MicroRNA-365 Knockdown Prevents Ischemic Neuronal Injury by Activating Oxidation Resistance 1-Mediated Antioxidant Signals

Micro RNA-365(mi R-365) is upregulated in the ischemic brain and is involved in oxidative damage in the diabetic rat. However, it is unclear whether mi R-365 regulates oxidative stress(OS)-mediated neuronal damage after ischemia. Here, we used a transient middle cerebral artery occlusion model in rats and the hydrogen peroxide-induced OS model in primary cultured neurons to assess the roles of mi R-365 in neuronal damage. We found that mi R-365 exacerbated ischemic brain injury and OS-induced neuronal damage and was associated with a reduced expression of OXR1(Oxidation Resistance 1). In contrast, mi R-365 antagomir alleviated both the brain injury and OXR1 reduction. Luciferase assays indicated that mi R-365 inhibited OXR1 expression by directly targeting the 30-untranslated region of Oxr1. Furthermore, knockdown of OXR1 abolished the neuroprotective and antioxidant effects of the mi R-365 antagomir. Our results suggest that mi R-365 upregulationincreases oxidative injury by inhibiting OXR1 expression,while its downregulation protects neurons from oxidative death by enhancing OXR1-mediated antioxidant signals.

Identification of protein targets for the antidepressant effects of Kai-Xin-San in Chinese medicine using isobaric tags for relative and absolute quantitation

Kai-Xin-San consists of Ginseng Radix, Polygalae Radix, Acori Tatarinowii Rhizoma, and Poria at a ratio of 3:3:2:2. Kai-Xin-San has been widely used for the treatment of emotional disorders in China. However, no studies have identified the key proteins implicated in response to Kai-Xin-San treatment. In this study, rat models of chronic mild stress were established using different stress methods over 28 days. After 14 days of stress stimulation, rats received daily intragastric administrations of 600 mg/kg Kai-Xin-San. The sucrose preference test was used to determine depression-like behavior in rats, while isobaric tags were used for relative and absolute quantitation-based proteomics to identify altered proteins following Kai-Xin-San treatment. Kai-Xin-San treatment for 2 weeks noticeably improved depression-like behaviors in rats with chronic mild stress. We identified 33 differentially expressed proteins: 7 were upregulated and 26 were downregulated. Functional analysis showed that these differentially expressed proteins participate in synaptic plasticity, neurodevelopment, and neurogenesis. Our results indicate that Kai-Xin-San has an important role in regulating the key node proteins in the synaptic signaling network, and are helpful to better understand the mechanism of the antidepressive effects of Kai-Xin-San and to provide objective theoretical support for its clinical application. The study was approved by the Ethics Committee for Animal Research from the Chinese PLA General Hospital(approval No. X5-2016-07) on March 5, 2016.

Arsenic Disulfide Induced Apoptosis and Concurrently Promoted Erythroid Differentiation in Cytokine-Dependent Myelodysplastic Syndrome-progressed Leukemia Cell Line F-36p with Complex Karyotype Including Monosomy 7

Objective： Acute myeloid leukemia progressed from myelodysplastic syndrome （MDS/AML） is generally incurable with poor prognosis for complex karyotype including monosomy 7 （-7）. Qinghuang Powder （青黄, QHP）, which includes Qing Dai （ Indigo naturalis） and Xiong Huang （realgar） in the formula, is effective in treating MDS or MDS/AML even with the unfavorable karyotype, and its therapeutic efficacy could be enhanced by increasing the Xiong huang content in the formula, while Xiong huang contains 〉 90% arsenic disulfide （As2S2）. F-36p cell line was established from a MDS/AML patient with complex karyotype including -7, and was in cytokine-dependent. The present study was to investigate the effects of As2S2 on F-36p cells. Methods： Cell proliferation was measured by an 3-（4,5-dimethylthiazol-2-yl）-2, 5-diphenyltetrazolium bromide （MTT） assay. Cell apoptosis was identified by Annexin V-staining. Cell viability was determined by a propidium iodide （PI） exclusion. Erythroid differentiation was evaluated by the expression of cell surface antigen CD235a （GpA）. Results： After treatment with As2S2 at concentrations of 0.5 to 16 p. mol/L for 72 h, As2S2 inhibited the proliferation of F-36p cells. The 50% inhibitory concentrations （IC50） of As2S2 against the proliferation of F-36p cells was 6 I~ mol/L. The apoptotic cells significantly increased in a dose-dependent mannar （P〈0.05）. The cell viabilities were significantly inhibited by As2S2 dose-dependent in a dose-dependent manner （P〈0.05）. Significant increases of CD235a-positive cells were concurrently observed （P〈0.05） also in a dose- dependent manner. Conclusions： As2S2 could inhibit proliferation and viability, induce apoptosis, and concurrently promote erythroid differentiation dose-dependently in F-36p cells. Respiratory syncytial virus(RSV)is the most common cause of viral bronchiolitis among children worldwide,yet there is no vaccine for RSV *** study investigates the potential of cube and sphere-shaped cerium oxide nanoparticles(CNP)to modulate reactive oxygen(ROS)and nitrogen(RNS)species and immune cell phenotypes in the presence of RSV infection in vitro and in *** and sphere-shaped CNP were synthesized by hydrothermal and ultrasonication methods,***-chemical characterization confirmed the shape of sphere and cube CNP and effect of various parameters on their particle size distribution and zeta *** vitro results revealed that sphere and cube CNP differentially modulated ROS and RNS levels in J774 ***,cube CNP significantly reduced RSV-induced ROS levels without affecting RNS levels while sphere CNP increased RSV-induced RNS levels with minimal effect on ROS *** CNP drove an M1 phenotype in RSV-infected macrophages in vitro by increasing macrophage surface expression of CD80 and CD86 with a concomitant increase in TNFαand IL-12p70,while simultaneously decreasing M2 CD206 *** administration of sphere and cube-CNP were well-tolerated with no observed toxicity in BALB/c ***,cube CNP preferentially accumulated in murine alveolar macrophages and induced their activation,avoiding enhanced uptake and activation of other inflammatory cells such as neutrophils,which are associated with RSV-mediated *** conclusion,we report that sphere and cube CNP modulate macrophage polarization and innate cellular responses during RSV infection.

