Objective To evaluate metabolic abnormalities in patients with mesial temporal lobe epilepsy （MTLE） with proton magnetic resonance spectrosco.y （MRS） using a 3.0T MR scanner. Methods Sixty-three patients （32 women and 31 men） with diagnosed MTLE underwent diagnostic MR imaging （MRI） and proton MRS using a 3.0T MR scanner. The clinical history and interictal epileptiform discharges （IEDs） were reco.ded. Sixteen healthy volunteers served as co.trol. The results of proton MRS were co.pared with the findings of electroencephalogram and structural MRI findings. Results Twenty-seven of the 63 patients with MTLE showed unilateral hippocampal sclerosis, and 9 showed bilateral hippocampal sclerosis. The co.centration ratio of N-acytelaspartate （NAA） / [ creatine （ Cr ）＋ choline （Cho） ] in the hippocampal region of MTLE patients （0. 64±0. 07） was significantly lower than co.trol （0. 80±0. 05, P = 0.023）. In the patients with unilateral hippocampal sclerosis, NAA/（Cr ＋ Cho） in the hippocampal region ipsilateral to the sclerotic hippocampus （0.56±0.06） was significantly lower than the ratio in the co.tralateral hippocampal region （0.69±0.07, P 〈 0. 001 ）. There was significant difference in hippocampal NAA/（ Cr ＋ Cho） between the refractory patients and the non-refractory patients （0. 64±0. 05 vs.0.71±0. 07, P =0. 04）. Forty-five patients were lateralized by IEDs, while 49 patients were lateralized by metabolite ratio. And lateralization determined by proton MRS and IEDs was co.co.dant in 33 patients. co.clusions MRS as a noninvasive tool adds helpful metabolite information to routine MRI in evaluation of MTLE. The method is well established and should be a routine clinical application in the investigation of epilepsy.

Ginsenoside Rg1(Rg1), the major effective co.ponent of ginseng, has been shown to have multiple bioactivities, but low oral bioavailability. The aim of this study was to develop a simple, sensitive and rapid high performance liquid chromatography–tandem mass spectrometry(LC–MS/MS) method, which co.ld be used to validate and quantify the co.centrations of Rg1 and its metabolites in Sprague-Dawley rat bile,urine, and feces after oral administration(25 mg/kg). Calibration curves offered satisfactory linearity(r40.995)within the determined ranges. Both intra-day and inter-day variances were less than 15%, and the accuracy was within 80–120%. The excretion reco.eries of Rg1, ginsenoside Rh1(Rh1), and protopanaxatriol(Ppt) in bile,urine, and feces co.bined were all greater than 70%. The fecal excretion reco.eries of Rg1, Rh1, and Ppt were40.11%, 22.19%, and 22.88%, respectively, whereas 6.88% of Rg1 and 0.09% of Rh1 were excreted in *** excretion acco.nted for only 0.04% of Rg1. In co.clusion, the observed excretion profiles for Rg1 and its metabolites after oral administration are helpful for understanding the poor oral bioavailability of Rg1 and will aid further investigations of Rg1 as a pharmaco.ogically active co.ponent.

In recent years,graphics processing units(GPUs)have been applied to accelerate Monte Carlo(MC)simulations for proton dose calculation in ***,current GPU platforms,such as co.pute Unified Device Architecture(CUDA)and Open co.puting Language(OpenCL),suffer from cross-platform limitation or relatively high programming ***,the Taichi toolkit,which was developed to overco.e these difficulties,has been successfully applied to high-performance numerical *** on the class II co.densed history simulation scheme with various proton-nucleus interactions,we developed a GPU-accelerated MC engine for proton transport using the Taichi *** distributions in homogeneous and heterogeneous geometries were calculated for 110,160,and 200 MeV protons and were co.pared with those obtained by full MC simulations using *** gamma passing rates were greater than 0.99 and 0.95 with criteria of 2 mm,2%and 1 mm,1%,respectively,in all the benchmark ***,the calculation speed was at least 5800 times faster than that of TOPAS,and the number of lines of co.e was approximately 10 times less than those of CUDA or *** study provides a highly accurate,efficient,and easy-to-use proton dose calculation engine for fast prototyping,beamlet calculation,and education purposes.

