BACKGROUND Mitochondrial genes are involved in tumor metabolism in ovarian cancer(OC)and affect immune cell infiltration and treatment *** To predict prognosis and immunotherapy response in patients diagnosed with OC using mitochondrial genes and neural *** Prognosis,immunotherapy efficacy,and next-generation sequencing data of patients with OC were downloaded from The Cancer Genome Atlas and Gene Expression *** genes were sourced from the MitoCarta3.0 *** discovery cohort for model construction was created from 70% of the patients,whereas the remaining 30% constituted the validation *** the expression of mitochondrial genes as the predictor variable and based on neural network algorithm,the overall survival time and immunotherapy efficacy(complete or partial response)of patients were *** In total,375 patients with OC were included to construct the prognostic model,and 26 patients were included to construct the immune efficacy *** average area under the receiver operating characteristic curve of the prognostic model was 0.7268[95% confidence interval(CI):0.7258-0.7278]in the discovery cohort and 0.6475(95%CI:0.6466-0.6484)in the validation *** average area under the receiver operating characteristic curve of the immunotherapy efficacy model was 0.9444(95%CI:0.8333-1.0000)in the discovery cohort and 0.9167(95%CI:0.6667-1.0000)in the validation *** The application of mitochondrial genes and neural networks has the potential to predict prognosis and immunotherapy response in patients with OC,providing valuable insights into personalized treatment strategies.

Recent developments in natural products for white adipose tissue browning

Excess accumulation of white adipose tissue(WAT) causes obesity which is an imbalance between energy intake and energy expenditure. Obesity is a serious concern because it has been the leading causes of death worldwide, including diabetes, stroke,heart disease and cancer. Therefore, uncovering the mechanism of obesity and discovering anti-obesity drugs are crucial to prevent obesity and its complications. Browning, inducing white adipose tissue to brown or beige(brite) fat which is brown-like fat emerging in WAT, becomes an appealing therapeutic strategy for obesity and metabolic disorders. Due to lack of efficacy or intolerable side-effects, the clinical trials that promote brown adipose tissue(BAT) thermogenesis and browning of WAT have not been successful in humans. Obviously, more specific means still need to be developed to activate browning of white adipose tissue. In this review, we summarized seven kinds of natural products(alkaloids, flavonoids, terpenoids, long chain fatty acids, phenolic acids, else and extract) promoting white adipose tissue browning which can ameliorate the metabolic disorders, including obesity, dislipidemia, insulin resistance and diabetes. Since natural products are important drug sources and the browning property plays a significant role in not only obesity treatment but also in type 2 diabetes(T2 DM) improvement, natural products of inducing browning may be an irreplaceable drug discovery orientation for obesity, diabetes and even other metabolic disorders.

Objective:To analyze the factors related to pregnancy of endometriosis and whether Chinese herbal medicines(CHMs)can improve pregnancy outcomes in patients with endometriosis in long-term ***:This multicenter cohort study retrospectively analyzed the clinical data of endometriosis patients with fertility needs from January 2019 to November 2019.A total of 252 patients with endometriosis from 5 level-ⅢGrade A hospitals in beijing were included in this *** and multivariate logistic regression analysis were performed for the relevant *** propensity score matching(PSM)function of SPSS software was used to match the CHMs group with the non-CHMs *** pregnancy rate and live birth rate were ***:The results of univariate analysis showed that age,disease course,presence of infertility,presence of adenomyosis,time after surgery or use of gonadotropin-releasing hormone agonist(Gn RH-a),use of CHMs and follow-up time were influencing factors of pregnancy in endometriosis patients(P6 months were independent risk factors(OR=0.445,0.348,0.140,respectively,P<0.05),time after surgery or use of Gn RH-a 6 months and use of CHMs were independent protective factors(OR=3.839,3.842,respectively,P<0.05).After PSM,99 pairs of two groups were matched *** pregnancy rate of the CHMs group was higher than that of the non-CHMs group[55.56%(55/99)vs.36.36%(36/99),P<0.05].The live birth rate of the CHMs group was higher than that of the non-CHMs group[49.49%(49/99)vs.35.35%(35/99),P<0.05].Conclusion:CHMs can effectively improve clinical pregnancy rate and live birth rate of patients with endometriosis in the chronic disease management.

