circRNA3669 promotes goat endometrial epithelial cells proliferation via miR-26a/RCN2 to activate PI3K/AKT-mTOR and MAPK pathways

The development of receptive endometrium(RE) from pre-receptive endometrium(PE) for successful embryo implantation is a complex dynamic process in which the morphology and physiological states of the endometrial epithelium undergo a series of significant changes, including cell proliferation and apoptosis. However, the molecular mechanisms are not yet fully understood. In this study, a higher circRNA3669 level was observed in PE than in RE of goats. Functional assays revealed that this overexpression promoted the proliferation of goat endometrial epithelial cells(GEECs) by activating the expression of genes related to the PI3K/AKT-mTOR and MAPK pathways,thereby inhibiting apoptosis in vitro. Furthermore, circRNA3669 functioned as a competing endogenous RNA(ceRNA) to upregulate Reticulocalbin-2(RCN2) expression at the post-transcriptional level by interacting with and downregulating miR-26a in GEECs. In addition, RCN2, which is highly expressed in the PE of goats, was found to be regulated by β-estradiol(E2) and progesterone(P4). Our results demonstrated that RCN2 also affected the key proteins PI3K, AKT, mTOR, JNK, and P38 in the PI3K/AKT-mTOR and MAPK pathways, thereby facilitating GEECs proliferation and suppressing their apoptosis in vitro. Collectively, we constructed a new circRNA3669-miR-26aRCN2 regulatory network in GEECs, which further provides strong evidence that circRNA could potentially play a crucial regulatory role in the development of RE in goats.

Honeybees are the most critical pollinators providing key ecosystem services that underpin crop production and sustainable *** a backdrop of rapid global change,this eusocial insect encounters a succession of stressors during nesting,foraging,and *** mites,together with vectored viruses,have been recognized as central biotic threats to honeybee health,while the spread of invasive giant hornets and small hive beetles also increasingly threatens colonies *** of agrochemicals,including acaricides used for mite treatment,and other pollutants of the environment have been widely documented to affect bee health in various ***,expanding urbanization,climate change,and agricultural intensification often result in the destruction or fragmentation of flower-rich bee *** anthropogenic pressures exerted by beekeeping management practices affect the natural selection and evolution of honeybees,and colony translocations facilitate alien species invasion and disease *** this review,the multiple biotic and abiotic threats and their interactions that potentially undermine bee colony health are discussed,while taking into consideration the sensitivity,large foraging area,dense network among related nestmates,and social behaviors of honeybees.

Mathematical modeling of anaerobic digestion is a powerful tool to predict gas yields and optimize the *** Anaerobic Digestion Model No.1(ADM1)is a widely implemented model for this ***,modeling full-scale biogas plants is challenging due to the extensive substrate and parameter characterization *** study describes the modification of the ADM1 through a simplification of individual process phases,characteristic components and required ***,the ability of the simplified model to simulate the co-digestion of grass silage and cattle slurry was evaluated using data from a full-scale biogas *** impacts of substrate composition(crude carbohydrate,protein and lipid concentration)and variability of carbohydrate degradability on simulation results were assessed to identify the most influential *** indicated that the simplified version was able to depict biogas and biomethane production with average model efficiencies,according to the Nash-Sutcliffe efficiency(NSE)coefficient,of 0.70 and 0.67,respectively,and was comparable to the original ADM1(average model efficiencies of 0.71 and 0.63,respectively).The variability of crude carbohydrate,protein and lipid concentration did not significantly impact biogas and biomethane output for the data sets *** contrast,carbohydrate degradability seemed to explain much more of the variability in the biogas and methane ***,the application of simplified models provides a reliable basis for the process simulation and optimization of full-scale agricultural biogas plants.

