To formulate the optimal strategy of combatting bacterial biofilms,in this review we update current knowledge on the growing problem of biofilm formation and its resistance to antibiotics which has spurred the search for new strategies to deal with this *** on recent findings,the role of bacteriophages in the prevention and elimination of biofilm-related infections has been *** vitro,ex vivo and in vivo biofilm treatment models with single bacteriophages or phage cocktails have been compared.A combined use of bacteriophages with antibiotics in vitro or in vivo confirms earlier reports of the synergistic effect of these agents in improving biofilm ***,studies on the application of phage-derived lysins in vitro,ex vivo or in vivo against biofilm-related infections are *** strategy of combined use of phage and antibiotics seems to be different from using lysins and *** findings suggest that phages and lysins alone or in combination with antibiotics may be an efficient weapon against biofilm formation in vivo and ex vivo,which could be useful in formulating novel strategies to combat bacterial *** findings proved to be relevant in the prevention and destruction of biofilms occurring during urinary tract infections,orthopedic implant-related infections,periodontal and peri-implant *** conclusion,it appears that most efficient strategy of eliminating biofilms involves phages or lysins in combination with antibiotics,but the optimal scheme of their administration requires further studies.

Bacillus species have been widely used as probiotics in a number of food ***,these are vulnerable to bacteriophage infections,which poses fermentation failure and eventually result in economic *** this,we designed this study in which the bacteriophage of lysogenic Bacillus methylotrophicus,phage BM-P1,was induced successfully,and its biological characteristics and genome information were *** obtained results showed that phage BM-P1 belonged to Myoviridae *** maximum number of infections for this phage was 10,with a burst size of 104.48±2.70 counts expressed per milliter per infective ***,this phage was temperature-and pH-sensitive and divalent ions(Ca2+,Mg2+)and chloramphenicol did not have a significant influence on its adsorption *** analysis revealed that the genome size of phage BM-P1 was 153087 bp with 41.94%GC content,including 258 coding ***,52 bac-teriophages of Bacillus were classified into two clades by using phylogenetic *** them,phage BM-P1 with phage VB_BsuM-Goe3 and vB_BveM-Goe7 had the highest average nucleotide identity values,95.23%and 95.28%,***,synteny analysis revealed transposition,deletion,and insertion in the genome of *** this,it could be extrapolated that this phage is a new lysogenic *** conclusion,this study furthers the knowledge on the properties of *** phages and provides seminal insights for designingeffective antiphage strategiesforfermentation industry.

In order to study structure-function details of TGF-β1, the recombinant mature form of rat TGF-β1 was expressed in bacteria. Synthesis of the 112 amino-acid carboxyl-terminal part of TGF-β1 (amino acid 279-390) was controlled by an inducible gene expression system based on bacteriophage T7 RNA polymerase. This system allowed an active and selective synthesis of recombinant TGF-β1. The molecular weight of expressed TGF-β1 monomer determined on SDS-polyacrylamide gel under reducing conditions was about 13 kD. Serial detergent washes combined with a single gel-filtration purification step were sufficient to purify the expression product to homogeneity. Amino-terminal sequencing revealed that the N-terminal of the recombinant protein was identical to the published data. In Western blot analysis the recombinant polypeptide showed excellent antigenicity against polyclonal TGF-β1 antibody. The mature recombinant rat TGF-β1 expressed in this study provides a useful tool for future detailed structural and functional studies.

In order to develop a desirable inexpensive,effective and safe vaccine against the very virulentinfectious bursal disease virus （vvIBDV）,we tried to take advantage of the emerging T4 bacteriophagesurface protein display *** major immunogen protein VP2 from the vvlBDV strain HK46 wasfused to the nonessential T4 phage surface capsid protein,a small outer capsid （SOC） protein,resulting inthe 49 kDa SOC-VP2 fusion protein,which was verified by sodium dodecylsulfate polyacrylamide gelelectrophoresis and Western *** showed that the recombinant VP2 protein wassuccessfully displayed on the surface of the T4 *** recombinant VP2 protein is antigenic and showedreactivities to various monoclonal antibodies （mAbs） against IBDV,whereas the wild-type phage T4 couldnot react to any *** addition,the recombinant VP2 protein is immunogenic and elicited specific antibodiesin immunized specific pathogen free （SPF） *** significantly,immunization of SPF chickenswith the recombinant T4-VP2 phage protected them from infection by the vvIBDV strain *** with the vvIBDV strain HK46 at a dose of 100 of 50% lethal dose (LD50) per chicken 4 weeksafter the booster was given,the group vaccinated with the T4-VP2 recombinant phage showed no clinicalsigns of disease or death,whereas the unvaccinated group and the group vaccinated with the wild-type T4phage exhibited 100% clinical signs of disease and bursal damages,and 30%—40% ***,the data herein showed that the T4-displayed VP2 protein might be an inexpensive,effective and safe vaccinecandidate against vvIBDV.

