Population Genetic Analysis of Sillago nigrofasciata (Perciformes:Sillaginidae) Along the Coast of China by Sequencing Mitochondrial DNA Control Region

Sillago nigrofasciata, a small to moderate size nearshore species, is newly found along the eastern and southern coasts of China. The present study is carried out in order to analyze the population genetics of the S. nigrofasciata. The control region sequence of mitochondrial DNA revealing 73 haplotypes were obtained from 162 individuals collected at 8 locations along the coast of China. The whole S. nigrofasciata population along the coast of China showed a low nucleotide diversity(0.012) and a high population diversity(haplotype diversity)(0.943), and all the 8 local populations showed low nucleotide diversities(0.014 – 0.001), suggesting the protective measures are effective. The haplotypes of the 8 local populations were widely distributed in haplotype network diagram and neighbor-joining phylogenetic tree, while no branch associating with sampling locations was detected. Recent gene flow analysis showed asymmetric gene exchanges among local populations. The pairwise FST values and unweighted pair-group method with arithmetic mean(UPGMA) tree revealed a certain amount of genetic difference among local populations. Moreover, analysis of molecular variance(AMOVA) reflected genetic differences between hypothetical subdivision groups. Neutral test and mismatch distribution of pairwise nucleotide suggested S. nigrofasciata may have experienced recent population expansion events. The historical geographic events associating with ice age may be the main explanation to the heterogeneity among local populations with short geographic distances, and the homogeneity among local populations with long geographic distances.

The present work documented two snapper species, Lutjanus fulvus Forster, 1801 and Lutjanus erythropterus Bloch, 1790, recently collected from the Saint Martin's Island, Bay of Bengal, Bangladesh, during a survey on coral associated fishes of Bangladesh from July 2017 to September 2018. These two species were identified with morphological characteristics and by analyzing partial mitochondrial COI sequences. The document also included an updated checklist of snappers available in Bangladesh.

Transcriptional responses of mu-, pi- and omega-class glutathione S-transferase genes in the hepatopancreas of Cipangopaludina cahayensis exposed to microcystin-LR

The glutathione S-transferases(GSTs)play important roles in detoxification of microcystins(MCs),but the responses of GSTs to MC-LR have not been well characterized in freshwater gastropod,Cipangopaludina *** the present study,we cloned full-length cDNAs of mu-and pi-class GSTs(GSTM and GSTP)and partial cDNA of omega-class GST(GSTO),and determined the transcriptional responses of the three GST genes to different concentrations of MC-LR(0,1,10 and 100 lg/L)in *** full-length cDNAs of GSTM and GSTP of *** were 813 and 820 bp,containing an open reading frame(ORF)of 648 bp(encoding 215 amino acids)and 624 bp(encoding 207 amino acids),*** mRNA expression of GSTM and GSTO significantly decreased after exposure to 10 lg/L MC-LR,and the mRNA expression of GSTP significantly decreased after100 lg/L MC-LR *** might contribute to the detoxication of MCs in ***,which is consistent with its sedentary life and filter-feeder *** mRNA expression of the three GST isoforms in *** could be used as biomarkers for water contamination.

Challenges in the implementation of food safety and quality assurance systems in small-scale fisheries

Ninety percent of the world’s fishers are directly involved in small-scale fisheries(SSFs),and many food safety and quality issues along the food value chains may be both irreversible and *** safe,high-quality aquatic products are key to sustaining life,promoting good health,and avoiding adverse health *** This paper aimed to highlight factors preventing the implementation of safety and quality assurance systems in SSFs by analysing the results from surveys on the implementation of Article 11 of the Food and Agriculture Organization of the united Nations(FAO)Code of Conduct for Responsible fisheries(2017,2019,and 2022).Materials and methods The analysis considered responses to 15 closed-ended questions on food safety and quality obtained from 143 FAO Members and the European Union,and 87 open-ended replies addressing challenges in *** The major factors preventing the implementation of the Code’s food safety and quality provisions were lack of sanitation and monitoring programmes,lack of infrastructure(particularly refrigeration facilities)lack of technical capacities and limited management of food safety risks along the value *** and quality assurance efforts were focused on international markets,resulting in less attention being paid to domestic markets where a high degree of informality was *** Improved implementation of safety and quality assurance systems will lead to safer food with higher nutritional quality,extended shelf-life and reduced post-harvest *** awareness of food safety issues in SSFs is a prerequisite for achieving several of the UN Sustainable Development Goals.

