White Spot Syndrome Virus (WSSV) was purified from hemolymph of infected shrimp. After nucleic acid extraction from the purified virus particles, EcoR I digested fragments of the WSSV genome were cloned; three of these fragments were used as non radioactive probes labeled with DIG 11 dUTP. The probes hybridized in situ, with sections located in the nuclei of all WSSV infected tissues. The virus was detected in the gill, stomach, epidermis, and connective tissue and so on, but not detected in healthy shrimp tissues and epithelial cells of hepatopancreatic tubules of diseased shrimp.

Using alternative plant-derived dietary protein to replace fishmeal,combined with practical evaluation indexes,is a recent focus for aquaculture *** 8-week feeding experiment with giant freshwater prawn Macrobrachium rosenbergii post-larvae was conducted to determine the eff ects of replacing fi shmeal(FM)with soybean meal in the feed,in terms of growth performance,antioxidant capacity,intestinal microbiota,and mRNA expression of target of rapamycin(TOR)and ribosomal protein S6 kinase B1(S6K1).Four isonitrogenous diets with isocaloric value were prepared to contain 100%,75%,50%,or 25%FM as the protein source(dietary treatments FM100,FM75,FM50,and FM25,respectively).Each diet was fed to post-larval prawns(mean weight 0.045±0.002 g)twice a day in four *** signifi cant diff erence in weight gain was observed among all groups,but the survival rate of prawns fed the FM50 and FM25 diets was signifi cantly lower than that of prawns fed the FM *** mRNA expression of both TOR and S6K1 were the lowest in hepatopancreas of prawns fed the FM25 *** dismutase activity of prawns fed the FM25 diet was significantly lower than that of prawns fed *** contrast,the malondialdehyde content was signifi cantly higher in prawns fed FM25 as compared with those fed *** proportion of fishmeal in the diet did not affect the composition of core(phylum-level)intestinal microbiota,but greater fishmeal replacement with soybean meal had a potential risk to increase the relative abundance of opportunistic pathogens in the gut when considered at the genus *** results suggest that fishmeal replacement with soybean meal should not exceed 50%in a diet for post-larval ***.

A histological and ultrastructural study was performed on the development of the head kidney, thymus, and spleen in Pseudosciaena crocea aging in range from just hatching to the 60th day after hatching （DAH）. Head kidney was first present on the 3rd DAH. Primordial haemopoietic stem cells were first observed in the head kidney which rapidly differentiate into different cellular types. Progenitor spleen was present on the 4th DAH, located close to the gut, which soon became rich in blood capillaries, red blood cells and thrombocytes. The thymus was obvious on the 4th DAH, and was located on either side of the upper comer of the opercular cavity, closely under the membrance of the opercuhr cavity. The thymus was the last lymphoid organ to appear but showed a quick development. This organ seemed to originate from haemopoietic stem cells migrating from the head region of the kidney. The thymus consisted of outer thymocytic and inner epithelioid zones. There was no obvious demarcation between them, but both zones were visible. Small lymphocytes appeared, or lymphoid organs become lymphoid, in the sequence thymus, head kidney and spleen. Only a small number of lymphocytes appeared in the later stages. Non-specific systems may play an important role in the immunocompetence mechanisms of P. crocea during its early development stage.

The morphological similarities of Pampus fishes have led to considerable confusion in species-level identification,and no accurate information on neotype or DNA barcoding of Pampus echinogaster is available. Two hundred and seven specimens of P. echinogaster were collected from the coastal waters of Dandong, Dongying, Qingdao,Nantong, Zhoushan, Wenzhou, Changle, Taiwan, and Wakayama（Japan）, from June 2010 to April 2013. The diagnostic characteristics of P. echinogaster are as follows： dorsal fin VIII-XI-43–51, anal fin V-VIII-43–49, pectoral fin 22–27, caudal fin 19–22, pelvic fin absent; first gill rakers sparse, slender（pointed）, 3–4＋12–16=15–20; vertebrae39–41; transverse occipital canal on top of head moderately small, wavy ridges not reaching upper origin of pectoral fin; ventral branch of lateral line canal spare, shorter than dorsal branch of lateral line canal. By combining congener sequences of the cytochrome oxidase I（COI） gene from Gen Bank, two absolute groups were detected among all specimens, which further indicated that two valid species were present based on genetic differences in amino acid sequences and the distance between the groups. The sequences of Group 1 can be regarded as DNA barcoding of P. echinogaster. The correct morphological redescription and DNA barcoding of *** are presented here to provide a guarantee for efficient and accurate studies, a theoretical basis for classification, and enable appropriate fishery management and conservation strategies for the genus Pampus in the future.

