Characterization and Optimization of Amylase Production in Strains LZ-10 and LZ-11 Belonging to Bacillus subtilis

In order to obtain pure enzyme with high activity,two amylase producing strains were isolated from soil samples,and named as the strains LZ-10 and *** to morphologic observation,physiology and biochemistry experiments,16S rRNA and gyrB gene analysis,the strains LZ-10 and LZ-11 were identified as Bacillus *** the method of ammonium sulfate,DEAE-52 anion purify enzyme,finally used polyacrylamide gel electrophoresis(SDS-PAGE)to detect molecular *** strain LZ-10 had an amylase activity of 123.3 U·mL^(-1),a purification factor of 6.8,a recovery rate of 69.5% and an optimal temperature of 50℃.The amylase activity of the strain LZ-11 was 59.91 U·mL^(-1),the purification factor was 4.5,the recovery rate was 60.5%,and the optimum temperature was 55℃.The commodity enzyme derived from Bacillus subtilis was 37.5 U·mL^(-1).The relative molecular weight of amylase activity from the two strains was 55 *** thermal stability and pH stability were higher than those of commercialized amylase.

We study the temperature-dependent Raman spectrum of hexagonal YMnO3 films prepared by a chemical solution method. There are seven Raman peaks （3A1 ＋ E1 ＋ 4E2） of the film identified at room temperature. From the results of temperature dependence of the Raman spectrum, it is deduced that the YMnO3 film has a magnetic phase transition temperature of about 123K. The temperature variation phonon mode at 685cm^-1 shows an anomalous frequency variation near 123 K, suggesting either a more complex mechanism of spin-phonon coupling or strong mixing of phonon modes. The reason for the higher antiferromagnetic Néel temperature TN of the film than that of the bulk counterpart is also discussed.

Metal halide perovskites(MHP)are potential candidates for the photocatalytic reduction of CO_(2)due to their long photogenerated carrier lifetime and charge diffusion ***,the conventional long-chain ligand impedes the adsorption and activation of CO_(2)molecules in practical ***,a ligand modulation technology is employed to enhance the photocatalytic CO_(2)reduction activity of lead-free Cs_(2)AgInCl_(6)microcrystals(MCs).The Cs_(2)AgInCl_(6)MCs passivated by Oleic acid(OLA)and Octanoic acid(OCA)are used for photocatalytic CO_(2)*** results show that the surface defects and electronic properties of Cs_(2)AgInCl_(6)MCs can be adjusted through ligand *** with the OLA-Cs_(2)AgInCl_(6),the OCA-Cs_(2)AgInCl_(6)catalyst demonstrated a significant improvement in the catalytic yield of CO and CH_(4).The CO and CH_(4)catalytic yields of OCA-Cs_(2)AgInCl_(6)reached 171.88 and34.15μmol g^(-1)h^(-1)which were 2.03 and 12.98 times higher than those of OLA-Cs_(2)AgInCl_(6),and the total electron consumption rate of OCA-Cs_(2)AgInCl_(6)was 615.2μmol g^(-1)h^(-1)which was 3.25 times higher than that of OLA-Cs_(2)AgInCl_(6).Furthermore,in situ diffuse reflectance infrared Fourier transform spectra revealed the enhancement of photocatalytic activity in Cs_(2)AgInCl_(6)MCs induced by ligand *** study illustrates the potential of lead-free Cs_(2)AgInCl_(6)MCs for efficient photocatalytic CO_(2)reduction and provides a ligand modulation strategy for the active promotion of MHP photocatalysts.

We execute the density functional theory(DFT) and time-dependent density functional theory(TDDFT) approaches to make a detailed exploration about excited state luminescent properties as well as excited state intramolecular proton transfer(ESIPT) mechanism for the novel 2,6-dimethyl phenyl(DMP-HBT-py) system. Firstly, we check and confirm the formation and stabilization of hydrogen bonding interaction for DMP-HBT-py. Via optimized geometrical parameters of primary chemical bond and infrared(IR) spectra, we find O–H··· N hydrogen bond of DMP-HBT-py should be strengthened in S1 state. Insights into frontier molecular orbitals(MOs) analyses, we infer charge redistribution and charge transfer(ICT)phenomena motivate ESIPT trend. Via probing into potential energy curves(PECs) in related electronic states, we come up with the ultrafast ESIPT behavior due to low potential barrier. Furthermore, we search the reaction transition state(TS)structure, the ultrafast ESIPT behavior and mechanism of DMP-HBT-py compound can be re-confirmed. We sincerely wish this work could play roles in further developing novel applications based on DMP-HBT-py compound and in promoting efficient solid emitters in OLEDs in future.

