Noise-induced hearing loss(NIHL) is a highly prevalent form of sensorineural hearing damage that has significant negative effects on individuals of all ages and there are no effective drugs approved by the US Food and drug Administration. In this study, we unveil the potential of superparamagnetic iron oxide nanoparticle assembly(SPIOCA) to reshape the dysbiosis of gut microbiota for treating NIHL. This modulation inhibits intestinal inflammation and oxidative stress responses, protecting the integrity of the intestinal barrier. Consequently, it reduces the transportation of pathogens and inflammatory factors from the bloodstream to the cochlea. Additionally, gut microbiota-modulated SPIOCA-induced metabolic reprogramming in the gut–inner ear axis mainly depends on the regulation of the sphingolipid metabolic pathway, which further contributes to the restoration of hearing function. Our study confirms the role of the microbiota–gut–inner ear axis in NIHL and provides a novel alternative for the treatment of NIHL and other microbiota dysbiosis-related diseases.

Inhibitory effect of M3 receptor antagonist 4-DAMP on melanoma proliferation of A375 cell line

Objective: he effect of M3 receptor antagonist 4-DAMP (4-diphenylacetoxy-N-methylpiperidine-methiodide) on the proliferation of melanoma were explored, and the development prospect and significance of M3 receptor antagonist in the field of anti-tumor were discussed. Methods: The viability of tumor cells was detected by MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide) colorimetry. The optimal concentration of 4-DAMP was screened and the effect of the drug was confirmed by EdU (5-ethynyl-2'-deoxyuridine) fluorescence staining. Adult male BALB/c nude mice were divided into control group, 5-Fu (5-Fluorouracil, 5-Fluorouracil, 20 mg/kg/d) group and 4-DAMP (2 mg/kg/d) group to establishtumor-bearing model in vivo. The tumor volume curve was calculated and plotted. Mice were killed after 21 days of continuous administration. Strip the tumor, take pictures and meatured the weighs, and calculate the inhibition rate. [1]The spleen was weighed and the spleen index was culculated. H & E staining was used to observe the pathological changes of tumor sections. The gene expression of M3R、MIA (Melanoma inhibitory activity, MIA)、SP-1 (Transcription factor Sp1, SP-1) and Lnc-SPRY4-IT1 (SPRY4 Intronic Transcript 1,Lnc-SPRY4-IT1) in A375 cell line was detected by real-time quantitative PCR (quantitative reverse-transcription polymerase chain reaction, qRT-PCR). The protein level of ERK1/2 (extracellular signal-regulated kinase 1/2,ERK1/2) was detected by Western Blot. Results: 4-DAMP could significantly inhibit the activity of A375 cell line (P < 0.01), and EdU staining demonstrated that 4-DAMP inhibited the growth of A375 cell line. The results of animal experiments showed that compared with the control group, the volume and mass of tumor in 5-Fu group and 4-DAMP group were significantly reduced (P < 0.05) and H & E staining showed that the intercellular space became larger, connective tissue increased, and tumor growth was obstructed. 目的：我们的目标是从源于胚胎体（EBs）的小鼠胚胎干细胞（mESCs）中分离运动神经元样细胞前体（MNLCPs）,以用于发展针对运动神经元疾病的药物筛选试验和移植疗法。天然Shh蛋白（或Shh通路激动剂）和维甲酸诱导的胚胎体中,MNLCPs和未分化细胞的含量是不确定的。如果不把未分化的细胞从细胞培养中充分去除,其可能会干涉药物筛选试验或在移植后增生。我们开发了一种以密度梯度离心法为基础的富集MNLCPs的方法。方法：我们用Wichterle等人2008年改进的方法,将mESCs（HBG3：eGFP：HB9）扩大和分化。通过化学和酶学的无研磨处理,含有绿色荧光蛋白的MNLCPs和未分化细胞的胚胎体被小心轻轻地离解成单细胞。利用OptiprepTM8%~20%逐步梯度离心技术将MNLCPs回收。拥有绿色荧光蛋白的MNLCPs的含量由流式细胞仪检测。结果：我们的结果表明,在胚胎体形成前,mESCs在明胶包被的培养板上生长,其分化为MNLCPs的能力减少。比较mESCs在明胶,明胶与PMEFs,及PMEFs包被的培养板上的生长发现,mESCs在PMEFs包被的培养板上产生含绿色荧光蛋白的MNLCPs的得率为（54.1±11.0）%（x±s;n=12）,在明胶包被的培养板上的得率为（2.8±1.1）%（x±s;n=9）。用密度梯度离心法获得的含绿色荧光蛋白的MNLCPs的平均含量为（87.7±5.5）%（x±s;n=3）。结论：我们的数据表明,不使用细胞分选器,无研磨解离和密度梯度离心法也能用于富集具有高存活率的MNLCPs。有必要对MNLCPs在体外、体内和表型上进行进一步的生理学意义（如神经轴突的生长及形成神经肌肉接头的能力）上的鉴定。

With the interdisciplinary convergence of biology,medicine and materials science,both research and clinical translation of biomaterials are progressing at a rapid ***,there is still a huge gap between applied basic research on biomaterials and their translational products-medical devices,where two significantly different perspectives and mindsets often work independently and non-synergistically,which in turn significantly increases financial costs and research *** this gap is well-known and often criticized in the biopharmaceutical industry,it is gradually *** this article,we critically examine the developmental pipeline of biodegradable biomaterials and biomaterial-based medical device *** based on clinical needs,market analysis,and relevant regulations,some ideas are proposed to integrate the two different mindsets to guide applied basic research and translation of biomaterial-based products,from the material and technical *** repair substitutes are discussed here as an ***,this will lay a strong foundation for biomaterial research and clinical translation,while reducing the amount of extra research effort and funding required due to the dissonance between innovative basic research and commercialization pipeline.