Neurological outcomes of untreated brainstem cavernous malformations in a prospective observational cohort and literature review

Background Haemorrhages of brainstem cavernous malformations(CMs)can lead to neurological deficits,the natural history of which is *** study aimed to evaluate the neurological outcomes of untreated brainstem CMs and to identify the adverse factors associated with worsened *** From 2009 to 2015,698 patients(321 women)with brainstem CMs were entered into the prospective cohort after excluding patients lost to follow-up(n=43).All patients were registered,clinical data were collected and scheduled follow-up was *** After a median follow-up of 60.9 months,prospective haemorrhages occurred in 167 patients(23.9%).The mean modified Rankin Scale scores at enrolment and at censoring time were 1.6 and *** status was improved,unchanged and worsened in 334(47.9%),293(42.0%)and 71(10.2%)patients,respectively;233(33.4%)recovered to normal *** crossing the axial midpoint(relative risk(RR)2.325,p=0.003)and developmental venous anomaly(DVA)(RR 1.776,p=0.036)were independently significantly related to worsened *** percentage of worsened outcomes was 5.3%(18 of 337)in low-risk patients(neither DVA nor crossing the axial point)and increased to 26.0%(13 of 50)in high-risk patients(with both DVA and crossing the axial point).The percentage of worsened outcomes significantly increased as the number of prospective haemorrhages increased(from 1.5%(8 of 531,if 0 prospective ictus)to 37.5%(48 of 128,if 1 ictus)and 38.5%(15 of 39,if>1 ictus)).Conclusions The neurological outcomes of untreated brainstem CMs were improved/unchanged in majority of patients(89.8%)with a fatality rate of 1.7%in our cohort,which seemed to be *** features significantly predicted worsened *** results provide evidence for clinical consultation and individualised *** referral bias of our cohort was underlined.

Effect of direct current pulse stimulating acupoints of JiaJi (T10-L3) and Ciliao (BL 32) with Han's Acupoint Nerve Stimulator on labour pain in women: a randomized controlled clinical study

OBJECTIVE: To assess the clinical effect and safety of direct current(DC) pulse produced by Han's Acupoint Nerve Stimulator in reduction(HANS) of labor ***: Totally 120 participants were enrolled in this clinical trial, and were randomly divided into4 groups including: HANS group, patient controlled intravenous analgesia(PCIA) group, patient-controlled epidural analgesia(PCEA) group and control group. The HANS group was treated by stimulating the acupoints of Jia Ji(T10-L3) and Ciliao(BL 32)with DC pulse of 100 Hz and 15-30 m A produced by a portable battery-powered Han's Acupoint Nerve Stimulator for 30 min. The PCIA group was intravenously infused Ondansetron(8 mg) for 5 min,then tramadol injection(1.5 mg/kg) was slowly dripped by using Baxter APⅡ electronic pump with50 m L tramadol(0.70%) + ondansetron(8 mg),background infusion 2 m L/h, PCA dose of 2 m L,lockout interval of 10 min. In PCEA group, women received intrathecal injection ropivacaine(3 mg) in L2-3, and epidural catheter was connected to Baxter APⅡ electronic pump, with 100 m L Ropivacaine(0.1%) and Sufentanil(50 ug), background infusion5 m L, Patient controlled analgesia(PCA) dose of 5m L, lockout interval of 10 min. The control group was not received analgesia. The visual analogue scale(VAS), stage and manner of labor, Apgar score of newborn, neonatal weights, oxytocin dosage,postpartum hemorrhage and side effects were monitored in all ***: The vital signs were all stable in the four analgesic groups. After analgesia, there was statistical difference in VAS score between HANS group and control group, between PCEA group and the control group, between PCIA group and control group. Up-regulation of mitochondrial antioxidation signals in ovarian cancer cells with aggressive biologic behavior

Objective： Recently, a high frequency of mutations in mitochondrial DNA （mtDNA） has been detected in ovarian cancer. To explore the alterations of proteins in mitochondria in ovarian cancer, a pair of human ovarian carcinoma cell lines （SKOV3/***1） with different metastatic potentials was examined. Methods： Cancer cells *** were derived from the ascitic tumor cells of nude mice bearing a tumor of ovarian cancer cells SKOV3. *** exhibited a higher degree of migration potential than its paired cell line SKOV3. The proteins in the mi- tochondria of these two cells were isolated and separated by 2-D gel electrophoresis. The differently expressed proteins were extracted and identified using matrix assisted laser desorption ionisation/time-of-flight/time-of-flight （MALDITOF/TOF）, and finally a selected protein candidate was further investigated by immunohistochemistry （IHC） method in nude mice bearing tumor tissues of these two cells. Results： A total of 35 spots with different expressions were identified between the two cells using 2D-polyacrylamide gel electrophoresis （PAGE） approach. Among them, 17 spots were detected only in either SKOV3 or *** cells. Eighteen spots expressed different levels, with as much as a three-fold difference between the two cells. Twenty spots were analyzed using MALDI-TOF/TOF, and 11 of them were identified successfully; four were known to be located in mitochondria, including superoxide dismutase 2 （SOD2）, fumarate hydratase （FH）, mitochondrial ribosomal protein L38 （MRPL38）, and mRNA turnover 4 homolog （MRTO4）. An increased staining of SOD2 was observed in *** over that of SKOV3 in IHC analysis. Conclusions： Our results indicate that the enhanced antioxidation and metabolic potentials of ovarian cancer cells might contribute to their aggressive and metastatic behaviors. The underlying mechanism warrants further study.