Geographical patterns and determinants of insect biodiversity in China

Insects play important roles in the maintenance of ecosystem functioning and the provision of livelihoods for millions of ***,compared with terrestrial vertebrates and angiosperms,such as the giant panda,crested ibis,and the metasequoia,insect conservation has not attracted enough attention,and a basic understanding of the geographical biodiversity patterns for major components of insects in China is ***,we investigated the geographical distribution of insect biodiversity across multiple dimensions(taxonomic,genetic,and phylogenetic diversity)based on the spatial distribution and molecular DNA sequencing data of *** analysis included 18 orders,360 families,5,275 genera,and 14,115 species of *** results revealed that Southwestern and Southeastern China harbored higher insect biodiversity and numerous older lineages,representing a museum,whereas regions located in Northwestern China harbored lower insect biodiversity and younger lineages,serving as an evolutionary *** also observed that mean annual temperature and precipitation had significantly positive effects,whereas altitude had significantly negative effects on insect biodiversity in most ***,cultivated vegetation harbored the highest insect taxonomic and phylogenetic diversity,and needleleaf and broadleaf mixed forests harbored the highest insect genetic *** results indicated that human activities may positively contribute to insect spatial diversity on a regional *** study fills a knowledge gap in insect spatial diversity in *** findings could help guide national-level conservation plans and the post-2020 biodiversity conservation framework.

Impacts of agri-food e-commerce on traditional wholesale industry:Evidence from China

Rapidly expanding studies investigate the effects of e-commerce on company operations in the retail ***,the interaction between agri-food e-commerce(AEC)and the traditional agri-food wholesale industry(AWI)has not received enough attention in the existing *** on the provincial panel data from 2013 to 2020 in China,this paper examines the effect of AEC on AWI,comprising three dimensions:digitalization(DIGITAL),agrifood e-commerce infrastructure and supporting services(AECI),and agri-food e-commerce economy(AECE).First,AWI and AEC are measured using an entropy-based combination of *** results indicate that for China as a whole,AWI has remained practically unchanged,whereas AEC exhibits a significant rising ***,the findings of the fixed-effect regression reveal that DIGITAL and AECE tend to raise AWI,whereas AECI negatively affects ***,threshold regression results indicate that AECI tends to diminish AWI with three-stage inhibitory intensity,which manifests as a first increase and then a drop in the inhibition *** results suggest that with the introduction of e-commerce for agricultural product circulation,digital development will have catfish effects that tend to stimulate the vitality of the conventional wholesale industry and promote technical ***,the traditional wholesale industry benefits financially from e-commerce even while it diverts part of the traditional wholesale circulation for agricultural products.

Combined effects of climate and land-use changes on the alpha and beta functional diversities of terrestrial mammals in China

Climate and land-use changes are predicted to impact biodiversity,threatening ecosystem services and ***,their combined effects on the functional diversity of mammals at the regional scale remain unclear,especially at the beta ***,we use projected climate and land-use changes in China to investigate their potential effects on the alpha and beta functional diversities of terrestrial mammals under low-and high-emission *** the current projection,we showed strong positive spatial correlations between functional richness and species *** evenness(FEve),functional specialization(FSpe),and functional originality(FOri)decreased with species richness,and functional divergence(FDiv)increased first and then *** beta diversity was dominated by its nestedness component,in contrast to the taxonomic *** changes in species richness are more strongly influenced by land-use change under the low-emission scenario,while under the high-emission scenario,they are more strongly influenced by climate *** in functional richness(FRic)were inconsistent with those in species richness,with a magnitude of decreases greater than predicted from species ***,mammal assemblages showed potential functional differentiation(FD)across the country,and the trends exceeded those towards taxonomic differentiation(TD).Our findings help us understand the processes underlying biodiversity responses to global changes on multiple facets and provide new insight for conservation plans.

