Dexamethasone impairs the differentiation and maturation of murine dendritic cells by Toll-like receptor 4-nuclear factor-κB pathway

Background Recent studies have demonstrated that dexamethasone （DEX） interferes with immune responses by targeting key functions of dendritic cells （DCs） at the earliest stage. However, the cellular and molecular mechanisms are still incompletely understood. This study aimed to explore the possible mechanisms by investigating the roles of DEX on differentiation, maturation ＆ function of murine DCs and the effects of DEX on DCs via Toll-like receptor 4 （TLR4）-nuclear factor （NF）-KB mediated signal pathway. Methods Immature DCs （imDCs） were cultured from murine bone marrow （BM） cells. We added DEX into culture medium at different time. The expression of CD11c, CD86 and I-Ab （mouse MHC class II molecule） was determined by flow cytometry. We determined the expression of NF-κB and its inhibitory protein I-κBα by electrophoretic mobility shift assay （EMSA） and Western blotting, respectively. The productions of interleukin （IL）-12p70 and IL-10 in cell culture supernatants were determined by enzyme-linked immunosorbent assay （ELISA）. Results DEX impaired differentiation of DCs from murine bone marrow progenitors, and inhibited lipopolysaccharide （LPS） induced maturation of DCs. DEX significantly inhibited NF-κB expression of normal DCs, the higher the DEX concentration or the longer the DEX treatment time, the more obvious the effect. However, DEX had little effect on LPS-induced NF-KB activation, and partially impaired LPS-induced I-κBα degradation. DEX significantly decreased LPS induced IL-12p70 production by DCs. Interestingly, our results showed a synergistic effect between DEX and LPS on the production of IL-10 by DCs. Conclusions DEX inhibits the differentiation and maturation of murine DCs involved in TLR4-I-κB-NF-κB pathway, and also indirectly impairs Thl development and interferes with the Thl-Th2 balance through IL-12 and/or IL-10 secretion by DCs.

The synthesis, X-ray crystal structure, thermal properties and electrochemistry of the new complex formulated as {Er（DMSO）7}PW12O40 are reported. The single-crystal X-ray analysis reveals that the crystal crystallizes in the monoclinic system, space group P21/c with a=11.767（2）, b=14.909（3）, c=34.905（7）A, β=98.97（3）°, Mr=3591.33, V=6049（2）A^3, Dc=3.944g/cm^3, Z=4, GOOF=1.098, F（000）=6340, R=0.0490 and wR=0.1202. Crystal structure analysis indicates that the Er（Ⅲ） is seven-coordinate with a distorted pentagonal bipyramid and combines to the anion [PW12O40]3- via static electric force.

After dengue virus(DENV)infection,antibody-dependent enhancement(ADE)is easy to occur when the neutralizing antibody(NAb)gradually decreases to a sub-neutralizing *** this cohort surveillance,we utilized sera samples collected from dengue fever patients at different convalescent phases in Jinghong City,to investigate the dynamic change rule of DENV-specific antibodies,and to analyze the risk of ADE caused by secondary infection with heterologous serotypes *** baseline serosurvey,191 four-year and 99 six-year sera samples during convalescence were collected in 2017 and 2019,*** positive rate of DENVspecific immunoglobulin G was 98.4%in 2017,which significantly decreased to 82.8%in *** geometric mean titer(GMT)of NAb decreased from 1:155.35 to 1:*** 290 overall samples,73 paired consecutive samples were used for follow-up *** four-year sera,the GMTs of NAb against DENV-3 and cross-reactive antibodies against DENV-1,DENV-2 and DENV-4 were 1:167.70,1:13.80,1:18.54 and 1:45.26,respectively,which decreased to 1:53.18,1:10.30,1:14.60 and 1:8.17 in six-year *** age-stratified analysis,due to the increasing number of ADE positive samples from 2017 to 2019 in 31–40 and 51–60 years groups,the risk of ADE in DENV-4 infection was positively associated with the extension of convalescent phase,and the odd ratio was higher than other *** the recovery period lengthened,the risk of secondary infection with DENV-1 and DENV-2 was *** results offer essential experimental data for risk prediction of severe dengue in hyper-endemic dengue areas,and provide crucial scientific insight for the development of effective dengue vaccines.

