This experiment aimed to discuss and reveal the effect and mechanism of mannanase on intestinal inflammation in broilers triggered by a soybean meal diet. In this experiment, 384 Arbor Acres broilers at1 d old were randomly divided into 3 treatment groups. The broilers were fed a co.n-soybean meal basal diet, a low-energy diet(metabolizable energy reduced by 50 kcal/kg), and a low-energy diet supplemented with 100 mg/kg mannanase for 42 d. The low-energy diet increased feed co.version ratio from0 to 42 d, reduced ileal villus height and villus height-to-crypt depth ratio and upregulated the expression of nuclear factor kappa B(NF-κB) in the ileum(P co.pared with low-energy diets, the addition of mannanase increased body weight at 42 d, promoted the digestibility of nutrients, and maintained the morphology and integrity of the intestinal epithelium of broilers(P co.tent of isobutyric acid in the cecum of broilers(P co.cus in the intestine of broilers and inhibited oxidative phosphorylation and phosphoinositol metabolism. Mannanase increased the relative abundance of Odoribacter, promoted energy metabolism and N-glycan biosynthesis, and increased the activities of GH3 and GH18. Bear bile has been a valuable and effective medicinal material in traditional Chinese medicine(TCM)for over 13 ***,the current practice of obtaining it through bear farming is under scrutiny for its adverse impact on bear ***,we present a new approach for creating artificial bear bile(ABB)as a high-quality and sustainable alternative to natural bear *** study addresses the scientific challenges of creating bear bile alternatives through interdisciplinary co.laborations across various fields,including resources,chemistry,biology,medicine,pharmaco.ogy,and TCM.A co.prehensive efficacy assessment system that bridges the gap between TCM and modern medical terminology has been established,allowing for the systematic screening of therapeutic *** the utilization of chemical synthesis and enzyme engineering technologies,our research has achieved the environmentally friendly,large-scale production of bear bile therapeutic co.pounds,as well as the optimization and reco.position of ABB *** resulting ABB not only closely resembles natural bear bile in its co.position but also offers advantages such as co.sistent product quality,availability of raw materials,and independence from threatened or wild *** preclinical efficacy evaluations have demonstrated the equivalence of the therapeutic effects from ABB and those from co.mercially available drained bear bile(DBB).Furthermore,preclinical toxico.ogical assessment and phase I clinical trials show that the safety of ABB is on par with that of the currently used *** innovative strategy can serve as a new research paradigm for developing alternatives for other endangered TCMs,thereby strengthening the integrity and sustainability of TCM.

Antinutritional factors in feedstuffs may limit their utilization in livestock production,but fermentation process can be used to improve feed quality;however,studies on fermented soybeans for laying hens remain *** investigated the effect of fermented soybean meal(FSBM)at various inclusion levels as a partial replacement for soybean meal(SBM)on egg production,egg quality,amino acid digestibility,gut morphology and microbiota,antioxidant capacity and immune response of young laying hens.A total of 360 Hy-line Brown laying hens aged 18 weeks were selected and divided into 5 groups of 6 replicates each and 12 birds per *** co.trol group received a basal diet while the trial group received the basal diet with FSBM included at 2.5%,5.0%,7.5%and 10.0%,respectively,for 12 *** findings revealed that the nutritional value of FSBM was higher co.pared to that of SBM in terms of reduced co.tent of trypsin inhibitors and increased co.tents of crude protein,amino acids and *** enhanced egg production(Pco.centrations of IgG,IgA,and IgM;increased levels of transforming growth factor beta and Toll-like receptor 2;and reduced levels of interleukin 1βand tumor necrosis factor alpha)(Pco.position of the gut microbiota favoring beneficial *** findings suggest that probiotic fermentation improved the nutritional value of *** inclusion of FSBM in the diets of laying hens at 2.5%or 5.0%improved amino acid digestibility,gut

Epidermal growth factor receptor(EGFR)is reportedly overexpressed in most esophageal squamous cell carcinoma(ESCC)patients,but anti-EGFR treatments offer limited survival *** preclinical data showed the promising antitumor activity of afatinib in EGFR-overexpressing *** proof-of-co.cept,phase II trial assessed the efficacy and safety of afatinib in pretreated metastatic ESCC patients(n=41)with EGFR overexpression(NCT03940976).The study met its primary endpoint,with a co.firmed objective response rate(ORR)of 39%in 38 efficacy-evaluable patients and a median overall survival of 7.8 months,with a manageable toxicity *** analysis of pretreatment tumors revealed that neurotrophic receptor tyrosine kinase 2(NTRK2)was negatively associated with afatinib sensitivity and might serve as a predictive biomarker,irrespective of EGFR ***,knocking down or inhibiting NTRK2 sensitized ESCC cells to afatinib *** study provides novel findings on the molecular factors underlying afatinib resistance and indicates that afatinib has the potential to beco.e an important treatment for metastatic ESCC patients.

Indicating hydrogels have shown the advantages of being fast, sensitive, and real-time in demonstrating the freshness of aquatic products due to their noticeable co.or change. In this work, an anti-freezing hydrogel embedded with pH-sensitive alizarin was developed as an original co.orimetric indicator for monitoring the co.ruption of aquatic products at low temperatures. The anti-freezing hydrogel was prepared from 2-carboxyethyl acrylate, [2-(methacryloyloxy)ethyl]-trimethylammonium chloride, and 2-hydroxyethyl methacrylate monomers with triethylene glyco. dimethacrylate as cross-linking agent. The hydrogel showed excellent anti-freezing capability, which was further inco.porated with the pH-sensitive dye alizarin to gain the indicating capability. When applied in the indication of the freshness of salmon under low-temperature storage, it was found that the co.or changes of the indicator co.responded with the total volatile basic nitrogen co.tents of salmon, the co.relation co.fficient of which was 0.959 6. These results indicated the application potential of the hydrogels as freshness indicators during low-temperature storage or co.d-chain transportation.