Simplified Approximate Expressions for the Boundary Layer Flow in Cylindrical Sections in Plankton Nets and Trawls

Trawls and plankton nets are basically made up of conical and cylindrical net sections. In conical sections the flow will pass through the inclined net wall with a noticeable angle of attack, and then the flow, filtration and drag can be suitably modelled e.g. by a pressure drop approach [1]. In cylindrical and other non-tapered net sections, such as foreparts and extension pieces in trawls and plankton nets, the flow is directed along the net wall and is best considered in terms of a boundary layer. Boundary layer theory and turbulence models can be used to describe such flow, but this requires extensive numerical modelling and computational effort. Simplified approximate formulas providing a qualitative description of the flow with some quantitative accuracy are therefore also useful. This work presents simplified parametric expressions for boundary layer flow in cylindrical net sections, including the boundary layer thickness and growth rate along the net, the filtration velocity out of the net wall, the decrease in mass flux through the net due to the growing boundary layer, and the effect of twine thickness, flow (towing) velocity and the dimensions of the net. These expressions may be useful for assessing the existence and extension of a boundary layer, for appropriate scaling of boundary layer effects in model tests, for proper placement of velocity measurement probes, for assessing the influence on filtration and clogging of plankton net sections, and more.

An 8-week feeding trial was conducted to determine the optimal dietary protein level for juvenile marbled *** semi-purified test diets were formulated to contain different protein levels(CP)including 42.7%,47.4%,53.3%,58.8%,and 64.5%(dry matter),named as CP42.7,CP47.4,CP53.3,CP58.8,and CP64.5,*** hundred and twenty-five juveniles(6.0±0.1 g)were randomly distributed into 15 tanks(300 L tanks),resulting in 35 fish per tank(n-3 tanks).Fish were fed the test diets 5 times per day until *** CP58.8 resulted in the highest gain in weight and the best efficiency in feed utilization among the tested protein levels(P<0.05).Fish fed the CP58.8 diet showed significantly higher whole-body protein and lipid contents than the fish that were fed the other diets(P<0.05).Fish fed the CP53.3,CP58.8,and CP64.5 diets showed a significantly higher dorsal-muscle lipid content than the fish that were fed the CP42.7 and CP47.4 diets(P<0.05).The one-slope straight broken-line regression analysis on the results of the thermal growth coefficient and feed conversion ratio indicated that the estimated optimum dietary protein level was 58.8%.Taken together,it is suggested that the dietary protein level of 58.8%is optimal for better growth and high efficiency in feed utilization for the juvenile marbled flounder.

The study investigated the genetic variation of Parachanna obscura from five rivers(Anambra,Ibbi,Imo,Katsina-Ala and Ogun)in Nigeria using the mitochondrial cytochrome oxidase 1 *** was extracted from 19,22,16,18 and 21 fin clips per river population,respectively and subjected to polymerase chain reaction.A total of 96 sequences,each with 671 bp were obtained with 38(5.6%)polymorphic,27(3.8%)parsimoniously informative and 659(98.2%)conserved *** nucleotide composition was C=28.07%,T=29.43%,A=22.18%,G=20.32%.A total of 40 haplotypes with 38 unique sequences as well as 24 substitutions with 22 transversions and two transitions were *** diversity among populations ranged from 0.00184 to 0.00888 representing Ibbi and Imo,respectively while haplotype diversity ranged from 0.77056 to 0.95000 also,from Ibbi and Imo,*** of molecular variance showed that the intra-population variation accounted for 50.05%.Topology from phylogenetic analyses revealed that *** from Imo River was distinctly different from the rest.

Neuropeptide Y(NPY) has a pivotal role in the regulation of many physiological processes. In this study, the gene encoding a NPY receptor-like from the common Chinese cuttlefish Sepiella japonica( SjNPYR-like) was identified and characterized. The full-length SjNPYR-like cDNA was cloned containing a 492-bp of 5′ untranslated region(UTR), 1 182 bp open reading frame(ORF) encoding a protein of 393 amino acid residues, and 228 bp of 3′ UTR. The putative protein was predicted to have a molecular weight of 45.54 kDa and an isoelectric point(pI) of 8.13. By informatic analyses, SjNPYR-like was identified as belonging to the class A G protein coupled receptor(GPCR) family(the rhodopsin-type). The amino acid sequence contained 12 potential phosphorylation sites and five predicted N-linked glycosylation sites. Multiple sequence alignment and 3D structure modeling were conducted to clarify SjNPYR bioinformatics characteristics. Phylogenetic analysis identifies it as an NPYR with identity of 33% to Lymnaea stagnalis NPFR. Transmembrane properties of SjNPYR-like were demonstrated in vitro using HEK293 cells and the p EGFP-N1 plasmid. Relative quantifi cation of SjNPYR-like mRNA level confi rmed a high level expression and broad distribution of SjNPYR-like in various tissues of female S. japonica. In addition, the transcriptional profile of SjNPYR-like in the brain, liver, and ovary during gonadal development was analyzed. The results provide basic understanding on the molecular characteristics of SjNPYR-like and its potentially physical functions.
