Detection of congenital cytomegalovirus in newborns using nucleic acid amplification techniques and its public health implications

Human cytomegalovirus(HCMV), a herpesvirus, is an important human pathogen that causes asymptomatic infections in healthy or immunocompetent individuals but can lead to severe and potentially life-threatening complications in immune-immature individuals such as neonates or immune-compromised patients such as organ-transplant recipients and HIV-positive *** HCMV infection represents a significant public health issue and poses substantial healthcare and economic burden to society. This virus causes the most common viral congenital infection worldwide, and is the leading non-genetic cause of sensorineural hearing loss in children in developed countries. Congenital HCMV infection is believed to fulfill the criteria of the American college of Medical Genetics to be considered as a condition targeted for a newborn screening program. This is because congenital HCMV infection can be identified during a time(within 2 days after birth) at which it would not ordinarily be detected clinically, and there are demonstrated benefits of early detection, timely intervention, and efficacious treatment of the condition. Recent progresses in developing polymerase chain reaction-based approaches to detect HCMV in samples obtained from newborns have generated much excitement in the field. In this review, we highlight the recent progress in diagnostic techniques that could potentially be used for the detection of HCMV infection in neonates and its direct implications in public health settings for diagnosing congenital HCMV infection.

The multiplexing capability of fluorescence microscopy is severely limited by the broad fluorescence spectral *** imaging offers potential solutions,yet typical approaches to disperse the local emission spectra notably impede the attain able *** we show that using a sin gle,fixed fluoresc ence emission detection band,through frame-synchronized fast seanning of the excitation wavelength from a white lamp via an acousto-optic tun able filter,up to six subcellular targets,labeled by common fluorophores of substantial spectral overlap,can be simultaneously imaged in live cells with low(〜1%)crosstalks and high temporal resolutions(down to~10 ms).The dem on strated capability to quantify the abundances of different fluorophores in the same sample through unmixing the excitation spectra next enables us to devise novel,quantitative imaging schemes for both bi-state and Forster resonance energy transfer fluorescent biosensors in live *** thus achieve high sensitivities and spatiotemporal resolutions in quantifying the mitochondrial matrix pH and intracellular macromolecular crowding,and further demonstrate,for the first time,the multiplexing of absolute pH imaging with three additional target organelles/proteins to elucidate the complex,Parkin-mediated mitophagy ***,excitation spectral microscopy provides exceptional opportunities for highly multiplexed fluorescence *** prospect of acquiring fast spectral images without the need for fluorescence dispersion or care for the spectral response of the detector offers tremendous potential.

Thermal Stability and Thermal Aging of Poly(vinyl chloride)/MgAl Layered Double Hydroxides Composites

MgAl-LDH（layered double hydroxides） were prepared with CO（NH2）2, NH4 Cl and NH3·H2O by the coprecipitation method, respectively. Corresponding composite membranes were prepared by the coating method. LDHs were characterized by WAXS, CO2-TPD and SEM. The morphology of the PVC/LDHs composite membranes were characterized by means of SEM. The thermal stability of the membranes was analyzed by air aging box and TGA-FTIR. The SEM results show that nano-particles can be compatible with poly（vinyl chloride）（PVC） matrix homogeneously by the stirring-ultrasound blend method with two steps. Furthermore, the air aging box results proved that MgAl-CO（NH2）2-LDH has the best effect on thermal stability of PVC. TGA-FTIR results show that MgAl-CO（NH2）2-LDH could adsorb more HCl that resulted from the degradation of PVC and improve the pyrolysis temperature of the first degradation stage by 15 K compared with PVC.

Application of Linear Mean-Square Estimation in Ocean Engineering

The attempt to obtain long-term observed data around some sea areas we concern is usually very hard or even impossible in practical offshore and ocean engineering situations. In this paper, by means of linear mean-square estimation method, a new way to extend short-term data to long-term ones is developed. The long-term data about concerning sea areas can be constructed via a series of long-term data obtained from neighbor oceanographic stations, through relevance analysis of different data series. It is effective to cover the insufficiency of time series prediction method＇s overdependence upon the length of data series, as well as the limitation of variable numbers adopted in multiple linear regression model. The storm surge data collected from three oceanographic stations located in Shandong Peninsula are taken as examples to analyze the number-selection effect of reference oceanographic stations（adjacent to the concerning sea area） and the correlation coefficients between sea sites which are selected for reference and for engineering projects construction respectively. By comparing the N-year return-period values which are calculated from observed raw data and processed data which are extended from finite data series by means of the linear mean-square estimation method, one can draw a conclusion that this method can give considerably good estimation in practical ocean engineering, in spite of different extreme value distributions about raw and processed data.

