Objective:To investigate a new noninvasive diagnostic model for nonalcoholic fatty liver disease(NAFLD) based on features of tongue ***:Healthy controls and volunteers confirmed to have NAFLD by liver ultrasound were recruited from China-Japan friendship hospital between September 2018 and May 2019,then the anthropometric indexes and sampled tongue images were *** tongue images were labeled by features,based on a brief protocol,without knowing any other clinical data,after a series of corrections and data *** algorithm was trained on images using labels and several anthropometric indexes for inputs,utilizing machine learning ***,a logistic regression algorithm and a decision tree model were constructed as 2 diagnostic models for ***:A total of 720 subjects were enrolled in this study,including 432 patients with NAFLD and 288 healthy *** them,482 were randomly allocated into the training set and 238 into the validation *** diagnostic model based on logistic regression exhibited excellent performance:in validation set,it achieved an accuracy of 86.98%,sensitivity of 91.43%,and specificity of 80.61%;with an area under the curve(AUC) of 0.93 [95% confidence interval(CI) 0.68–0.98].The decision tree model achieved an accuracy of 81.09%,sensitivity of 91.43%,and specificity of 66.33%;with an AUC of 0.89(95% CI 0.66–0.92) in validation ***:The features of tongue images were associated with *** the 2 diagnostic models,which would be convenient,noninvasive,lightweight,rapid,and inexpensive technical references for early screening,can accurately distinguish NAFLD and are worth further study.

Bisphosphonate-related osteonecrosis of jaw(BRONJ)is characterized by impaired osteogenic differentiation of orofacial bone marrow stromal cells(BMSCs).Corin has recently been demonstrated to act as a key regulator in bone development and orthopedic ***,the role of corin in BRONJ-related BMSCs dysfunction remains unclarified.A m6A epitranscriptomic microarray study from our group shows that the CORIN gene is significantly upregulated and m6A hypermethylated during orofacial BMSCs osteogenic *** knockdown inhibits BMSCs osteogenic differentiation,whereas corin overexpression or soluble corin(sCorin)exerts a promotion ***,corin expression is negatively regulated by bisphosphonates(BPs).Corin overexpression or sCorin reverses BPs-impaired BMSCs differentiation ***,we find altered expression of phos-ERK in corin knockdown/overexpression BMSCs and BMSCs under sCorin ***98059(a selective ERK inhibitor)blocks the corin-mediated promotion *** regard to the high methylation level of corin during osteogenic differentiation,we apply a non-selective m6A methylase inhibitor,Cycloleucine,which also blocks the corin-mediated promotion ***,we demonstrate that METTL7A modulates corin m6A modification and reverses BPs-impaired BMSCs function,indicating that METTL7A regulates corin expression and thus contributes to orofacial BMSCs differentiation *** conclude,our study reveals that corin reverses BPs-induced BMSCs dysfunction,and METTL7A-mediated corin m6A modification underlies corin promotion of osteogenic differentiation via the ERK *** hope this brings new insights into future clinical treatments for BRONJ.