Phage display technique provides a powerful approach for the discovery of new tumor-specific ***,the peptides isolated through this technique usually did not possess high tumor-specific property.A pre-clearing step was introduced to increase the efficiency of biopanning by removal of particles that could interact with ubiquitously expressed cellular receptors in the non-target *** randomized Ph.D-CX7C phage library (Phage III) was first pre-cleared in normal mice to reduce vasculatureor organ-targeting phages to get the pre-cleared phage library,and then the tumor-targeting bacteriophage particles (Phage I) were screened from pre-clearing phage library in S180 tumor-bearing *** biodistribution results of 99mTc-labeled phages in mice bearing S180 tumor show that the uptake of 99mTc-labeled Phage I in tumor is high but low in normal organs,and the tumor-to-liver and tumor-to-spleen ratios of 99mTc-labeled Phage I are higher than those of 99mTc-labeled Phage II (tumor-specific phages screened from the original CX7C library) and Phage III (unscreened phages from the original CX7C library).It indicates that the yield of tumor-targeting bacteriophage particles could be improved and the non-specific binding in organs becomes ***,the pre-clearing phage display method could improve the yield of positive hits by reducing the non-target organ accumulation of bacteriophage particles.

Background:Antibiotic growth promoters are widely used to improve weight ***,the abuse of antibiotics can have many negative effects on *** alternatives to antibiotics is an urgent need in livestock *** aimed to perform a meta-analysis and network meta-analysis(NMA)to investigate the effects of feed additives as potential antibiotic substitutes(ASs)on bacteriostasis,growth performance,intestinal morphology and ***,the primary,secondary,and tertiary ASs were defined by comparing their results with the results of ***:Among 16,309 identified studies,37 were summarized to study the bacteriostasis effects of feed additives,and 89 were included in the meta-analysis and NMA(10,228 pigs).We summarized 268 associations of 57 interventions with 32 *** order of bacteriostasis effects was as follows:antimicrobial peptides(AMPs)≈antibiotics>organic acids>plant extracts>*** detected associations of 11 feed additives and 11 *** with a basal diet,plant extract,AMPs,probiotics,microelements,organic acids,bacteriophages,lysozyme,zymin,and oligosaccharides significantly improved growth performance(Pbacteriophage,lysozyme,microelements,oligosaccharides,organic acids,plants,plant extracts,probiotics,and zymin doses were 0.100%,0.150%,0.012%,0.010%,0.050%,0.750%,0.20%,0.040%,0.180%,and 0.100%,*** with antibiotics,all investigated feed additives exhibited no significant difference in effects on growth performance,IgG,and diarrhoea index/rate(P>0.05);AMPs and microelements significantly increased V/C(P<0.05);and zymin significantly improved lymphocyte levels(P<0.05).Furthermore,linear weighting sum models were used to com

A kinetic model of the interactions between operators and regulators is developed to study the stabilities of genetic states and lysogeny/lysis switch in Eschemchia coli infected by bacteriophage lambda. Using adiabatic approximation, the kinetic evolutions of mRNA and regulator concentrations can be deduced from operators＇ equations. Furthermore, the stability of each state of the system is studied. The results show that the lysogenic state switches to the lytic state through two bifurcations： one from a single stable state to a three-point state, and the other from a three-point state to a single stable state. Then we indicate that the property of the lysogeny/lysis switch satisfies the topological characteristics theorem. Finally, the influence of the left operators on the lysogeny/lysis switch is briefly discussed. The results show that the cooperativity of the CI2 bound to left and right operators makes the lysogenic state more stable.