The poor gas selectivity problem has been a long-standing issue for miniaturized chemical-resistor gas *** electronic nose(e-nose)was proposed in the 1980s to tackle the selectivity issue,but it required top-down chemical functionalization processes to deposit multiple functional ***,we report a novel gas-sensing scheme using a single graphene field-effect transistor(GFET)and machine learning to realize gas selectivity under particular conditions by combining the unique properties of the GFET and e-nose *** of using multiple functional materials,the gas-sensing conductivity profiles of a GFET are recorded and decoupled into four distinctive physical properties and projected onto a feature space as 4D output vectors and classified to differentiated target gases by using machine-learning *** single-GFET approach coupled with trained pattern recognition algorithms was able to classify water,methanol,and ethanol vapors with high accuracy quantitatively when they were tested ***,the gas-sensing patterns of methanol were qualitatively distinguished from those of water vapor in a binary mixture condition,suggesting that the proposed scheme is capable of differentiating a gas from the realistic scenario of an ambient environment with background *** such,this work offers a new class of gas-sensing schemes using a single GFET without multiple functional materials toward miniaturized e-noses.

Precisely quantifying transition metal(TM) redox in bulk is a key to understand the fundamental of optimizing cathode materials in secondary batteries. At present, the commonly used methods to probe TM redox are hard X-ray absorption spectroscopy(hXAS) and soft X-ray absorption spectroscopy(sXAS).However, they are both facing challenges to precisely quantify the valence states of some transition metals such as Mn. In this paper, Mn-L iPFY(inverse partial fluorescence yield) spectra extracted from Mn-L m RIXS(mapping of resonant inelastic X-ray scattering) is adopted to quantify Mn valence states. Mn-L i PFY spectra has been considered as a bulk-sensitive, non-distorted probe of TM valence ***, the exact precision of this method is still unclear in quantifying practical battery ***, a series of LiMn_(2)O_(4) electrodes with different charge and discharge states are prepared. Based on their electrochemical capacity(generally considered to be very precise), the precision of Mn iPFY in quantifying bulk Mn valence state is confirmed, and the error range is unraveled. Mn-L mRIXS iPFY thus is identified as one of the best methods to quantify the bulk Mn valence state comparing with hXAS and sXAS.

Determination of water level design for an estuarine city

Based on the extreme value theory, self-affinity, and scale invariance, we studied the temporal and spatial relationship and the variation of water level and established a model of Gumbel-Pareto distribution for designed flood calculation. The model includes the previous extreme value models, the over-threshold data, and the fractal features shared by previous extreme value models. The model was simplified into a logarithmic normal distribution and a Pareto distribution for specific parameter values, and was used to calculate the designed flood values for the Shanghai Wusong Station in 100- and 1 000-year return periods. The calculated results show that the value of the designed flood height calculated in the Gumbel-Pareto distribution is between those in the Gumbel and Pearson-Ⅲ distributions. The designed flood values in the 100- and 1 000-year return periods of the model were 0.03% and 0.11% lower, respectively, than the Gumbel distribution and 0.06% and 1.54% higher, respectively, than the Pearson-Ⅲ distribution. Compared to the traditional model based solely on extreme probability, the Gumbel-Pareto distribution model could better describe the probabilistic characteristics of extreme marine elements and better use the data.

KT Boundary Chromites Determined to be Terrestrial:Cr Isotopic Evidence for Excavation and Ejection of Mafic/Ultramafic Rocks by the KT Boundary Impact

Evidence for a mantle and/or basaltic component in KT boundary distal ejecta is apparently inconsistent with ejection from Chicxulub Crater since it is located on;5km thick continental crust（De Paolo et al.,1983;Montanari et al.,1983;Hildebrand and Boynton,1988,1990）.Evidence for mafic/ultramafic target rocks was reinforced by discovery of chromites,some with shock planar deformation features（PDF）,in impact layer samples from sites in southern Colorado and eastern Wyoming（Bohor et al.,1990）.However,until now it was unclear whether the chromites originated with an impactor or with terrestrial target *** this end,high-precision 54Cr/52Cr isotope ratios were measured on KT boundary chromites along with known terrestrial *** find a terrestrial 54Cr/52Cr ratio in KT boundary chromites from impact layer samples collected at the above sites over the last several years（Fig.1）.Ejected terrestrial chromites suggest the impact sampled terrestrial mafic and/or ultramafic target rocks not known to exist in the Chicxulub target area.

Peridynamic modeling and simulation of thermo-mechanical de-icing process with modified ice failure criterion

De-icing technology has become an increasingly important subject in numerous applications in recent ***,the direct numerical modeling and simulation the physical process of thermomechanical deicing is *** work is focusing on developing a numerical model and tool to direct simulate the de-icing process in the framework of the coupled thermo-mechanical peridynamics ***,we adopted the fully coupled thermo-mechanical bond-based peridynamics(TM-BB-PD)method for modeling and simulation of *** the framework of TM-BB-PD,the ice constitutive model is established by considering the influence of the temperature difference between two material points,and a modified failure criteria is proposed,which takes into account temperature effect to predict the damage of quasi-brittle ice ***,thermal boundary condition is used to simulate the thermal load in the de-icing *** comparing with the experimental results and the previous reported finite element modeling,our numerical model shows good agreement with the previous *** on the numerical results,we find that the developed method can not only predict crack initiation and propagation in the ice,but also predict the temperature distribution and heat conduction during the de-icing ***,the influence of the temperature for the ice crack growth pattern is discussed *** conclusion,the coupled thermal-mechanical peridynamics formulation with modified failure criterion is capable of providing a modeling tool for engineering applications of de-icing technology.
