Activation of parvalbumin interneurons in anterior cingulate cortex impairs observational fear

The ability to detect conspecific＇s distress is crucial for animal survival. In rodent models, observational fear （OF） occurs when one animal perceives another fear related negative emotions, which may model certain behaviors caused by witnessing traumatic experiences in humans. Anterior cingulate cortex （ACC） has been showed to play a crucial role in OF. However, cellular and neural circuit basis relating to ACC governing OF is poorly understood. Here, we used Designer Receptor Exclusively Activated by a Designer Drug （DREADD） system to investigate the cell type specific circuit mechanism of ACC in OF. Firstly, inhibitory hM4D （Gi） designer receptor together with clozapine N-oxide （CNO） injection was applied to inactivate ACC neurons in the observer mice. We found that, chemogenetic inhibition of ACC resulted in a decreased freezing response in the observer mice. Next, combining PV-ires-Cre mice and Cre-dependent DREADD system, we selectively targeted the ACC parvalbumin （PV） interneurons with the excitatory hM3D （Gq） designer receptor. Activation of ACC PV interneurons following CNO injection reduced freezing response in the observer mice, while had no effect on freezing response in the demon- strator mice. Finally, monosynaptic rabies retrograde tracing revealed that ACC PV interneurons receive inputs from the mediodorsal thalamic nucleus （MD） and the ventromedial thalamic nucleus （VM）, both known for their roles in OF. Taken together, these findings reveal that ACC activation is important for OF, during which PV interneurons in ACC play an important regulatory role. Abnormal function of ACC PV interneurons might contribute to the pathology of empathy- deficits related diseases, such as autism and schizoohrenia.

Consecutively hospitalized patients with confirmed coronavirus disease 2019(COVID-19)in Wuhan,China were retrospectively enrolled from January 2020 to March 2020 to investigate the association between the use of renin–angiotensin system inhibitor(RAS-I)and the outcome of this *** between the use of RAS-I(angiotensin-converting enzyme inhibitor(ACEI)or angiotensin receptor blocker(ARB)),ACEI,and ARB and in-hospital mortality were analyzed using multivariate Cox proportional hazards regression models in overall and subgroup of hypertension status.A total of 2771 patients with COVID-19 were included,with moderate and severe cases accounting for 45.0%and 36.5%,respectively.A total of 195(7.0%)patients ***-I(hazard ratio(HR)=0.499,95%confidence interval(CI)0.325–0.767)and ARB(HR=0.410,95%CI 0.240–0.700)use was associated with a reduced risk of all-cause mortality among patients with *** patients with hypertension,RAS-I and ARB applications were also associated with a reduced risk of mortality with HR of 0.352(95%CI 0.162–0.764)and 0.279(95%CI 0.115–0.677),***-I exhibited protective effects on the survival outcome of *** use was associated with a reduced risk of all-cause mortality among patients with COVID-19.

Aerodynamic characteristics of morphing wing withflexible leading-edge

The morphing wing can improve the flight performance during different ***,research has been subject to limitations in aerodynamic characteristics of the morphing wing with a flexible *** computational fluid dynamic method and dynamic mesh were used to simulate the continuous morphing of the flexible *** comparing the steady aerodynamic characteristics of morphing and conventional wings,this study examined the unsteady aerodynamic characteristics of morphing wings with upward and downward deflections of the leading-edge at different *** numerical results show that for the steady aerodynamic,the leading-edge deflection mainly affects the stall *** downward deflection of the leading-edge increases the stall angle of attack and nose-down pitching *** results are opposite for the upward *** the unsteady aerodynamic,at a small angle of attack,the transient lift coefficient of the upward deflection,growing with the increase of deflection frequency,is larger than that of the static *** transient lift coefficient of the downward deflection,decreasing with the increase of deflection frequency,is smaller than that of the static ***,at a large angle of attack,an opposite effect of deflection frequency on the transient lift coefficient was *** transient lift coefficient is larger than that of the static case when the leading edge is in the nose-up stage,and lower than that of the static one in the nose-down stage.

Time-domain nonlinear aeroelastic analysis and wind tunnel test of a flexible wing using strain-based beam formulation

A theoretical formulation for time-domain nonlinear aeroelastic analysis of a flexible wing model is presented and validated by wind tunnel tests. A strain-based beam model for nonlinear structural analysis is combined with the Unsteady Vortex Lattice Method(UVLM) to form the complete framework for aeroelastic analysis. The nonlinear second-order differential equations are solved by an implicit time integration scheme that incorporates a Newton-Raphson sub-iteration technique. An advanced fiber optic sensing technique is firstly used in a wind tunnel for measuring large structural deformations. In the theoretical study, the nonlinear flutter boundary is determined by analyzing the transient response about the nonlinear static equilibrium with a series of flow velocities. The gust responses of the wing model at various gust frequencies are also studied. Comparisons of the theoretical and experimental results show that the proposed method is suitable for determining the nonlinear flutter boundary and simulating the gust response of flexible wings in the time domain.

2 D nanomaterials generally exhibit enhanced physiochemical and biological functions in biomedical applications due to their high surface-to-volume ratio and surface charge. Conventional cancer chemotherapy based on nanomaterials has been hindered by their low drug loading and poor penetration in tumor tissue. To overcome these difficulties, novel materials systems are urgently needed. Hereby, the lanthanide-based porphyrin metal–organic framework(MOF) nanosheets(NSs) with promising cancer imaging/chemotherapy capacities are fabricated, which display superior performance in the drug loading and tumor tissue penetration. The biodegradable PPF-Gd NSs deliver an ultrahigh drug loading(>1500%)and demonstrate the stable and highly sensitive stimuli-responsive degradation/release for multimodal tumor imaging and cancer chemotherapy. Meanwhile, PPF-Gd NSs also exhibit excellent fluorescence and magnetic resonance imaging capability in vitro and in vivo. Compared to the traditional doxorubicin(DOX)chemotherapy, the in vivo results confirm the evident suppression of the tumor growth by the PPF-Gd/DOX drug delivery system with negligible side effects. This work further supports the potential of lanthanide-based MOF nanomaterials as biodegradable systems to promote the cancer theranostics technology development in the future.