Effect of probiotics on pro-inflammatory cytokines and NF-κB activation in ulcerative colitis

AIM:To demonstrate the therapeutic effect of probiotics in patients with ulcerative colitis(UC),and their effect on inflammatory mediators and nuclear factor(NF)κB activation in these ***:Thirty patients with mild to moderate UC were randomly classified into two groups:sulfasalazine group,who received sulfasalazine 2400 mg/d;and probiotic group,who received sulfasalazine 2400 mg/d with *** patients were investigated before and after 8 wk of treatment with probiotic(Lactobacillus delbruekii and Lactobacillus fermentum).Colonic activity of myeloperoxidase(MPO)was assayed with UV spectrophotometry,the colonic content of interleukin(IL)-6 was determined by enzyme-linked immunosorbent assay(ELISA),fecal calprotectin was determined by ELISA,and expression of NF-κB p65 and tumor necrosis factor (TNF)-αproteins in colonic tissue was identified by immunohistochemistry and reverse transcription polymerase chain reaction,***:At the start of the study,colonic mucosal injury and inflammation were demonstrated in UC patients by hematoxylin and eosin staining,and an increase in colonic MPO activity,fecal calprotectin,and expression of colonic TNF-αand NF-κB p65 *** use of probiotic for 8 wk significantly ameliorated the inflammation by decreasing the colonic concentration of IL-6,expression of TNF-αand NF-κB p65,leukocyte recruitment,as demonstrated by a decrease in colonic MPO activity,and the level of fecal calprotectin compared to sulfasalazine group and the control group(P<0.05).CONCLUSION:Oral supplementation with probiotics could be helpful in maintaining remission and preventing relapse of UC.

Antimalarial activity of Ageratum conyzoides in combination with chloroquine and artesunate

Objective:To determine the suppressive and curative activity of aqueous leaf extract of Ageratum conyzoides(***) in combination with chloroquine and artesunate, respectively against Plasmodium berghei infection in ***:Using malaria(Plasmodium berghei) infected albino mice of both sexes,aqueous extracts of *** in combination with chloroquine and artesunate were tested for antimalarial activity,***-day suppressive test and Rane’s curative test were carried ***:Suppressive tests showed significant dose dependent reduction in parasitemia level produced by the extract-chloroquine and extract-artesunate *** activities of both extract-drug combinations were greater than the individual drugs ***-chloroquine(100:5) produced the highest suppressive effect(98%suppression).Curative tests showed absolute survival in two extract-drug *** extract-drug combinations produced higher curative effects than the individual drugs *** highest dose combinations of extract-chloroquine(100:5) and extract-artesunate(100:5) produced absolute parasitemia clearance(cure) in the infected mice. Conclusions:The study indicated that aqueous extract of *** had the ability to potentiate the antimalarial activity of chloroquine and artesunate against induced plasmodiasis in *** contributes a lot in the malaria endemic and poverty stricken tropics.

Indonesia is one of the countries with the highest burden of tuberculosis and drug-resistant tuberculosis(DR-TB)across the *** on data from the World Health Organization(WHO)Global TB Report 2023,it is estimated that there are 10000 cases of DR-TB in ***,a novel antitubercular drug,has been implemented to treat DR-TB *** was administered either a shorter(9 months)or individualized treatment regimen(18 months).However,long treatment duration with various adverse events affects patient ***,a short treatment with less medication is urgently *** 2022,the WHO announced an alternative regimen-bedaquiline,pretomanid,linezolid,and moxifloxacin(BPaLM)to treat DR-TB patients for six months without resistance to fluoroquinolones[1].This recommendation is based on previous clinical trials of TB,Zenix TB,and *** introduction of BPaL and BPaLM provided a bright future for treating DR-TB patients.
