Phytase is a kind of enzyme that hydrolyzes phytic acid and its salts to produce inositol and phosphoric acid. As a new feed additive, phytase has great potential in animal nutrition and environmental protection. Because of its good stability, large-scale production and high activity, microbial phytase has become a hot spot in industrial application. Here, we reported the predicted structure and enzymatic properties of a phytase from Bacillus subtilis, which was named as phyS. It was clear that the optimal temperature is 35°C, and the optimal pH is 8. Meanwhile, the enzyme activity was kept at above 90% in the range of pH 8 - 9, this result demonstrated that phyS is an alkaline phytase. This study lays a foundation for the extensive application of phyS.

[Objectives]This study was conducted to optimize the optimal process conditions for extracting polysaccharides from Rose laevigate *** hot water.[Methods]The extraction rate of *** was used as an evaluation index,and extraction temperature,material-to-liquid ratio,extraction time and extraction times were selected as influencing *** the basis of single factor tests,L 9(34)orthogonal tests were carried out for optimization,and the content of polysaccharides was determined by the phenol-sulfuric acid *** in vitro antioxidant activity of the extracted *** polysaccharides was studied.[Results]The effects of various influencing factors on the extraction rate of *** polysaccharides ranked as material-liquid ratio>extraction temperature>extraction times>extraction *** optimal extraction process was extraction temperature 80℃,extraction time 2 h,and material-to-liquid ratio 1∶25,extraction times 3 times,under which the extraction rate was(9.55±0.12)%.*** polysaccharides had certain scavenging capacities of DPPH free radicals and hydroxyl free radicals,and its scavenging capacity of DPPH free radicals was better than that of hydroxyl free radicals.[Conclusions]This study provides a theoretical basis for the optimization of the extraction process of *** polysaccharides and the in-depth development and application of polysaccharides.

Mass spectrometry (MS) stands as a pivotal tool for chemical analysis in the textile industry, capable of scrutinizing the vast majority of reported harmful chemicals. With the ever-increasing number of banned chemicals in textiles, there is a pressing need for a more systematic compilation of relevant literature on MS utilization to identify and quantify harmful substances in textiles. This article comprehensively reviews the role of MS in textile analysis over the past two decades, encompassing a wide range of sample preparation techniques, such as energy-assisted extraction, solid-phase microextraction, liquid-phase microextraction, and QuEChERS. Furthermore, it delves into MS techniques, such as gas chromatography–MS, liquid chromatography–MS, inductively coupled plasma–MS, ion mobility spectrometry–MS, and ambient ionization MS. Moreover, it offers a brief glimpse into the future of MS development in this field, highlighting ongoing research into additional sample preparation techniques for detecting hazardous chemicals in textiles. Notably, it underscores the growing interest in ambient ionization MS and the coupling of chromatography with high-resolution MS.

The impacts of ambiguity in preparation of 80% sulfuric acid solution and shaking time control of calibration solution on the determination of transparent exopolymer particles

The quantification of transparent exopolymer particles(TEP) by colorimetric method is of large error and low repeatability,one major reason of which is related to the absence of clear definition and evaluation for part steps of the original *** is obscure that the 80% sulfuric acid solution,acted as the extraction solution in the determination of TEP,is prepared based on a volume ratio or mass ***,the change of solubility of recently available Gum Xanthan(GX) from the market means that the original protocol is no longer applicable,and the grinding of GX stock solution with a tissue grinder is replaced by shaking with a rotating shaker in the study to prevent the excessive dissolution of *** found that different preparation techniques could result in the varied concentrations of 80% H_(2)SO_(4).The duration of shaking during the preparation of standard solution significantly affected the slope of the calibration curve,which caused different correction results of *** impacts of different extraction solution concentrations and shaking time of GX solution on the quantification of TEP were investigated based on the field sampling and laboratory *** extraction capacities of H_(2)SO_(4) with different concentrations for Alcian Blue were distinct,but had limited effect on the final measuring result of *** change of the standard curve slope came along with the variation of shaking time,which markedly altered the detection limit and calibration result,and the extended shaking time was in favor of the determination of low-concentration *** was suggested that the extraction solution concentration,shaking time and filtration volume of standard solution are required to be well controlled and selected to obtain more accurate results for TEP with different concentrations.

The aim of this work was to develop a liquid chromatography-tandem mass spectrometry method for the determination of milk allergen and egg allergen in food *** peptides GGLEPINFQTAADQAR,VGINYWLAHK,VLVLDTDYK,FFVAPFPEVFGK,and NAVPITPTLNR were confirmed and synthesized as the quantitative peptide of ovalbumin,α-lactalbumin,β-lactoglobulin,α_(S1)-casein andα_(S2)-casein,the relative isotope-labeled internal standards were used in the quantitative *** range was in the range of0.5-5000.0 nmol/L for egg and milk allergen in bread,cake,cookie,rice crust and wheat flour samples with free from egg and milk,the limits of detection of milk allergens and egg allergen were in the range between0.94 mg/100 g and 56.71 mg/100 g,limits of quantification of milk allergens and egg allergen were in the range between 2.36 mg/100 g and 141.78 mg/100 *** recoveries ranged from 76.7%to 122.8%,the relative standard deviations were in the range of 1.60%-15.60%.The developed method has been successfully used for the detection of egg and milk allergen in various food samples.
