Analysis of hydroxylated polybrominated diphenyl ethers in plant samples using ultra performance liquid chromatography-mass spectrometry

A method was developed for the analysis of hydroxylated brominated diphenyl ethers (OH-PBDEs) in plant samples using an ultra performance liquid chromatography-triple quadrupole mass spectrometer (UPLC-ESI-MS/MS) in negative mode. Plant samples were extracted and cleaned up through florisil column, resolved on a 100 mm C18 column with linear gradient elution and detected by mass spectrometry in multiple reaction monitoring (MRM) mode. The method provided good recoveries rang- ing from 68.2% to 94.6%, relative standard deviation (RSD) in the range of 3.2% - 9.1%, and limits of quantification (LOQ) defined as the signal-to-noise ratio of 10 of 0.3-2.1 ng/g. It allowed a fast separation and sensitive quantification of the isomers and homologues of seven OH-PBDE congeners 2′-OH-BDE-3, 3′-OH-BDE-7, 4′-OH-BDE-17, 3′-OH-BDE-28, 3-OH-BDE-47, 5-OH-BDE-47 and 6-OH-BDE-47. The method was successfully applied to identify and quantify the formation of hydroxylat- ed metabolites in alfalfa exposed to BDE-209. Five OH-PBDEs were detected in plant tissues, and more congeners were found in roots than in shoots. To our knowledge, this work represents the first attempt to validate UPLC-MS/MS method to quantify OH-PBDEs in plant samples without derivatization.

The application of near-infrared spectra micro-image in the imaging analysis of biology samples

In this research,suitable imaging methods were used for acquiring single compound images ofbiology samples of chicken pectorales tissue section,tobacco dry leaf,fresh leaf and plantglandular hair,*** adverse effects caused by the high water content and thethermal effect of near infrared(NIR)light were effectively solved during the experiment procedures and the data *** algorithm was applied to the NIR micro-image of chickenpectorales *** the loading vector of PC3 with the NIR spectrum of dry albumen,the information of PC3 was confimmed to be provided mainly by protein,i.e.,the 3rd score imagerepresents the distribution trend of protein *** algorithm was applied to the NIR micro-image of tobacco dry *** information of PC2 was confimed to be provided by carbohydrateindluding starch *** to the 2nd score image of tobacco dry leaf,the comparedcorelation image with the reference spectrum of starch had the same distribution trend as the 2nd score *** comparative correla tion images with the reference spectra of protein,glucose,fructose and the total plant alkaloid were acquired to confirm the distribution trend ofthese compounds in tobacco dry leaf *** correlation images of fresh leafwith the reference spectra of protein,starch,fructose,ghucose and water were acquired to confim the distribution trend of these compounds in fresh *** imaging of plant glandularhair was acquired to show the tubular structure clearly.

Global sensitivity analysis of wheat grain yield and quality and the related process variables from the DSSAT-CERES model based on the extended Fourier Amplitude Sensitivity Test method

A crop growth model,integrating genotype,environment,and management factor,was developed to serve as an analytical tool to study the influence of these factors on crop growth,production,and agricultural planning.A major challenge of model application is the optimization and calibration of a considerable number of *** analysis(SA) has become an effective method to identify the importance of various *** this study,the extended Fourier Amplitude Sensitivity Test(EFAST) approach was used to evaluate the sensitivity of the DSSAT-CERES model output responses of interest to 39 crop genotype parameters and six soil *** outputs for the SA included grain yield and quality(take grain protein content(GPC) as an indicator) at maturity stage,as well as leaf area index,aboveground biomass,and aboveground nitrogen accumulation at the critical process *** key results showed that:(1) the influence of parameter bounds on the sensitivity results was slight and less than the impacts from the significance of the parameters themselves;(2) the sensitivity parameters of grain yield and GPC were different,and the sensitivity of the interactions between parameters to GPC was greater than those between the parameters to grain yield;and(3) the sensitivity analyses of some process variables,including leaf area index,aboveground biomass,and aboveground nitrogen accumulation,should be performed ***,some parameters,which improve the model’s structure and the accuracy of the process simulation,should not be ignored when maturity output as an objective variable is studied.

The detection of apple juice is very important for its quality and safety *** developing of fast,non-invasive detection system is beneficial to the improvement of producing efficiency and *** this study,the homemade near infrared（NIR） spectrometer was applied to detect the soluble solid contents（SSC） and conductivity of apple juice,and the performance of homemade NIR analyzer and the feasibility of detecting conductivity by NIR spectra were *** varieties of apples（totally 120 samples） were collected,including Fuji apple,American green apple,and Chinese green *** fresh pellucid apple juice was made and their spectra were measured by a homemade charge coupled device（CCD） NIR *** result showed that homemade CCD NIR spectrometer could accurately predict the SSC of apple juice in the wavelength range of780-1100 nm combined with partial-least square *** correlation coefficient of calibration was r = 0.96,the standard deviation of prediction was SEP = 0.45°Brix,the relative of SEP was SEP%= 3.75%.The results indicated that the homemade NIR analyzer applied in this study had good performance,and the predictive ability of model was as good as that obtained by FT-NIR *** spectra and the conductivity of apple juice had some correlation（r = 0.77）,but the prediction accuracy was *** prediction result was SEP = 0.30,SEP%= 15.14%.From the analysis of regression coefficients of PLS model,it can be concluded that the conductivity and NIR spectra may have some indirect relation through the ionization of organic acid.

[Background]Water is an important limiting factor in crop irrigation *** the demand for domestic and industrial water has increased with the growing population,there is increasing competition in agriculture water resource *** resources for agriculture are becoming increasingly scarce in irrigation agriculture regions,for example,the North China Plain(NCP).Water productivity(WP) or water use efficiency(WUE,which is advocated as an indicator to analyze water use) to aid in water *** crop production,WUE has been generally defined as the amount of marketable product(e.g.,kilograms of grain,kg) in relation to the amount of input needed to produce that output(cubic meters of water,m3).In the NCP,50%of the total water resource is applied for field irrigation management in winter wheat,which is the key factor causing a decrease in groundwater *** current problems of serious water shortages and higher demand for crop yield can be solved by improving WUE in this region.[Materials and methods]The objectives of this study were:(i) to develop and test a model for assessing WUE in winter wheat,where the estimation methods,the empirical statistics spectral indices method and data assimilation method are tested;and(ii) to compare performance of the proposed models using the empirical statistics spectral indices method and the data assimilation *** variables and concurrent biomass,yield and WUE parameters of samples were acquired at the Xiaotangshan experimental site in beijing,China,during the 2008/2009,2009/2010,2010/2011 and 2011/2012 winter wheat growing *** data assimilation scheme for remote sensing and AquaCrop model is *** sensitivity analysis method is used to select crop parameters for AquaCrop model,the database lookup table(DLT)was established according to the related files,regression analysis methods were constructed between the remote sensing data and the measured biomass to use remote sensing data for
