An indigenous and industrially important manganese peroxidase （MnP） isolated from solid state culture of Pleurotus ostreatus IBL-02 was 2.1-fold purified and immobilized using sol-gel matrix of trimethoxysilane and proplytetramethoxysilane. Lower KM, higher Vmax, hyper-activation, enhanced acidic and thermal stability up to 70 ℃, were the improved]enhanced catalytic features of the presently sol-gel immobilized MnP. A maximum of 100% decolorization was achieved within a short time period for the different real dyes containing textile industry effluents i.e. Nishat textile （NIT）, K＆N textile （KIT）, Sitara textile （SIT）, and Crescent textile （CRT）.

β-Glucosidases are important in the formation of floral tea aroma and the development ofresistance to pathogens and herbivores in tea plants.A novel β-glucosidase was purified 117-fold to homogeneity,with a yield of 1.26%.from tea leaves by chilled acetone and ammonium sulfate precipitation,ion exchangechromatography(CM-Sephadex C-50)and fast protein liquid chromatography(FPLC;Superdex 75,ResourceS).The enzyme was a monomeric protein with specific activity of 2.57 U/*** molecular mass of theenzyme was estimated to be about 41 kDa and 34 kDa by SDS-PAGE and FPLC gel filtration on Superdex200,*** enzyme showed optimum activity at 50℃ and was stable at temperatures lower than40℃.It was active between pH 4.0 and pH 7.0,with an optimum activity at pH 5.5,and was fairly stablefrom pH 4.5 to pH *** enzyme showed maximum activity towards pNPG,low activity towardspNP-Galacto,and no activity towards pNP-Xylo.

Coronatine Alleviates Water Deficiency Stress on Winter Wheat Seedlings

With the aim to determine whether coronatine （COR） alleviates drought stress on wheat, two winter wheat （Triticum aestivum L.） cultivars, ChangWu134 （drought-tolerant） and Shan253 （drought-sensitive） were studied under hydroponic conditions. Seedlings at the three-leaf stage were eultured in a Hoagland solution containing COR at 0.1 ~M for 24 h, and then exposed to 20% polyethylene glycol 6000 （PEG- 6000）. Under simulated drought （SD）, COR increased the dry weight of shoots and roots of the two cultivars significantly; the root/shoot ratio also increased by 30% for Shan253 and 40% for ChangWu134. Both cultivars treated with COR under SD （0.1COR-I-PEG） maintained significantly higher relative water content, photosynthesis, transpiration, intercellular concentration of CO2 and stomatal conductance in leaves than those not treated with PEG. Under drought, COR significantly decreased the relative conductivity and malondialdehyde production, and the loss of 1,1-diphenyl-2-picrylhydrazyl scavenging activity in leaves was significantly alleviated in COR-treated plants. The activity of peroxidase, catalase, glutathione reductase and ascorbate peroxidase were adversely affected by drought. Leaves of plants treated with COR under drought produced less abscisic acid （ABA） than those not treated. Thus, COR might alleviate drought effects on wheat by reducing active oxygen species production, activating antioxidant enzymes and changing the ABA level.

Two models for prediction of the activity and stability of site-directed mutagenesis on tumor necrosis factor-α are established. The models are based on straightforward structural considerations, which do not require the elaboration of site-directed mutagenesis on the protein core and the hydrophobic surface area by analyzing the properties of the mutated amino acid residues. The reliabilities of the models have been tested by analyzing the mutants of tumor necrosis factor-α (TNF-α) whose two leucine residues (L29, L157) were mutated. Based on these models, a TNF-α mutant with high activity was created by molecular design.

Antibacterial peptides have received increasing attention as a new pharmaceutical substance. But the molecular mechanism of lysis is still poorly understood. CMIV gene and mutant CMIV gene in GST fusion system were expressed. After cleaving with different cleavage reagents, the peptide with an excess of N-terminus and with an un-amidated C-terminus stopped the activity while the peptide with an excess Asn at the C-terminus had the activity level the same as natural CMIV. The results showed that the terminal structure of cecropin CMIV played an important role in its biological activity.

A cDNA coding mutated cecropin CMIV from Bombyx mori was synthesized according to its amino acid sequense using E .coli biased codons .The gene was cloned into the fusion expression vector pEZZ318 and was expressed in E .coli *** fusion protein produced was purified by affinity chromatography to yield 26 mg/L fusion product .The anti-bacterial activities of recombinant cecropin CMIV were recovered after cleavage by chemical method.

Nucleotide oligomerization domain 2(NOD2) is a major cytoplasmic sensor for pathogens and is critical for the clearance of cytosolic bacteria in ***, studies regarding NOD2, especially the initiated signaling pathways, are scarce in teleost species. In this study, we identified a NOD2 molecule(PaNOD2) from ayu(Plecoglossus altivelis).Bioinformatics analysis showed the structure of NOD2 to be highly conserved during vertebrate evolution. Dual-luciferase reporter assays examined the activation of NF-κB signaling and Western blotting analysis detected the phosphorylation of three MAP kinases(p-38, Erk1/2, and JNK1/2).Functional study revealed that, like its mammalian counterparts, PaNOD2 was the receptor of the bacterial cell wall component muramyl dipeptide(MDP), and the leucine-rich repeat motif was responsible for the recognition and binding of Pa NOD2 with the ligand. Overexpression of PaNOD2 activated the NF-κB signaling pathway, leading to the upregulation of inflammatory cytokines, including TNF-α and IL-1β in HEK293 T cells and ayu head kidney-derived monocytes/macrophages(MO/MΦ).Particularly, we found that PaNOD2 activated the MAPK signaling pathways, as indicated by the increased phosphorylation of p-38, Erk1/2, and JNK1/2, which have not been characterized in any teleost species previously. Our findings proved that the NOD2 molecule and initiated pathways are conserved between mammals and ayu. Therefore, ayu could be used as an animal model to investigate NOD2-based diseases and therapeutic applications.
