In August 2021,three students with diarrhea from the same school visited a local hospital in the S district of *** epidemic investigation showed that there were more students with diarrhea in the same school and they had one meal *** jejuni was isolated from both patients with diarrhea and asymptomatic food handlers;however,the latter also carried Campylobacter *** analysis showed that there was a campylobacteriosis outbreak among the students,and the asymptomatic food handler may have been the source of the *** inspection and surveillance for Campylobacter is needed for the food producing staff,particularly those cooking in the cafeteria in schools or other public food services.

Objective To investigate molecular characterization of streptococcus pyogenes isolates involved in an outbreak of scarlet fever in China in 2011. Methods Seventy-four 5treptococcol pyogenes involved in an outbreak of scarlet fever were isolated from pediatric patients in the areas with high incidence in China from May to August of 2011. Emm genotyping, pulsed-field gel electrophoresis （PFGE）, superantigen （SAg） genes and antimicrobial susceptibility profiling were analyzed for these isolates. Results A total of 4 different emm types were identified. Emm12 was the most prevalent type which contained four predominating PFGE patterns corresponding to four different virulence and superantigen profiles. Emm12（79.7%） and emml （14.9%） accounted for approximately 94% of all the isolates. The speA gene was all negative in emm12 isolates and positive in emml isolates. All strains were resistant to erythromycin, and 89.4% of them were resistant to erythromycin, tracycline, and clindamycin simultaneously. Conclusion Several highly diversified clones with a high macrolide resistance rate comprise a predominant proportion of circulating strains, though no new emm type was found in this outbreak. The data provide a baseline for further surveillance of scarlet fever, which may contribute to the explanation of the outbreak and development of a GAS vaccine in China.

Objective To construct a rapid and high-throughput assay for identifying recombinant bacteria based on mass spectrometry. Methods Matrix assisted laser desorption ionization time-of-flight mass spectrometry （MALDI-TOF MS） techniques were used to identify 12 recombinant proteins （10 of Yersinia pestis, 1 of Campylobacter jejuni and 1 of Helicobacter pylori）. A classification model for the various phase of recombinant bacteria was established, optimized and validated, using MALDI-TOF MS-CIinProTools system. The differences in the peptide mass spectra were analyzed by using Biotyper and FIexAnalysis softwares. Results Models of GA, SNN, and QC were established. After optimizing the parameters, the GA recognition model showed good classification capabilities： RC=100%, mean CVA=98.7% （the CVA was 96.4% in phase 1, 100% in phase 2, 98.4% in phase 3, and 100% in phase 4, respectively） and PPV=95}. This model can be used to classify the bacteria and their recombinant, which only requires 3.7x103 cells for analysis. The total time needed is only 10 min from protein extraction to reporting the result for one sample. Furthermore, this assay can automatically detect and test 96 samples concurrently. A total of 48 specific peaks （9, 16, 9, and 14 for the four stages, respectively） was found in the various phase of recombinant bacteria. Conclusion MALDI-TOF MS can be used as a fast, accurate, and high-throughput method to identify recombinant bacteria, which provide a new ideas not only for recombinant bacteria but also for the identification of mutant strains and bioterrorism pathogens.

Introduction:Human sapovirus(HuSaV)is an enteric virus responsible for sporadic cases and outbreaks of acute gastroenteritis(AGE)globally.A seven-year active surveillance study was conducted to investigate the molecular epidemiology of HuSaVs associated with AGE outbreaks in Chaoyang district of beijing Municipality,China from January 2015 to December ***:Fecal and anal swab samples were obtained from patients experiencing AGE *** were identified through reverse transcription polymerase chain reaction(RT-PCR),and partial viral protein 1(VP1)sequences(approximately 434 base pairs)were utilized for genotyping,single nucleotide polymorphism(SNP)analysis,and phylogenetic ***:HuSaVs were identified in 71 AGE outbreaks,demonstrating a detection rate of 10.5%,second only to *** primary demographic affected by HuSaV were children under the age of 5 in kindergarten *** rates tended to peak during two distinct periods:May to June and September to *** genotyping,seven distinct genotypes ***.3 was the most prevalent,accounting for 54.9%of cases,followed by GI.1(12.7%),GI.2(9.9%),GII.5(7.0%),GI.5(2.8%),GI.6(1.4%),GII.1(1.4%),and untyped cases(9.9%).A phylogenetic analysis of GII.3 identified three distinct groups,with 15 notable SNPs ***:This study offers a comprehensive analysis of the persistent prevalence of HuSaV outbreaks in Chaoyang district,beijing Municipality,*** time,the diversity of HuSaV subtypes has shifted,and it is now recognized as the second leading viral agent responsible for AGE *** highlights the importance of ongoing surveillance in the future.

Objective To establish and compare the pulsed-field gel electrophoresis （PFGE）, multiple-locus variable number tandem repeat analysis （MLVA） and automated ribotyping for subtyping of Citrobacter strains. Methods PFGE protocol was optimized in terms of plug preparation procedure, restriction enzymes and configuration of electrophoretic parameters. MLVA method was evaluated by finding variable number tandem repeats in two genomes of Citrobacter strains. The ribotyping was performed by using the automated RiboPrinter system. Results We optimized the plug preparation procedure, focused on the cell suspension concentration （turbidity of 2.5 to 3.5）, SDS addition （no SDS needed） and lysis time （1 h）, and selected the appropriate restriction enzyme （Xbal） and the electrophoretic parameters （1.0 s-20.0 s for 19 h） of PFGE. There was nearly no discriminatory power of MLVA between Citrobacter strains. For 51 Citrobacter strains, automated ribotyping gave a D-value of 0.9945, while PFGE gave a D-value of 0.9969. Both PFGE and automated ribotyping clustered strains from the same sources （with the same species from the same place at the same time identified as the same source） and divided strains from different sources （from different years, places and hosts） into different subtypes. Conclusion PFGE protocol established in this paper and automated ribotyping are suitable for application in Citrobacter subtyping.