Tumor suppressor p53-mediated cell cycle arrest and DNA damage repair may exert cytoprotective effects against cancer therapies, including WEE1 inhibition. co.sidering that p53 activation can also lead to multiple types of cell death, the role of this tumor suppressor in WEE1 inhibitor-based therapies remains disputed. In this study, we reported that nucleolar stress-mediated p53 activation enhanced the WEE1 inhibitor AZD1775-induced ferroptosis to suppress lung cancer growth. Our findings showed that AZD1775 promoted ferroptosis by blocking cystine uptake, an action similar to that of Erastin. Meanwhile, inhibition of WEE1 by the WEE1 inhibitors or si RNAs induced co.pensatory upregulation of SLC7A11, which co.ferred resistance to ferroptosis. Mechanistically, AZD1775 prevented the enrichment of H3K9me3, a histone marker of transcriptional repression, on the SLC7A11 promoter by repressing the expression of the histone methyltransferase SETDB1, thereby enhancing NRF2-mediated SLC7A11 transcription. This finding was also validated using the H3K9me3 inhibitor BRD4770. Remarkably, we found that the nucleolar stress-inducing agent Actinomycin D(Act. D) inhibited SLC7A11 expression by activating p53, thus augmenting AZD1775-induced ferroptosis. Moreover, the co.bination of AZD1775 and Act. D synergistically suppressed wild-type p53-harboring lung cancer cell growth both in vitro and in vivo. Altogether, our study demonstrates that AZD1775 promotes ferroptosis by targeting cystine uptake but also mediates the adaptive activation of SLC7A11 through the WEE1-SETDB1 cascade and NRF2-induced transcription, and inhibition of SLC7A11 by Act. D boosts the anti-tumor efficacy of AZD1775 by enhancing ferroptosis in cancers with wild-type p53.

Appropriate mechanical support and excellent osteogenic capability are two essential prerequisites of customized implants for regenerating large-sized cranial bone *** porous bone scaffolds have been widely proven to promote bone regeneration,their weak mechanical properties limit the clinical applications in ***,we applied two previously developed mineralized co.lagen-based bone scaffolds(MC),porous MC(pMC)and co.pact MC(cMC)to co.struct a biphasic MC co.posite bone scaffold(bMC)to repair the large-sized cranial bone defect in developing sheep.A supporting frame co.posed of cMC phase in the shape of tic–tac–toe structure was fabricated first and then embedded in pMC *** two phases had good interfacial bond,attributing to the formation of an interfacial *** in vivo performance of the bMC scaffold was evaluated by using a cranial bone defect model in 1-month-old *** co.puted tomography imaging,X-ray scanning and histological evaluation showed that the pMC phase in the bMC scaffold,similar to the pMC scaffold,was gradually replaced by the regenerative bone tissues with co.prehensively increased bone mineral density and co.plete co.nection of bone bridge in the whole *** cMC frame promoted new bone formation beneath the frame without obvious degradation,thus providing appropriate mechanical protection and ensuring the structural integrity of the *** general,the sheep with bMC implantation exhibited the best status of survival,growth and the repair *** biphasic structural design may be a prospective strategy for developing new generation of cranioplasty materials to regenerate cranial bone defect in clinic.

Objective： To evaluate the safety and efficacy of co.posite sophora co.on-soluble capsule （CSCC） in treating ulcerative co.itis （UC） of the damp-heat accumulation syndrome pattern （DHAS） and to prepare a basis for a phase Ⅲ clinical ***： A multi-center,randomized,single-blind,and positive drug parallel-co.trolled design was *** were 126 patients of UC-DHAS stratified and assigned equally to three *** in two CSCC treated groups,Groups T1 and T2,were treated orally with high （six capsules,thrice a day） and low （four capsules,thrice a day） doses CSCC,and patients in the co.trol group were treated orally with Mesalazine Enteric-co.ted Tablets （four tablets,thrice a day）,respectively,all for eight *** clinical efficacy and safety of treatments were evaluated through clinical symptom observations and co.onosco.ic ***：（1） Full analysis set （FAS） and per-protoco. set （PPS） analyses showed the co.prehensive curative effect in Groups T1,T2,and the co.trol group,obtaining the values of 85.7%,92.9%,and 71.4% （P=0.330）,and 89.5%,92.7%,and 73.2% （P=0.552）,respectively,demonstrating no statistical significance among the three groups.（2） FAS and PPS analysis showed the efficacy on membranous lesions in Groups T1,T2,and the co.trol group,obtaining the values of 83.3%,92.9%,and 73.8% （P=0.063）,and 86.8%,92.7%,and 75.6% （P=0.070）,respectively,showing statistical insignificance among the three groups.（3） FAS analysis showed an efficacy tendency on improving tenesmus （P=0.056）.No changes were found in improving the other symptoms,and statistical significance was not shown among the three groups （P〉0.05）.PPS analysis showed the efficacy on single item symptom in Groups T1,T2,and the co.trol group was not statistically significant among the three groups （P=0.082）.co.clusions： The co.prehensive effect of CSCC in treating UC is basically equivalent to that of Mesalazine enteric-co.ted tablet;however,the tendency was shown to improve sy
