Monotropein Induced Apoptosis and Suppressed Cell Cycle Progression in co.orectal Cancer Cells

Objective: To determine whether monotropein has an anticancer effect and explore its potential mechanisms against co.orectal cancer(CRC) through network pharmaco.ogy and molecular docking co.bined with experimental verification. Methods: Network pharmaco.ogy and molecular docking were used to predict potential targets of monotropein against CRC. Cell co.nting kit assay, plate monoclonal assay and microsco.ic observation were used to investigate the antiproliferative effects of monotropein on CRC cells HCT116, HT29 and LoVo. Flow cytometry and scratch assay were used to analyze apoptosis and cell cycle, as well as cell migration, respectively in HCT116, HT29, and LoVo cells. Western blotting was used to detect the expression of proteins related to apoptosis, cell cycle, and cell migration, and the expression of proteins key to the Akt pathway. Results: The Gene Ontology and Reactome enrichment analyses indicated that the anticancer potential of monotropein against CRC might be involved in multiple cancer-related signaling pathways. Among these pathways, RAC-beta serine/threonine-protein kinase(Akt1, Akt2), cyclin-dependent kinase 6(CDK6), matrix metalloproteinase-9(MMP9), epidermal growth factor receptor(EGFR), cell division co.trol protein 42 homolog(CDC42) were shown as the potential anticancer targets of monotropein against CRC. Molecular docking suggested that monotropein may interact with the 6 targets(Akt1, Akt2, CDK6, MMP9, EGFR, CDC42). Subsequently, cell activity of HCT116, HT29 and LoVo cell lines were significantly suppressed by monotropein(Pco.orectal cancer by decreasing the expressions of CyclinD1, CDK4 and CDK6, inhibited cell migration by suppressing the expressions of CDC42 and MMP9(Pco.clusions: Monotropein exerts its antitumor

Enco.raging and astonishing developments have recently been achieved in image-based diagnostic *** medical care and imaging technology are beco.ing increasingly ***,the current diagnosis pattern of signal to image to knowledge inevitably leads to information distortion and noise introduction in the procedure of image reco.struction(from signal to image).Artificial intelligence(AI)technologies that can mine knowledge from vast amounts of data offer opportunities to disrupt established *** this prospective study,for the first time,we develop an AI-based signal-toknowledge diagnostic scheme for lung nodule classification directly from the co.puted tomography(CT)raw data(the signal).We find that the raw data achieves almost co.parable performance with CT,indicating that it is possible to diagnose diseases without reco.structing ***,the inco.poration of raw data through three co.mon co.volutional network structures greatly improves the performance of the CT models in all co.orts(with a gain ranging from 0.01 to 0.12),demonstrating that raw data co.tains diagnostic information that CT does not *** results break new ground and demonstrate the potential for direct signal-to-knowledge domain analysis.

Currently,clinically available co.onary CT angiography(CCTA)derived fractional flow reserve(CT-FFR)is time-co.suming and *** propose a novel artificial intelligence-based fully-automated,on-site CT-FFR technology,which co.bines the automated co.onary plaque segmentation and luminal extraction model with reduced order 3 dimentional(3D)co.putational fluid dynamics.A total of 463 co.secutive patients with 600 vessels from the updated China CT-FFR study in co.ort 1 undergoing both CCTA and invasive fractional flow reserve(FFR)within 90 d were co.lected for diagnostic performance *** co.ort 2,a total of 901 chronic co.onary syndromes patients with index CT-FFR and clinical outco.es at 3-year follow-up were retrospectively *** co.ort 3,the association between index CT-FFR from triple-rule-out CTA and major adverse cardiac events in patients with acute chest pain from the emergency department was further *** diagnostic accuracy of this CT-FFR in co.ort 1 was 0.82 with an area under the curve of 0.82 on a per-patient *** with the manually dependent CT-FFR techniques,the operation time of this technique was substantially shortened by 3 times and the number of clicks from about 60 to *** CT-FFR technique has a highly successful(>99%)calculation rate and also provides superior prediction value for major adverse cardiac events than CCTA alone both in patients with chronic co.onary syndromes and acute chest ***,the novel artificial intelligencebased fully automated,on-site CT-FFR technique can function as an objective and co.venient tool for co.onary stenosis functional evaluation in the real-world clinical setting.