Progress in active compounds effective on ulcerative colitis from Chinese medicines

Ulcerative colitis(UC), a chronic inflammatory disease affecting the colon, has a rising incidence worldwide. The known pathogenesis is multifactorial and involves genetic predisposition, epithelial barrier defects, dysregulated immune responses, and environmental factors. Nowadays, the drugs for UC include 5-aminosalicylic acid, steroids, and immunosuppressants. Long-term use of these drugs, however, may cause several side effects, such as hepatic and renal toxicity, drug resistance and allergic reactions. Moreover, the use of traditional Chinese medicine(TCM) in the treatment of UC shows significantly positive effects, low recurrence rate, few side effects and other obvious advantages. This paper summarizes several kinds of active compounds used in the experimental research of anti-UC effects extracted from TCM, mainly including flavonoids, acids, terpenoids, phenols, alkaloids, quinones, and bile acids from some animal medicines. It is found that the anti-UC activities are mainly focused on targeting inflammation or oxidative stress, which is associated with increasing the levels of anti-inflammatory cytokine(IL-4, IL-10, SOD), suppressing the levels of pro-inflammatory cytokines(TNF-α, IL-1β, IL-6, IL-8, IL-23, NF-κB, NO), reducing the activity of MPO, MDA, IFN-γ, and iNOS. This review may offer valuable reference for UC-related studies on the compounds from natural medicines.

Formulation development and evaluation of gastroretentive floating beads with Brucea javanica oil using ionotropic gelation technology

In the present study, a gastric retention floating system for Brucea javanica oil, composed of alginate and carrageenan, was prepared using ionotropic gelation. Parameters for floatability, drug load, encapsulation efficiency, bead morphology, in vitro release, and in vivo gastric retention were evaluated. The optimized formulation via Box–Behnken design consisted of 1.7% alginate(W/V), 1.02% carrageenan(W/V), 1.4% CaCO_3(W/V), and a gelling bath of pH 0.8. The alginate–carrageenan–Brucea javanica oil beads had a porous structure and exhibited up to 24 h of in vitro floatability with a load capacity of 45%–55% and an encapsulation efficiency of 70%–80%. A 6-h sustained release was observed in vitro. The beads had a prolonged gastric retention(> 60% at 6 h) in fasted rats, compared to non-floating beads(15% at 6 h), as measured by gamma scintigraphy with single-photon emission tomography/computed tomography(SPET/CT). In conclusion, the alginate–carrageenan–Brucea javanica oil system showed enhanced oil encapsulation efficiency, excellent floating and gastric retention abilities, and a favorable release behavior.

The fabrication of light-weight,highly impact-resistant,and energy-absorbent materials is urgently demanded in many facets of the society from body armor to aerospace engineering,especially under an extreme *** nanotubes(CNTs),one of the strongest and toughest materials ever found,also have good conductivity,chemical stability,and thermal stability,etc,making them a competitive candidate as building blocks to help achieve the above *** this work,a kind of CNT network was prepared by using chlorosulfonic acid(CSA)to release the internal stress of super-aligned carbon nanotube films(SA-CNTF)and dendritic polyamide amine(PAMAM)to further introduce multiple hydrogen bonds and interlocking *** fabricated bioinspired carbon nanotube network films(PAMAM@C-CNTF)have a high toughness of 45.97 MJ/m3,showing an increase of 420%compared to neat *** importantly,the anti-impact performance of the films(e.g.,with a maximum specific energy absorption of 1.40 MJ/kg under 80-100 m/s projectile impact)is superior to that of conventional protective materials from steel and Kevlar fiber,and also exceeds that of any other reported carbon-based *** hierarchical energy dissipation mechanism was further revealed through experiment and *** functions including intelligent heating/anti-icing,ultraviolet protection,as well as electromagnetic interference shielding properties make these network films have great potential in practical multi-protection applications,especially under an extreme environment.