Ingestion of Brachionus plicatilis under different microalgae conditions

The effects of four microalgae, Chlorella vulgaris, Platymonas helgolandicavar, Isochrysis galbana, and Nitzschia closterium on the grazing and filtering rates of the marine rotifer, Brachionus plicatilis, were evaluated under laboratory conditions. The grazing rates in separate cultures of the four microalga were as follows： C. vulgaris 〉 P. helgolandicavar 〉 L galbana 〉 N. closterium. However, the filtering rates occurred in the following order： P. helgolandicavar 〉 N. closterium 〉 C. vulgaris 〉 L galbana. A mixed diets experiment revealed that P. helgolandicavar was the preferred diet of B. plicatilis. In addition, the grazing rate of B. plicatilis increased gradually as the density of the microalgae increased, until concentrations of 2.5×10^6 cells mL^-1 for C. vulgaris and 1.5×10^6 cells mL^-1 for I. galbana were obtained. Furthermore, the filtering rate increased slightly when the density of the microalgae was low, after which it declined as the microalgal density increased. The grazing rates of B. plicatilis were as follows during the different growth phases： stationary phase 〉 exponential phase 〉 lag phase 〉 decline phase. Additionally, the filtering rates during the growth phases were： exponential phase 〉 lag phase 〉 stationary phase 〉 decline phase. The results of this study provide foundational information that can be used to explore the optimal culture conditions for rotifers and to promote the development of aquaculture.

Profile and development of microsatellite primers for Acanthogobius ommaturus based on high-throughput sequencing technology

Water quality, habitat, and fish assemblage relationships in middle-order agriculture and forest streams of the Mississippi Alluvial Plain

Background Agriculture has greatly influenced water quality, habitats, and fish assemblages in streams of the Mississippi Alluvial Plain(MAP) ecoregion. However, MAP streams have historically been understudied compared to streams in other agricultural regions of the USA. In this study, water quality, habitat, and fish assemblage composition were assessed seasonally(spring, summer, and fall) in eight representative MAP streams located across three U.S. states. The study design included four streams containing highly agricultural watersheds(herein termed “agriculture” streams) and four streams containing mostly forested watersheds(herein termed “forest” streams), which were intended to represent reference conditions for MAP *** In general, forest streams contained significantly better instream and riparian habitats than agriculture streams(P = 0.010–0.040) whereas agriculture streams contained significantly greater levels of primary nutrients(P < 0.001–0.010). Differences between agriculture and forest streams with respect to other physical and chemical variables were intermittent and season dependent. Fish assemblages in agriculture and forest streams were structured primarily along an environmental gradient reflecting instream habitat conditions, water nutrient concentrations, and benthic chlorophyll-a production. Structurally, fish assemblages in both stream types contained many regionally common species, though some species appeared to exhibit affinities for a particular stream type. Functionally, fish assemblages in agriculture streams contained more tolerant species, more omnivores, and fewer insectivores compared to forest stream assemblages, which were nearly all insectivores. The teleost Scatophagus argus is a species whose females grows faster than *** hormone(gh)mRNA abundance in females pituitary is higher than that in males;however the mechanism underlining such differential is still *** hormone(GH)is tightly associated with GH-releasing hormone(Ghrh)in *** this study,Ghrh gene(ghrh)and its receptor gene,ghrhr,were isolated from *** expression analysis showed that ghrh and ghrhr were mainly expressed in hypothalamus while ghrhr was expressed in pituitary and gh was predominantly expressed in *** cultured *** individuals were used to compare ghrh,ghrhr and gh mRNA abundances,120 g and 181 g average weight for male(n=11)and female(n=9),***-time PCR indicated that the ghrh and ghrhr mRNA abundances in male hypothalamus were significantly higher than those in female hypothalamus while that of gh mRNA abundance was significantly higher in female pituitary than in male *** ghrh and ghrhr mRNA abundances were significantly up-regulated in female hypothalamus 3 h after injection of 0.1 mg kg^-1 body weight Ghrh while pituitary ghrhr and gh mRNA abundances were not *** female hypothalamus,ghrh and ghrhr m RNA abundances were not affected at 6 h post-injection of 4 mg kg^-1 body weight 17α-methyltes-tosterone(17α-MT)or 17β-Estradiol(E2).In female pituitary,ghrhr m RNA abundance was down-regulated by 17α-MT while that of gh m RNA abundance was up-regulated by *** findings indicated that E2,rather than Ghrh,plays an important role in up-regulating the expression of gh in female ***,which should aid to understand the sexual dimorphism of teleost growth.

