Effect of biochar application on rice,wheat,and corn seedlings in hydroponic culture

In recent years,biochar has attracted considerable attention for soil quality improvement and carbon sequestration due to its unique physicochemical properties.However,the mechanism by which biochar application negatively affects the growth of crop seedlings has not been fully investigated.In this study,a hydroponic experiment was conducted to evaluate the response of rice,wheat,and corn seedlings to biochar application(CK,0 g/L;BC1,0.5 g/L;and BC2,1.0 g/L).Compared with the CK treatment,the BC1 and BC2 treatments decreased the fresh shoot and root weights of rice and corn seedlings(P0.05).For the contents of nutrient elements in seedlings,both BC1 and BC2 treatments hindered the roots from absorbing Fe and Cu and increased the uptake of Ca and Mn.Compared with the CK treatment,the translocation factor(TF)values of Ca,Mn,and Zn were significantly decreased especially in rice seedlings(35.3%-36.8%,68.7%-76.5%,and 29.8%-22.0%,respectively)under the BC1 and BC2 treatments,while only Mn was significantly decreased in wheat and corn seedlings(P<0.05).Transmission electron microscope(TEM)analysis of root cross-sections showed that nano-sized biochar particles(10~23 nm)were found in the root cells under BC2 treatment conditions.Our findings reveal that a large amount of biochar application can reduce nutrient absorption and translocation,and hinder rice,wheat,and corn seedlings,particularly rice seedling,in hydroponic system.

Halogenated benzoquinones(HBQs)could cause bladder cancer,but there were few related studies on the generation and control.In this study,the impact of different precursors,pH,bromide concentration,and algae-derived organic matters on the formation of HBQs and the removal efficiency by activated carbon were investigated.It was found that the chlorination of bisphenol A produced the most 2,6-dichloro-1,4-benzoquinone(2,6-DCBQ),reaching 14.86μg/L at 1 hr,followed by tyrosine,2-chlorophenol,P-hydroxybenzoic acid,trichlorophenol,and N-methylaniline.The production of 2,6-DCBQ increased first and then decreased from 0 to 36 hr(chlorination doses 0-20 mg/L),indicating that HBQs were unstable in water.Trihalomethanes(THMs)were detected during chlorination,and the concentration increased with prolongation of reaction time.2,6-DCBQ production decreased and 2,6-dibromo-1,4-benzoquinone(2,6-DBBQ)production increased with increment bromide concentration and the bromide promoted the formation of tribromomethane.The production of 2,6-DCBQ decreased with increase of pH,and the maximum production was 141.38μg/L at pH of 5.Microcystis aeruginosa,Chlorella algae cells,and intracellular organic matters(IOM)could be chlorinated as potential precursors for HBQs.The most amount of 2,6-DCBQ was generated from algae cells of Microcystis aeruginosa,followed by Chlorella algae cells,Microcystis aeruginosa IOM,and Chlorella IOM.This study compared the removal efficiency of HBQs by granular activated carbon(GAC)and columnar activated carbon(CAC)under different carbon doses and initial concentrations of HBQs.It was found that the removal efficiency by GAC(80.1%)was higher than that by CAC(51.8%),indicating that GAC has better control for HBQs.

Passive sampling technology has good application prospects for monitoring trace pollutants in aquatic environments.Further research on the sampling mechanism of this technology is essential to improve the measurement accuracy and extend the application scope of this approach.In this study,adsorption and permeation experiments were performed to investigate the sorption and mass transfer properties of five chiral pharmaceuticals at the enantiomeric level on polyethersulfone(PES)and polytetrafluoroethylene(PTFE)membranes used in a polar organic chemical integrative sampler.Batch adsorption experiments showed that the PES membrane had an adsorption phenomenon for most selected pollutants and an insignificant sorption behavior was observed for all selected pharmaceuticals on the PTFE membrane except for R(S)-fluoxetine.The diffusion coefficients of selected pharmaceuticals onto the PTFE membrane were approximately one order of magnitude higher than those onto the PES membrane.The permeation experiment indicated that under different hydraulic conditions,the change of the relative pollutant concentration through the PTFE membrane for the composite pollutant system was more obvious than that for the single pollutant system,and mass transfer hysteresis exists for both contaminant systems through PES membranes.Using the first-order equation or 3-component model to estimate the overall mass transfer coefficients,the results showed that the overall mass transfer coefficient values of pollutants in the composite pollutant system onto both membranes were higher than those in the single pollutant system.This parameter was mainly influenced by the synergistic effects of the multi-analyte interaction and diminished water boundary layers during the mass transfer process.

Succession of bacteria communities during production and application of dairy bedding by membrane-covered aerobic fermentation

The cost of dairy manure treatment and bedding material purchase increases the operating cost of the dairy farm.Membrane-covered aerobic fermentation system has been widely used for dealing with dairy manure and recycling the final product as bedding material.However,the microbial safety in each processing step is still uncertain.To better understand the bacterial community dynamics during the whole bedding conversion process,a full-chain and large-scale experiment including 16-day membrane-covered aerobic fermentation and 11-day bedding material application was conducted.The results showed that the pile temperatures in the fermentation stage rapidly increased to 80°C and maintained>50°C for more than 11 days and the use of fermentation product as bedding material provided cows with a stable and comfortable bedding environment.The Chao1 and Shannon index decreased at the end of the fermentation stage and remained stable in the application stage,indicating that membrane-covered aerobic fermentation effectively killed some pathogenic bacteria and guaranteed both the maturity and stability of the final product.The dominant bacteria in the fermentation stage were Acinetobacter,Thermus,and Rhodothermus at genus level.Seven common potential pathogens of mastitis(Staphylococcus,Enterococcus,Serratia,Pseudomonas,Corynebacterium,Mycobacterium,and Bacillus)were found at the end of fermentation stage but the relative abundance was low(0.0025%-0.2727%).The dominant bacteria in the application stage mainly included Acinetobacter,Pseudomonas,and Flavobacterium at the genus level.The relative abundance of Pseudomonas increased in the application stage,which was a reminder to the dairy farm to pay attention to the disinfection and timely replacement of bedding material to prevent the occurrence of dairy mastitis.The results of this study contributed deep understanding of the microorganism-driven bedding conversion process and provide practical guidance and cautions for the bedding materials application.