Hypergravity experiments on multiphase media evolution

The gravitational field affects the evolution of multiphase media, such as rocks, soil, and alloy melts. Hypergravity increases the body force of matter, enhancing the driving force of the relative motion between substances with different densities and accelerating the evolution of multiphase media. Hypergravity experiments provide a new approach to exploring the motion of multiphase media and solving engineering problems. Hypergravity experiments have been conducted in different disciplines,such as materials science, geological science, and geotechnical engineering. However, the knowledge barriers between various research fields have caused the development of centrifuges/inflight devices and theoretical research on the mechanisms of matter in motion in hypergravity to lag behind the application of hypergravity experiments, limiting the progress in these *** article systematically summarizes and proposes the fundamentals of hypergravity experiments, while the scientific challenge of the nonlinear hypergravity effect induced by high hypergravity on multiphase media evolution is clarified. Evaluation criteria are proposed for the noninertial frame effects of the centrifugal hypergravity field. The development of the high-centrifugal acceleration, large-capacity, and long-beam centrifuges are determined as the future research direction. Representative cases are used to demonstrate the effectiveness and great potential of the hypergravity experiments for the solidification of alloy melts and physical modeling. Challenges in the experimental methodology are also clarified. This paper reviews the fundamentals and applications of hypergravity experiments in various disciplines, pointing out the research direction of hypergravity experiments on multiphase media evolution.

Impact of Pitx3 gene knockdown on glial cell line-derived neurotrophic factor transcriptional activity in dopaminergic neurons

Pitx3 is strongly associated with the phenotype, differentiation, and survival of dopaminergic neurons. The relationship between Pitx3 and glial cell line-derived neurotrophic factor（GDNF） in dopaminergic neurons remains poorly understood. The present investigation sought to construct and screen a lentivirus expression plasmid carrying a rat Pitx3 short hairpin（sh）RNA and to assess the impact of Pitx3 gene knockdown on GDNF transcriptional activity in MES23.5 dopaminergic neurons. Three pairs of interference sequences were designed and separately ligated into GV102 expression vectors. These recombinant plasmids were transfected into MES23.5 cells and western blot assays were performed to detect Pitx3 protein expression. Finally, the most effective Pitx3 sh RNA and a dual-luciferase reporter gene plasmid carrying the GDNF promoter region（GDNF-luciferase） were cotransfected into MES23.5 cells. Sequencing showed that the synthesized sequences were identical to the three Pitx3 interference sequences. Inverted fluorescence microscopy revealed that the lentivirus expression plasmids carrying Pitx3-sh RNA had 40-50% transfection efficiency. Western blot assay confirmed that the corresponding Pitx3 of the third knockdown sequence had the lowest expression level. Dual-luciferase reporter gene results showed that the GDNF transcriptional activity in dopaminergic cells cotransfected with both plasmids was decreased compared with those transfected with GDNF-luciferase alone. Together, the results showed that the designed Pitx3-sh RNA interference sequence decreased Pitx3 protein expression, which decreased GDNF transcriptional activity.

Central black hole mass determination for blazars

In this paper, we use a method to dctermine some basic parameters for the γ-ray loud blazars. The parameters include the central black mass （M）, the boosting factor （δ）, the propagation angle （Ф）, the distance along the axis to the site of the γ-ray production （d）. A sample including 32 γ-ray loud blazars with available variability time scales has been used to discuss the above properties. In this method, the γ-ray energy, the emission size and the property of the accretion disc determine the absorption effect. If we take the intrinsic γ-ray luminosity to be λ times the Eddington luminosity, i.e. Lγ^in =λLEdd, then we have the following results： the mass of the black hole is in the range of （0.59 - 67.99） ×10^7M⊙ （λ = 1.0） or （0.90 - 104.13） × 10^7M⊙ （λ = 0.1）; the boosting factor （5） in the range of 0.16 - 2.09（λ = 1.0） or 0.24 - 2.86 （λ = 0.1）; the angle （Ф） in the range of 9.53°- 73.85° （λ =1.0） or 7.36° - 68.89° （λ = 0.1）; and the distance （d/Rg） in the range of 22.39 - 609.36 （λ = 1.0） or 17.54 - 541.88 （λ = 0.1）.