Starvation effects on pathogenic Vibrio alginolyticus in natural seawater

To get a better understanding of the starvation survival strategy of pathogenic Vibrio alginolyticus, log-phase cells were inoculated into sterile natural seawater for starvation studies. The results showed that all of total bacteria number, viable bacteria number and CFU number of V. alginolyticus increased remarkably at the initial starvation stage; after reaching their peaks at 5 d, both total bacteria number and viable bacteria number of V. alginolyticus fell slowly, while the CFU number fell more quickly after reaching its peak at 10 d; V. alginolyticus elongated their cells at the prophase of starvation, and then shrunk their volume and turned their shapes into ovals from rods at the anaphase of starvation; starved cells showed more sensitivity to heating and UV; starved cells showed no significant difference from unstarved ones at the lowest detection limit determined by indirect enzyme-linked immu- nosorbent assay （ELISA） ; starved cells＇ ability to adhere to the skin mucus of large yellow croaker （ Pseudosciaena crocea） showed a sharp decline as the starvation time increases; the cellular protein of V. alginolyticus increased remarkably at the ariaphase of starvation. The results indicated that pathogenic V. alginolyticus could survive in starvation for relatively long periods of time （ ≥2 months） in 28℃ natural seawater due to the morphological and physiological changes; however, starved V. alginolyticus cells showed less virulence and higher sensitivity under environmental stresses.

Growth and Resource Accumulation of Drifting Sargassum horneri(Fucales, Phaeophyta) in Response to Temperature and Nitrogen Supply

The abnormal increase of drifting brown alga Sargassum horneri was initially documented in *** formed blooms along the coast of East China Sea and Yellow Sea in *** this study,we investigated the changes of specific growth rate and resource accumulation of drifting *** in response to temperature and nitrogen richness at different growth stages under laboratory *** investigation lasted from June 2015 to April 2016 with the observation made every two *** results showed that the life cycle consists of a few growth stages dividable with morphological *** growth can be divided into shedding and withering(August),rapid growing(October to September),slow growing(February),rapid growing(April)and maturation(June)*** the experimental condition,algal segments were found to grow at temperatures ranging from 5 to 25℃in 12 days even when nitrogen is deficient.A significant difference in the special growth rate(SGR)between nitrogen-enriched and nitrogen-removed treatments was found in most months(P<0.05).SGR was lower in August and February than that in other *** and chlorophyll contents in algal segments were different among different temperatures,nitrogen supply and *** content was higher in February and April than that in other months in both nitrogen-enriched and nitrogen-removed *** results showed that the demand of *** for nitrogen increased in spring when it grows *** is likely that the high temperature and nitrogen concentration in winter and spring lead to the high biomass accumulation of drifting ***.