Bitter melon fruit is susceptible to yellowing,softening,and rotting under room-temperature storage conditions,resulting in reduced commercial value.Nitric oxide(NO)is an important signaling molecule and plays a crucial role in regulating the fruit postharvest quality.In this study,we investigated the effects of NO treatment on changes in sensory and firmness of bitter melon fruit during postharvest storage.Moreover,transcriptomic,metabolomic,and proteomic analyses were performed to elucidate the regulatory mechanisms through which No treatment delays the ripening and senescence of bitter melon fruit.Our results show that differentially expressed genes(DEGs)were involved in fruit texture(CSLE,β-Gal,and PME),plant hormone signal transduction(ACS,JAR4,and AUX28),and fruit flavor and aroma(SUS2,LOX,and GDH2).In addition,proteins differentially abundant were associated with fruit texture(PLY,PME,and PGA)and plant hormone signal transduction(PBL15,JAR1,and PYL9).Moreover,No significantly increased the abundance of key enzymes involved in the phenylpropanoid biosynthetic pathway,thus enhancing the disease resistance and alleviating softening of bitter melon fruit.Finally,differential metabolites mainly included phenolic acids,terpenoids,and flavonoids.These results provide a theoretical basis for further studies on the physiological changes associated with postharvest ripening and senescence of bitter melon fruit.

Glucosinolates are important phytochemicals in Brassicaceae.We investigated the effect of CaCl_(2)-HCl electrolyzed water(CHEW)on glucosinolates biosynthesis in broccoli sprouts.The results showed that CHEW treatment significantly decreased reactive oxygen species(ROS)and malondialdeh yde(MDA)contents in broccoli sprouts.On the the 8^(th)day,compared to tap water treatment,the the total glucosinolate content of broccoli sprouts with CHEW treatment increased by 10.6%and calcium content was dramatically enhanced from 14.4 mg/g DW to 22.7 mg/g DW.Comparative transcriptome and metabolome analyses revealed that CHEW treatment activated ROS and calcium signaling transduction pathways in broccoli sprouts and they interacted through MAPK cascades.Besides,CHEW treatment not only promoted the biosynthesis of amino acids,but also enhanced the expression of structural genes in glucosinolate synthesis through transcription factors(MYBs,bHLHs,WRKYs,etc.).The results of this study provided new insights into the regulatory network of glucosinolates biosynthesis in broccoli sprouts under CHEW treatment.

The circadian clock coordinates rhythms in numerous physiological processes to maintain organismal homeostasis. Since the suprachiasmatic nucleus(SCN) is widely accepted as the circadian pacemaker, it is critical to understand the neural mechanisms by which rhythmic information is transferred from the SCN to peripheral clocks. Here, we present the first comprehensive map of SCN efferent connections and suggest a molecular logic underlying these projections. The SCN projects broadly to most major regions of the brain, rather than solely to the hypothalamus and thalamus. The efferent projections from different subtypes of SCN neurons vary in distance and intensity, and blocking synaptic transmission of these circuits affects circadian rhythms in locomotion and feeding to different extents. We also developed a barcoding system to integrate retrograde tracing with in-situ sequencing, allowing us to link circuit anatomy and spatial patterns of gene expression. Analyses using this system revealed that brain regions functioning downstream of the SCN receive input from multiple neuropeptidergic cell types within the SCN, and that individual SCN neurons generally project to a single downstream brain region.This map of SCN efferent connections provides a critical foundation for future investigations into the neural circuits underlying SCNmediated rhythms in physiology. Further, our new barcoded tracing method provides a tool for revealing the molecular logic of neuronal circuits within heterogeneous brain regions.

Melatonin is a conserved pleiotropic molecule in animals and plants.Melatonin is involved in many development processes and stress responses;thus,exploring its function in plants,particularly in horticultural plants,has become a rapidly developing field.Many studies have revealed that phytomelatonin acts as a plant biostimulant and increase its tolerance to various abiotic stressors,including extreme temperature,drought,osmotic disturbance,heavy metals,and ultraviolet(UV).Melatonin appears to have roles in the scavenging of reactive oxygen species(ROS)and other free radicals,affecting the primary and secondary metabolism of plants,regulating the transcripts of stress-related enzymes and transcription factors,and crosstalk with other hormones under different environmental conditions.This pleiotropy makes phytomelatonin an attractive regulator to improve resistance to abiotic stress in plants.The recent discovery of the potential phytomelatonin receptor CAND2/PMTR1 and the proposition of putative models related to the phytomelatonin signaling pathways makes phytomelatonin a new plant hormone.Based on relevant studies from our laboratory,this review summarizes the phytomelatonin biosynthetic and metabolic pathways in plants and the latest research progress on phytomelatonin in abiotic stress of horticultural plants.This study will provide a reference for elucidating the regulatory mechanism of phytomelatonin affecting the resistance to abiotic stress in plants.