Objective: To explore the antitumor and potential off-target effects of systemically delivered cholesterol-conjugatedlet-7a mimics(Chol-let-7a) and control mimics(Chol-miRCtrl) on hepatocellular carcinomain ***: The antitumor effects of two intravenous dosing regimens ofChol-let-7a on heptocellular carcinoma growth were compared using an orthotopic xenograft mouse model. Off-targets were analyzed with histopathological and ultrapathological features of heparenal tissue and cells in theChol-let-7a-, Chol-miRCtrl-, and saline-treated (blank) xenograft mice and normal control mice. Then,let-7a abundance in orthotopic tumors, corresponding paracancerous hepatic tissue, and normal liver tissue from healthy nude mice was examined by reverse transcription-polymerase chain reaction. The distribution ofChol-let-7a andChol-miRCtrl in vivo was examined by whole-animal imaging and frozen-sections observation. The experiments were approved by the Institutional Research Board of Peking Union Medical College ***: Continuous treatment withChol-let-7a resulted in tumors that were 35.86% and 40.02% the size of those in theChol-miRCtrl and blank xenograft group (P < 0.01 andP < 0.01, respectively), while intermittent dosing withChol-let-7a resulted in tumors that were 65.42% and 56.66% the size of those in theChol-miRCtrl and the blank control group, respectively (P < 0.05 andP < 0.05). In addition, some histopathological and ultrapathological features were only observed after treatment with the two cholesterol-conjugated molecules, however mild with intermittent dosingChol-let-7a treatment, such as diffuse sinusoidal dilation and edema, primarily around the centrolobular vein in heptic tissues;mild hypercellularity with dilated capillary lumens in the renal tissue;and some organelle abnormalities found in heptic and renal cells. Background and objectives:Coronavirus disease 2019(COVID-19)is a pandemic that has become a major source of morbidity and mortality worldwide,affecting the physical and mental health of individuals influencing *** the threat,it poses to maternal health in sub-Saharan Africa and Nigeria,there is little or no data on the impact it has on fertility,conception,gestation and *** compare the birth rate between pre-COVID and COVID times using selected months of the *** and methods:This was a secondary analysis of cross-sectional analytical study data from the birth registries of three tertiary hospitals,comparing two years[2019(Pre-COVID)]versus[2020(COVID era)]using three months of the year(October to December).The data relied upon was obtained from birth registries in three busy maternity clinics all within tertiary hospitals in South-East Nigeria and we aimed at discussing the potential impacts of COVID-19 on fertility in *** secondary outcome measures were;mode of delivery,booking status of the participants,maternal age and ***:There was a significant decrease in tertiary-hospital based birth rate by 92 births(P=0.0009;95%CI:-16.0519 to-4.1481)among mothers in all the three hospitals in 2020 during the COVID period(post lockdown months)of October to *** was a significant difference in the mode of delivery for mothers(P=0.0096)with a 95%confidence interval of 1.0664 to 1.5916,as more gave birth through vaginal delivery during the 2020 COVID-19 period than ***:Tertiary-hospital based birth rates were reduced during the *** multi-centre study extrapolated on possible factors that may have played a role in this decline in their birth rate,which includes but is not limited to;decreased access to hospital care due to the total lockdowns/curfews and worsening inflation and economic recession in the country.

左归复方对MKR转基因2型糖尿病鼠胰岛素表达与分泌的影响

目的观察左归复方(滋阴益气活血解毒组方)干预治疗MKR小鼠后糖代谢、血清胰岛素及胰岛B细胞胰岛素表达的变化。方法40只MKR小鼠经鉴定后,随机分为模型组,左归复方高、低剂量组,阳性对照组(文迪雅),每组10只,另设10只C57小鼠为野生鼠组... 详细信息

目的观察左归复方(滋阴益气活血解毒组方)干预治疗MKR小鼠后糖代谢、血清胰岛素及胰岛B细胞胰岛素表达的变化。方法40只MKR小鼠经鉴定后,随机分为模型组,左归复方高、低剂量组,阳性对照组(文迪雅),每组10只,另设10只C57小鼠为野生鼠组。各药物组分别以相应剂量连续给药30d,模型组和野生鼠组灌胃蒸馏水。于给药前、第15日及第30日分别作空腹血糖测定;末次给药后0.5h心脏取血,放射免疫法测血清胰岛素,免疫组织化学SABC法检测胰岛B细胞胰岛素表达。结果左归复方干预治疗后,其高剂量具有显著的降低MKR小鼠空腹血糖、改善高胰岛素血症的作用。MKR小鼠胰岛B细胞胰岛素表达显著高于C57野生型小鼠;左归复方高剂量组干预治疗后胰岛B细胞胰岛素表达显著下调;左归复方高剂量组下调胰岛B细胞胰岛素表达与对照药文迪雅作用相当。结论左归复方具有改善MKR小鼠糖代谢、减轻胰岛B细胞的工作负荷、减少胰岛素表达及分泌的作用。
