Difference of the gut microbiota of premature ovarian insufficiency in two traditional Chinese syndromes

PURPOSE: To investigate the differences in gut microbial characteristics between two traditional Chinese syndromes of premature ovarian insufficiency(POI). METHODS: Forty women with POI were recruited from the Department of Traditional Chinese Medicine at Shenzhen maternity and Child Healthcare hospital between June and December 2020. Women with POI were divided into the kidney deficiency and blood stasis syndrome(SDBS) and Qi and blood deficiency syndrome(QBDS) groups. Gut microbial community profiles were analyzed by 16S rRNA gene sequencing using an Illumina Mi Seq system. A retrospective study comparing hormone levels and gut microbiota information was performed between the SDBS and QBDS groups. RESULTS: Compared with the QBDS group, the serum levels of estradiol(E2) and anti-Müllerian hormone(AMH) were significantly decreased in the SDBS group. The quantities of Adlercreutzia, Eggerthella, Klebsiella, and Paraprevotella significantly increased in the SDBS group, whereas Lactobacillus decreased significantly. Moreover, alterations in the microbiome in the SDBS and QBDS groups were closely related to the levels of E2 and AMH. The area under the receiver operating characteristic curve for the classification of the two syndromes by the gut microbiome was 0.71. CONCLUSIONS: There were significant differences in the dominant microbiota between the SDBS and QBDS groups, and the change in Proteobacteria in the QBDS group was more significant. The characteristics of gut microbiota help us differentiate between the SDBS and QBDS groups, which may provide a basis for the objectification of TCM syndrome types.

Electroacupuncture enhances the mitophagy of granulosa cells in premature ovarian insufficiency model mice by inactivating the hippo-yes-associated protein/transcriptional co-activator with postsynaptic density protein,drosophila disc large tumor suppresso

OBJECTIVE: To investigate the potential mechanism of electroacupuncture（EA） in alleviating premature ovarian insufficiency（POI） and to provide a theoretical basis for EA treatment of POI. METHODS: For this purpose, a POI mice model was developed by injecting 12 mg/kg busulfan and 120 mg/kg cyclophosphamide intraperitoneally to induce POI. It was then proceeded by EA intervention at Guanyuan（CV4） acupoint on the second day following modeling. Similarly, apoptosis in ovarian granulosa cells was detected by terminal deoxynucleotidyl transferase d UTP nick end labeling staining, while enzyme-linked immunosorbent assay was employed for measuring serum folliclestimulating hormone（FSH）, luteinizing hormone（LH）, estrogen（E2）, and anti-müllerian hormone（AMH） levels. Moreover, transmission electron microscopy（TEM） was employed for examining mitochondrial morphology, while autophagy and hippo-yes-associated protein/transcriptional co-activator with postsynaptic density protein, drosophila disc large tumor suppressor, and zonula occludens-1 protein binding motif（YAP/TAZ） pathway related protein levels in ovarian tissue were detected via Western blotting. RESULTS: Analysis of serum levels of various hormones indicated that serum FSH and LH were reduced in EA compared to the POI group, while E2and AMH levels were found to be elevated in EA compared to the POI group. The EA was found to inhibit apoptosis in granulosa cells in POI model mice, in addition to improved mitochondrial damage and significantly improved mitophagy. Pathway analysis revealed that EA was involved in activating the hippo-YAP/TAZ pathway, followed by reversing EA effects on granulosa cell apoptosis and mitophagy with the use of verteporfin, an autophagy and YAP-T-cell factor/enhancer of split and activator of transcription domain family member interaction inhibitor. Protein arginine methyltransferase-6 regulates heterogeneous nuclear ribonucleoprotein-F expression and is a potential target for the treatment of neuropathic pain

Colposcopy is an important technique in the diagnosis of cervical cancer. The development of computer-aided diagnosis methods can mitigate the shortage of colposcopists and improve the accuracy and efficiency of colposcopy examinations in China. This study proposes the Dense-U-Net model for colposcopy image recognition. This was a man–machine comparison cohort study. It presents a novel artificial intelligence (AI) model for the diagnosis of cervical lesions through colposcopy images using a Dense-U-Net image semantic segmentation algorithm. The Dense-U-Net model was created by applying the methods of “deepening the network structure,” “applying dropout” and “max pooling.” Moreover, image-based and population-based diagnostic performances of the AI algorithm and physicians with different levels of specialist experience were compared. In total, 2,475 participants were recruited, and 13,084 colposcopy images were included in this study. The diagnostic accuracy of the Dense-U-Net model increased significantly with increasing colposcopy images per patient. As the number of images in the training set increased, the diagnostic accuracy of the Dense-U-Net model for cervical intraepithelial neoplasm 3 or worse (CIN3+) diagnosis increased (P = 0.035). The rate of diagnostic accuracy (0.89 vs 0.85, P < 0.001) of CIN3+ lesions using the Dense-U-Net model was higher than that of expert colposcopists, and the missed diagnosis (0.06 vs 0.07, P = 0.002) and false positive (0.05 vs 0.08, P < 0.001) were lower. Moreover, Dense-U-Net is more accurate in diagnosing the type III cervical transformation zone, which is difficult to diagnose by experts (P < 0.001). The Dense-U-Net model also showed higher diagnostic accuracy for CIN3+ in an independent test set (P < 0.001). To diagnose the same 870 test images, the Dense-U-Net system took 1.76 ± 0.09 min, while the expert, senior, and junior colposcopists took 716.3 ± 49.76, 892.1 ± 92.30, and 3034.7 ± 259.51 min, respectively. Relationship between parenting stress and behavioral and emotional problems in preschool children: A mediation effect analysis

BACKGROUND Emotional reactions,such as anxiety,irritability,and aggressive behavior,have attracted clinical attention as behavioral and emotional problems in preschool-age *** To investigate the current status of family rearing,parental stress,and behavioral and emotional problems of preschool children and to analyze the mediating effect of the current status of family rearing on parental stress and behavioral/emo-tional *** We use convenience sampling to select 258 preschool children in the physical examination center of our hospital from October 2021 to September *** children and their parents were evaluated using a questionnaire ***'s correlation was used to analyze the correlation between child behavioral and emotional problems and parental stress and family rearing,and the structural equation model was constructed to test the mediating *** The score for behavioral/emotional problems of 258 preschool children was(27.54±3.63),the score for parental stress was(87.64±11.34),and the score for parental family rearing was(31.54±5.24).There was a positive correlation between the behavioral and emotional problems of the children and the“hostile/mandatory”parenting style;meanwhile,showed a negative correlation with the“support/participation”parenting style(all P<0.05).The intermediary effect value between the family upbringing of parents in parental stress and children's behavior problems was 29.89%.CONCLUSION Parental family upbringing has a mediating effect between parental stress and behavioral and emotional problems of *** paying attention to the behavioral and emotional problems of preschool-age children,clinical medical staff should provide correct and reasonable parenting advice to their parents to promote the mental health of preschool-age children.

Human fetal germ cells (FGCs), referring to the embryonic precursors of oocytes and sperm, are crucial for the transmission of genetic and epigenetic information across generations. The finely orchestrated development of FGCs is of great importance for maintaining reproductive health. In the past several years, various omics techniques have been applied to human FGCs to reveal the global epigenetic reprogramming and transcriptional resetting processes during FGC development. Single-cell RNA sequencing (RNAseq) analysis of human FGCs unraveled the dynamic transcriptome, with simultaneous expression of both pluripotency genes and germline-specific genes (Guo et al., 2015; Li et al., 2017).