Triggering receptor expressed on myeloid cells-1 (TREM-1) is a member of the immunoglobulin superfamily. As an amplifier of the inflammatory response, TREM-1 is mainly involved in the production of inflammatory mediators and the regulation of cell survival. TREM-1 has been studied in infectious diseases and more recently in non-infectious disorders. More and more studies have shown that TREM-1 plays an important pathogenic role in kidney diseases. There is evidence that TREM-1 can not only be used as a biomarker for diagnosis of disease but also as a potential therapeutic target to guide the development of novel therapeutic agents for kidney disease. This review summarized molecular biology of TREM-1 and its signaling pathways as well as immune response in the progress of acute kidney injury, renal fibrosis, diabetic nephropathy, immune nephropathy, and renal cell carcinoma.

Hepatic ischemia reperfusion injury(HIRI) is a clinical condition which may lead to cellular injury and organ dysfunction. The role of nitric oxide(NO) in HIRI is complicated and inconclusive. NO produced by endothelial nitric oxide synthase(e NOS) activation plays a protective role during early HIRI. But e NOS overexpression and the resulting excessive NO bioavailability can aggravate liver injury. NO induced by inducible nitric oxide synthase(i NOS) may have either a protective or a deleterious effect during the early phase of HIRI, but it may protect the liver during late HIRI. Here, we reviewed the latest findings on the role of NO during HIRI:(1) NO exerts a protective effect against HIRI by increasing NO bioavailability, downregulating p53 gene expression, decreasing inflammatory chemokines, reducing ROS via inhibiting the mitochondrial respiratory chain, activating s GCGTP-c GMP signal pathway to reduce liver cell apoptosis, and regulating hepatic immune functions;(2) e NOS protects against HIRI by increasing NO levels, several e NOS/NO signal pathways(such as Akt-e NOS/NO, AMPK-e NOS/NO and HIF-1α-e NOS/NO) participating in the anti-HIRI process, and inhibiting over-expression of e NOS also protects against HIRI; and(3) the inhibition of i NOS prevents HIRI. Thus, the adverse effects of NO should be avoided, but its positive effect in the clinical treatment of diseases associated with HIRI should be recognized.

Bacteria-mediated anti-tumor therapy has received widespread attention due to its natural tumor-targeting ability and specific immune-activation *** has made significant progress in breaking the limitations of monotherapy and effectively eradicating tumors,especially when combined with traditional therapy,such as *** to their different biological characteristics,bacteria and their derivatives can not only improve the sensitivity of tumor radiotherapy but also protect normal ***,genetically engineered bacteria and bacteria-based biomaterials have further expanded the scope of their applications in *** this review,we have summarized relevant researches on the application of bacteria and its derivatives in radiotherapy in recent years,expounding that the bacteria,bacterial derivatives and bacteria-based biomaterials can not only directly enhance radiotherapy but also improve the anti-tumor effect by improving the tumor microenvironment(TME)and immune ***,some probiotics can also protect normal tissues and organs such as intestines from radiation via anti-inflammatory,anti-oxidation and apoptosis *** conclusion,the prospect of bacteria in radiotherapy will be very extensive,but its biological safety and mechanism need to be further evaluated and studied.

Cyanobacterial blooms occur in eutrophic lakes worldwide, and greatly impair these ecosystems. To explore influences of cyanobacterial blooms on dynamics of both particulate organic matter(POM) and dissolved organic matter(DOM), which are at the base of the food chain, an investigation was conducted from December 2014 to November 2015 that included various stages of the seasonal cyanobacterial blooms(dominated by M icrocystis) in a large-shallow eutrophic Chinese lake(Taihu Lake). Data from eight sites of the lake are compiled into a representative seasonal cycle to assess general patterns of POM and DOM dynamics. Compared to December, 5-fold and 3.5-fold increases were observed in July for particulate organic carbon(POC, 3.05–15.37 mg/L) and dissolved organic carbon(DOC, 5.48–19.25 mg/L), respectively, with chlorophyll a(Chl a) concentrations varying from 8.2 to 97.7 μg/L. Approximately 40% to 76% of total organic carbon was partitioned into DOC. All C, N, and P in POM and DOC were significantly correlated with Chl a. POC:Chl a ratios were low, whereas proportions of the estimated phytoplankton-derived organic matter in total POM were high during bloom seasons. These results suggested that contributions of cyanobacterial blooms to POM and DOC varied seasonally. Seasonal average C:P ratios in POM and DOM varied from 79 to 187 and 299 to 2 175, respectively. Both peaked in July and then sharply decreased. Redundancy analysis revealed that Chl a explained most of the variations of C:N:P ratios in POM, whereas temperature was the most explanatory factor for DOM. These findings suggest that dense cyanobacterial blooms caused both C-rich POM and DOM, thereby providing clues for understanding their influence on ecosystems.
