Coupled single-cell and bulk RNA-seq analysis reveals the engulfment role of endothelial cells in atherosclerosis

The clearance of apoptotic cell debris,containing professional phagocytosis and non-professional phagocytosis,is essential for maintaining the homeostasis of healthy ***,we discovered that endothelial cells could engulf apoptotic cell debris in atherosclerotic ***-cell RNA sequencing(RNA-seq)has revealed a unique endothelial cell subpopulation in atherosclerosis,which was strongly associated with vascular injury-related ***,integrated analysis of three vascular injury-related RNA-seq datasets showed that the expression of scavenger receptor class B type 1(SR-B1)was up-regulated and specifically enriched in the phagocytosis pathway under vascular injury ***-cell RNA-seq and bulk RNA-seq indicate that SR-B1 was highly expressed in a unique endothelial cell subpopulation of mouse aorta and strongly associated with the reorganization of cellular adherent junctions and cytoskeleton which were necessary for ***,SR-B1 was strongly required for endothelial cells to engulf apoptotic cell debris in atherosclerotic plaque of both mouse and human ***,this study demonstrated that apoptotic cell debris could be engulfed by endothelial cells through SR-B1 and associated with the reorganization of cellular adherent junctions and cytoskeleton.

Accurate mapping of loess waterworn gully(LWG)is essential to further study gully erosion and geomorphological evolution for the Chinese Loess Plateau(CLP).Due to the vertical joint and collapsibility of loess,LWGs have the characteristics of zigzag and unique slope abruptness under synthetic action of hydraulic force and *** forces existing LWG mapping methods to either focus on the improvement of mapping accuracy or center on the increase of mapping ***,simultaneously achieving accurate and efficient mapping of LWG is still in its infancy under complex topographic ***,we proposed a method that innovatively integrates the loess slope abruptness feature into an improved deep learning semantic segmentation framework for LWG mapping using 0.6 m Google imagery and 5 m DEM *** selected four study areas representing typical loess landforms to test the performance of our *** proposed method can achieve satisfactory mapping results,with the F1 score,mean Intersection-over-Union(mIoU),and overall accuracy of 90.5%,85.3%,and 92.3%,*** addition,the proposed model also showed significant accuracy improvement by inputting additional topographic information(especially the slope of slope).Compared with existing algorithms(Random forests,original DeepLabV3+,and Unet),the proposed approach in this study achieved a better accuracy-efficiency ***,the method can ensure high accuracy and efficiency of the LWG mapping for different loess landform types and can be extended to study various loess gully mapping and water and soil conservation.

We experimentally demonstrate tunable dual-comb soliton rains in a polarization multiplexing fiber laser based on a singlewalled carbon *** repetition frequency difference of dual-comb pulses is about 39 Hz,with a maximum extinction ratio of 29 *** suitable polarization states,one of the dual-comb pulses switches into soliton rain sequence with chirped isolating soliton *** signal-to-noise ratio reaches 61 dB,which is 11 dB higher than that of the normal dual-comb *** intervals between chirped isolating solitons are distributed progressively,and the number of isolating solitons can be flexibly tuned from 2 to 11 by adjusting polarization state or pump *** work will provide support for further understanding of interaction dynamics of solitons and give a new route to the application of precision measurement.

This paper presents a numerical study to improve the understanding of the complex subject of penetration and perforation of concrete targets impacted by low-velocity *** main focus is on the damage mechanisms and the major factors that account for the target resistance of the *** improved continuous surface cap model recently proposed was *** model was first equipped with element erosion criteria and was adequately validated by comparisons with ballistic *** numerical simulations were carried out where the individual influence of tensile,shear,and volumetric behaviors(pore collapse)of a concrete target on its ballistic performance was *** demonstrated that cratering on the front face and scabbing on the rear face of the concrete target were mainly dominated by its tensile *** major target resistance came from the second tunneling stage which was primarily governed by the shear and volumetric behaviors of the ***,this study captured the pore collapse-induced damage phenomenon during the high-pressure tunneling stage,which has been extensively reported in experiments but has usually been neglected in previous numerical investigations.

Research and Exploration of Ideological and Political Education in the Course of Pathophysiology

Course based ideological and political education (CIPE) is an important way to improve the quality of ideological and political work and talent cultivation. This study explores for the first time the implementation of ideological and political education in the teaching of pathophysiology courses, and also analyzes the evaluation of student psychological status and the importance of mental health education in the implementation of IPE courses. A survey was conducted on 211 students at Yangtze University to understand their motivation and behavior towards learning ideological, political, and pathophysiological courses. In addition, a questionnaire survey was used to explore the relationship between pathophysiology and ideological and political courses, as well as the importance of their satisfaction with the implementation of ideological and political courses in pathophysiology and mental health education. The research results indicate that factors such as educational background and gender differences affect the learning of CIPE. Graduate students are more interested in ideological and political courses, while female students find it difficult to study pathophysiology;In addition, the results of one-way ANOVA indicate that the implementation effect of IPE in pathophysiology courses depends on the level of interest in IPE and pathophysiology courses, the level of consideration for the importance of professional courses, the professional gains after studying pathophysiology, and the level of understanding of the relationship between IPE and CIPE. 81.04% of students believe that in the CIPE process, telling stories by teachers themselves is the most popular way of communication and education. This reflects the importance of mental health education from the perspective of CIPE. Objective Renal fibrosis is the ultimate pathway of various forms of acute and chronic kidney ***,the knockout of transient receptor potential channel 6(TRPC6)has shown promise in alleviating renal ***,the regulatory impact of TRPC6 on renal fibrosis remains *** In vivo,TRPC6 knockout(TRPC6−/−)mice and age-matched 129 SvEv(WT)mice underwent unilateral renal ischemia-reperfusion(uIR)injury surgery on the left renal pedicle or sham *** and serum were collected on days 7,14,21,and 28 after *** vitro,primary tubular epithelial cells(PTECs)were isolated from TRPC6−/−and WT mice,followed by treatment with transforming growth factorβ1(TGFβ1)for 72 *** anti-fibrotic effect of TRPC6−/−and the underlying mechanisms were assessed through hematoxylin-eosin staining,Masson staining,immunostaining,qRT-PCR,and Western *** Increased TRPC6 expression was observed in uIR mice and PTECs treated with TGFβ***6−/−alleviated renal fibrosis by reducing the expression of fibrotic markers(Col-1,α-SMA,and vimentin),as well as decreasing the apoptosis and inflammation of PTECs during fibrotic progression both in vivo and in ***,we found that the phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT)/glycogen synthase kinase 3 beta(GSK3β)signaling pathway,a pivotal player in renal fibrosis,was down-regulated following TRPC6 *** These results suggest that the ablation of TRPC6 may mitigate renal fibrosis by inhibiting the apoptosis and inflammation of PTECs through down-regulation of the PI3K/AKT/GSK3β*** TRPC6 could be a novel therapeutic strategy for preventing chronic kidney disease.
