times more effective in inhibiting the D10 and W2 strains of the parasite,respectively,compared to non-encapsulated *** findings support a role for novel o/w nanoemulsions as delivery vehicles for antimalarial drugs.

Molecular mechanism of inbreeding effects based on RNA-Seq analysis of the adductor muscle of bay scallop(Argopecten irradians)

Understanding the effects of inbreeding is critical for the long-term viability of shellfish breeding and for understanding inbreeding depression(ID) of the offspring of selfed individuals and full-sib crosses. The bay scallop(Argopecten irradians) is a simultaneous hermaphroditic marine bivalve with distinct male and female gonads and is a species with external fertilization. It is a canonical model invertebrate for analysis of the genetics of ID and evolution of outcrossing mating system. ID in bivalves is well documented with regard to the phenotype and molecular markers aspects, but little is known on how inbreeding affects genome-wide gene ***, we compare the gene expression profiles of adductor muscle of A. irradians lines inbred to F=0.5 with those of its counterpart outbred lines which are originated from the same paternal individual. Inbred and outbred individuals have some differences in growth traits. The shell length and the total weight of inbred scallops are lower than the counterpart of outbred lines. We found that 1 175 genes were up-regulated while 1 390 were downregulated in inbred lines. Regarding components, differentially expressed genes(DEGs) were concentrated on sarcomeres, contractile filaments, and cell membranes. Among others, whereas regarding their function, the Gene Ontology(GO) was concentrated on transferase, phosphotransferase, peptidase, and others. The DEGsenriched metabolic pathways mainly belonged to the oxidative phosphorylation pathway and apoptosis *** used another two families to verify our bioinformatic results. The selfed and the outbred family originated from different ancestors, but the selfed family showed obvious ID. By using real-time quantitative reverse transcriptase polymerase chain reaction(qRT-PCR), we verified that DEGs with inbreeding are involved in the oxidative phosphorylation and cell apoptosis pathways, suggesting that inbreeding may affect energy metabolism and homeostasis.