Uganda has been implementing the Global Health Security Agenda(GHSA)since 2015 to build its capacity according to World Health Organization(WHO)Benchmarks on International Health Regulations *** country remains prone to outbreaks,with more than 20 disease outbreaks reported in the past five years,including Ebola virus disease,Crimean-Congo haemorrhagic fever,Marburg haemorrhagic fever,measles,yellow fever,coronavirus disease 2019(COVID-19),and *** resistance(AMR)is an ongoing *** scored capacity level 3 on infection prevention and control(IPC)and antimicrobial stewardship(AMS)in the 2017 Joint External Evaluation(JEE)*** gaps were being addressed after a self-assessment in *** paper describes the technical assistance approaches provided to Uganda by the Medicines,Technologies,and Pharmaceutical services Program,funded by the United States(U.S.)Agency for International Development,and implemented by Management Sciences for *** program,through a One Health approach,supported systematic capacity strengthening based on the JEE's capacity advancement framework for global health security,specifically relating to *** program's interventions impacted 32 WHO benchmark actions(7 for AMR multisectoral coordination,16 for IPC,and 9 for AMS),contributing to Uganda's strengthened GHSA *** success built on the AMR platform,the program trained 745 health workers in IPC for the Ebola virus and provided support for simulation exercises by eight district IPC *** program also worked with the Ministry of Health to coordinate IPC for the COVID-19 response in five health regions,covering 45 districts and reaching 5,452 health workers at 858 health facilities.

The Flooding Pampa grasslands are the last remnant of the Rio de la Plata grasslands in ***-genic interventions have led to severe degradation and,as a result,the ecosystem services provided by the grass-lands are declining,in terms of provisioning,regulating,and supporting *** synthesized the existing literature on the ecosystem goods and services provided by these grasslands under grazing in different conditions and conservation *** found that plant and animal diversity and primary production are the most studied ecosystem services,while climate regulation,water supply,nutrient cycling,meat production and erosion control,in that order,are less *** services are *** grazing and glyphosate spraying are the main drivers of grassland *** grazing and conservative stocking rates have been shown to reverse degradation and demonstrate that livestock production is compatible with ecosystem conserva-tion by maintaining regulating and provisioning *** these management strategies are poorly integrated,improving their implementation will require important changes in farmers’decisions and the development of policies that create the economic conditions for this to *** is needed to understand the conditions that prevent the knowledge generated from being transferred to producers and translated into practices that would improve the provision of ecosystem services.

Peripartal rumen-protected methionine supplementation to higher energy diets elicits positive effects on blood neutrophil gene networks, performance and liver lipid content in dairy cows

Background： Main objectives were to determine to what extent Smartamine M（SM） supplementation to a prepartal higher-energy diet could alter neutrophil（PMN） and liver tissue immunometabolic biomarkers, and whether those responses were comparable to those in cows fed a prepartal lower-energy diet（CON）.Results： Twenty-eight multiparous Holstein cows were fed CON（NEL= 1.24 Mcal/kg DM） during d-50 to d-22 relative to calving. From d-21 to calving, cows were randomly assigned to a higher-energy diet（OVE, n = 9; NEL= 1.54 Mcal/kg DM）, OVE plus SM（OVE ＋ SM, n = 10; SM = 0.07 % of DM） or remained on CON（n = 9）. Al cows received the same basal lactation diet（NEL= 1.75 Mcal/kg DM）. Supplementation of SM（OVE ＋ SM） continued until 30 d postpartum. Liver biopsies were harvested at d-10, 7, and 21 relative to parturition. Blood PMN isolated at-10, 3, and 21 d relative to calving was used to evaluate gene expression. As expected, OVE increased liver lipid content postpartum; however,cows fed OVE ＋ SM or CON had lower concentrations than OVE. Compared with OVE, cows in CON and OVE ＋ SM had greater DMI postpartum and milk production. Furthermore, cows fed OVE ＋ SM had the greatest milk protein and fat percentage and lowest milk SCC despite having intermediate PMN phagocytic capacity. Adaptations in PMN gene expression in OVE ＋ SM cows associated with the lower SCC were gradual increases from-10 to 21 d in genes that facilitate migration into inflammatory sites（SELL, ITGAM）, enzymes essential for reducing reactive oxygen metabolites（SOD1, SOD2）, and a transcription factor（s） required for controlling PMN development（RXRA）. The greater expression of TLR4 on d 3, key for activation of innate immunity due to inflammation, in OVE compared with CON cows suggests a more pronounced inflammatory state. Feeding OVE ＋ SM dampened the upregulation of TLR4, despite the fact that these cows had similar expression of the pro-inflammatory genes NFKB1 and TNF as OVE. Cows in CON had lower overall expr