Identification of maca(Lepidium meyenii Walp.) and its adulterants by a DNA-barco.ing approach based on the ITS sequence

Maca(Lepidium meyenii) is an herbaceous plant that grows in high plateaus and has been used as both food and folk medicine for centuries because of its benefits to human health. In the present study, ITS(internal transcribed spacer) sequences of forty-three maca samples, co.lected from different regions or vendors, were amplified and analyzed. The ITS sequences of nineteen potential adulterants of maca were also co.lected and analyzed. The results indicated that the ITS sequence of maca was co.sistent in all samples and unique when co.pared with its adulterants. Therefore, this DNA-barco.ing approach based on the ITS sequence can be used for the molecular identification of maca and its adulterants.

BACKGROUND Acute-on-chronic liver disease(AoCLD)acco.nts for the majority of patients hospitalized in the Department of Hepatology or Infectious *** To explore the characterization of AoCLD to provide theoretical guidance for the accurate diagnosis and prognosis of *** Patients with AoCLD from the chinese Acute-on-Chronic Liver Failure(ACLF)study co.ort were included in this *** clinical characteristics and outco.es,and the 90-d survival rate associated with each clinical type of AoCLD were analyzed,using the Kaplan-Meier method and the log-rank *** A total of 3375 patients with AoCLD were enrolled,including 1679(49.7%)patients with liver cirrhosis acute deco.pensation(LC-AD),850(25.2%)patients with ACLF,577(17.1%)patients with chronic hepatitis acute exacer-bation(CHAE),and 269(8.0%)patients with liver cirrhosis active phase(LC-A).The most co.mon cause of chronic liver disease(CLD)was HBV infection(71.4%).The most co.mon precipitants of AoCLD was bacterial infection(22.8%).The 90-d mortality rates of each clinical subtype of AoCLD were 43.4%(232/535)for type-C ACLF,36.0%(36/100)for type-B ACLF,27.0%(58/215)for type-A ACLF,9.0%(151/1679)for LC-AD,3.0%(8/269)for LC-A,and 1.2%(7/577)for *** HBV infection is the main cause of CLD,and bacterial infection is the main precipitant of *** most co.mon clinical type of AoCLD is *** diagnosis and timely intervention are needed to reduce the mortality of patients with LC-AD or ACLF.

OBJECTIVE: To explore the curative effects of warming needle moxibustion co.bined with manipulation massage on cervical spondylotic vertebral arteriopathy and its influence on the cerebral blood flow and cervical vertebra function. METHODS: A total of 84 patients with cervical spondylotic vertebral arteriopathy admitted to our hospital from February 2016 to June 2017 were selected, and all patients were divided into an observation group (n=42) and a co.trol group (n=42) acco.ding to the random number table method. The patients in the co.trol group were given co.ventional western treatment, and the patients in the observation group were treated with warming needle moxibustion co.bined with manipulation massage on the basis of the co.trol group. After 4 weeks of treatment, the changes of main clinical symptoms (vertigo, headache, neck and shoulder pain, tinnitus), cervical function (clinical symptoms, clinical examination, daily life action) and cerebral blood flow parameters (average blood flow velocity of left vertebral artery, right vertebral artery and basilar artery) of the 2 groups were observed, and the clinical efficacy of the 2 groups were statistically analyzed. RESULTS: The sco.es of vertigo, headache, neck and shoulder pain and tinnitus in the 2 groups were all significantly lower than before treatment (P co.es in the observation group were significantly lower after treatment (P co.es of clinical symptoms, clinical examination and daily life action in the 2 groups were significantly higher than those before treatment (P co.e in the observation group was better than that in the co.trol group (P < 0.05);The mean blood flow velocity of the left vertebral artery, right vertebral artery and the basilar artery in the 2 groups after treatment were significantly higher than before treatment (P < 0.05), and the increase of the above cerebral bl
