Dear Editor,Activated hepatic stellate cells(HSCs)synthesizing a large amount of extracellular matrix(ECM)are the key events to hepatic fibrosis.1 However,there are no effective drugs for curing liver fibrosis in the clinic.1 Therefore,clarifying the formation mechanism of liver fibrosis is of great importance for finding anti-liver fibrosis drug ***,an increasing amount of research has shown that necroptosis regulated by receptor-interacting protein kinase 3(RIP3)plays an important role in inflammatory disease injury and could be used as a drug target.2 We found that RIP3 was highly expressed in liver tissues of wildtype(WT)mice infected with Schistosoma japonicum(***),mainly at 6 w(weeks)and 8 w post-infection(Fig.1a)。

Multi-principal element alloys(MPEAs)have shown extraordinary properties in different ***,the composition design of MPEAs is still challenging due to the complicated interactions among principal elements(PEs),and even more challenging with precipitates *** can be either beneficial or detrimental in alloys,thus it is important to control precipitates formation on purpose during alloy *** this work,cluster-plus-glue-atom model(CGM)composition design method which is usually used to describe short-range order in traditional alloys has been successfully extended to MPEAs for precipitation *** key challenge of extending CGM to MPEAs is the determination of center atom since there are no solvent or solute in *** has found that the element type of center atom was related not only with chemical affinity,but also with atomic volume difference in MPEAs,which has inevitable effect on atomic *** on experimental data of MPEAs with precipitates,it was found that elements with either stronger chemical affinity or larger volume difference with other PEs would occupy the center site of ***,a cluster index(P_(C)),which considers both chemical affinity and atomic volume factors,was proposed to assist the determination of center atom in *** on the approach,a solid-solution Zr-Ti-V-Nb-Al BCC alloy was obtained by inhibiting the precipitation,while precipitation-strengthened Al-Cr-FeNi-V FCC alloy and Al-Co-Cr-Fe-Ni BCC alloy were designed by promoting the *** experimental results demonstrated that the approach could provide a relatively simple and accurate predication of precipitation and the compositions of precipitations were in line with PEs in cluster in *** research may open an effective way for composition design of MPEAs with desired phase structure.

AIM: To investigate the intratumoral expression of metastasis-associated in colon cancer 1 (MACC1) and c-Met and determine their clinical values associated with hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC). METHODS: A retrospective study admitted three hundred fifty-four patients with HBV-related HCC. The expression and distribution of MACC1 and c-Met were assessed by quantitative real-time polymerase chain reaction and immunohistochemistry staining. Prognostic factors influencing survival, metastasis and recurrence were assessed. RESULTS: Intratumoral MACC1 level was found to be associated with HCC disease progression. Both median tumor-free survival (TFS) and overall survival (OS) were significantly shorter in the postoperative HCC patients with high intratumoral MACC1 expression, as compared to those with low intratumoral MACC1 levels (TFS: 34 mo vs 48.0 mo, P < 0.001; OS: 40 mo vs 48 mo, P < 0.01). Multivariable analysis indicated that high MACC1 expression or co-expression with c-Met were independent predictors for HCC clinic outcome (P < 0.001). CONCLUSION: High intratumoral MACC1 expression can be associated with enhanced tumor progression and poor outcome of HBV-related HCC. MACC1 may serve as a prognostic biomarker for postoperative HCC.

Lesion mimic often exhibits leaf disease-like symptoms even in the absence of pathogen infection,and is characterized by a hypersensitive-response(HR)that closely linked to plant disease *** this,only a few lesion mimic genes have been identified in *** this investigation,a lesion mimic wheat mutant named je0297 was discovered,showing no alteration in yield components when compared to the wild type(WT).Segregation ratio analysis of the F_(2)individuals resulting from the cross between the WT and the mutant revealed that the lesion mimic was governed by a single recessive gene in *** Bulked segregant analysis(BSA)and exome capture sequencing,we mapped the lesion mimic gene designated as lm6 to chromosome *** gene fine mapping using 3315 F_(2)individuals delimited the lm6 within a 1.18 Mb *** this region,we identified 16 high-confidence genes,with only two displaying mutations in ***,one of the two genes,responsible for encoding flavonol synthase,exhibited altered expression *** phenotype analysis of TILLING mutants confirmed that the gene encoding flavonol synthase was indeed the causal gene for *** sequencing analysis revealed that the DEGs between the WT and mutant were significantly enriched in KEGG pathways related to flavonoid biosynthesis,including flavone and flavonol biosynthesis,isoflavonoid biosynthesis,and flavonoid biosynthesis ***,more than 30 pathogen infection-related(PR)genes exhibited upregulation in the *** to this expression pattern,the flavonoid content in je0297 showed a significant decrease in the 4^(th)leaf,accompanied by a notable accumulation of reactive oxygen,which likely contributed to the development of lesion mimic in the *** investigation enhances our comprehension of cell death signaling pathways and provides a valuable gene resource for the breeding of disease-resistant wheat.

AIM: To study the biological and clinical characteristics of transcription factor forkhead box protein 3 (FOXP3) in hepatocellular carcinoma (HCC). METHODS: We analyzed the expression and localization of FOXP3 in HCC tissues and cell lines to evaluate its biological features. The relationship between FOXP3 staining and clinical risk factors of HCC was assessedto identify the clinical characteristics of FOXP3 in HCC. RESULTS: The mRNA and protein expression of FOXP3 were found in some hepatoma cell lines. Immunohistochemical (IHC) analysis of HCC sections revealed that 48% of HCC displayed FOXP3 staining, but we did not find any FOXP3 staining in normal liver tissues and para-tumor tissues. IHC and Confocal analysis showed that the expressions of FOXP3 were mainly present in the nucleus and cytoplasm of tumor cells in tissues or cell lines. In HCC, the distribution of FOXP3 was similar to that of the cirrhosis, but not to the hepatitis B virus. Those findings implicate that FOXP3 staining seems to be associated with the high risk of HCC. CONCLUSION: The clinical characteristics of FOXP3 in HCC warrants further studies to explore its functions and roles in the cirrhosis and development of HCC.
