Variation in Bombyx mori immune response against fungal pathogen Beauveria bassiana with variability in cell wallβ-1,3-glucan

Entomopathogenic fungi are protected by a cell wall with dynamic structure for adapting to various environmental conditions.β-1,3-Glucan recognition proteins activate the innate immune system of insects by recognizing surface molecules of fungi.However,the associations between pathogenicity and the different components of entomopathogenic fungal cell walls remain unclear.Three Beauveria bassiana strains were selected that have significantly differing virulence against Bombyx mori.The molecular mechanisms underlying the immune response in B.mori were investigated using RNA sequencing,which revealed differences in the immune response to different B.bassiana strains at 12 h post-infection.Immunofluorescence assays revealed thatβ-1,3-glucan content had an opposite trend to that of fungal virulence.β-1,3-Glucan injection upregulated BmβGRP4 expression and significantly reduced the virulence of the high-virulence strain but not that of the medium-virulence or low-virulence strains.BmβGRP4 silencing in B.mori with RNA interference resulted in the opposite virulence pattern,indicating that the virulence of B.bassiana was affected by the cell walls’content ofβ-1,3-glucan,which could be recognized by BmβGRP4.Furthermore,interference with the gene CnA(calcineurin catalytic A subunit)involved inβ-1,3-glucan synthesis eliminated differences in virulence between B.bassiana strains.These results indicate that strains of a single species of pathogenic fungi that have differing cell wall components are recognized differently by the innate immune system of B.mori.

Background The timing and incidence of recurrent bone metastasis(BM)after radical gastrectomy in patients with gastric cancer(GC)as well as the survival of these patients were not fully understood.The aim of this study was to analyze the data of an observational GC cohort and identify patients who underwent curative gastrectomy and had recurrent BM to describe and clarify the pattern and profile of BM evolution after surgery.Methods Data were retrieved from a hospital-based GC cohort,and patients who underwent upfront radical gastrectomy were selected.The time points of specific organ metastatic events were recorded,and the person-year incidence rate of metastatic events was calculated.The latency period of BM events after gastrectomy was measured and compared with that of the other two most common metastatic events,liver metastasis(LM)and distant lymph node metastasis(LNM),using analysis of variance.Propensity score matching and subgroup analysis were used for sensitivity analysis.Results A total of 1324 GC cases underwent radical gastrectomy between January 2011 and December 2021.Of these,67 BM,218 LM,and 248 LNM occurred before the last follow-up.The incidence of BM events was 1.7/100 person-years,which was approximately 3-fold lower than that of LM and distant LNM events(5.5 and 6.3 per 100 person-years,respectively).BM events had a significantly longer latency(median time,16.5 months)than LM and LNM events(11.1 and 12.0 months,respectively).Recurrent BM led to a worse prognosis(median survival,4.5 months)than those of LM and LNM events(median survival,7.7 and 7.1 months,respectively).However,no difference in overall survival after gastrectomy was observed among the groups.Conclusions Compared with other common metastatic events,BM in GC after gastrectomy is a late-onset event indicating poor survival.

In situ meso-tetra(4-carboxyphenyl) porphyrin ligand substitution in Hf-MOF for enhanced catalytic activity and stability in photoredox reactions

Though there are numerous intrinsic merits of metal-organic frameworks（MOFs）,low charge separation efficiency has imposed heavy restrictions on their photocatalytic application.Herein,in situ porphyrin ligand substitution,as a strategy for improving the charge separation efficiency and increasing the amounts of active sites,has been designed and realized in a Hf-biphenyl dicarboxylic acid（BPDC） MOF.Specifically,a size and geometry matched meso-tetra（4-carboxyphenyl） porphyrin（TCPP）ligand was selected and doped into Hf-BPDC MOF by forming coordinating bonds with Hf centers,forming dualligand Hf-BPDC-TCPP MOF.The resultant Hf-BPDC-TCPP MOF showed significantly improved activity and chemical stability in the photocatalytic H2generation(261 μmol·g-1·h-1) and tetracycline（TC） degradation reactions（95.8%）,which was 48 and 1.47 folds higher than that of the Hf-BPDC MOF.Photophysical and electrochemical studies revealed that the introduction of porphyrin ligand could generate a stronger internal electric field for boosting the charge separation and transfer,increase the specific surface area for providing more active sites,and narrow the band gap to enhance the visible light absorption.This in situ ligand substitution method provides a promising approach to build a tunable platform for constructing high-performance MOF photocatalysts.

Adaptive Sensor-Fault Tolerant control of Unmanned Underwater Vehicles With Input Saturation

This paper investigates the tracking control problem for unmanned underwater vehicles(UUVs)systems with sensor faults,input saturation,and external disturbance caused by waves and ocean currents.An active sensor fault-tolerant control scheme is proposed.First,the developed method only requires the inertia matrix of the UUV,without other dynamic information,and can handle both additive and multiplicative sensor faults.Subsequently,an adaptive fault-tolerant controller is designed to achieve asymptotic tracking control of the UUV by employing robust integral of the sign of error feedback method.It is shown that the effect of sensor faults is online estimated and compensated by an adaptive estimator.With the proposed controller,the tracking error and estimation error can asymptotically converge to zero.Finally,simulation results are performed to demonstrate the effectiveness of the proposed method.

This paper investigates the problem of outlier-resistant distributed fusion filtering(DFF)for a class of multi-sensor nonlinear singular systems(MSNSSs)under a dynamic event-triggered scheme(DETS).To relieve the effect of measurement outliers in data transmission,a self-adaptive saturation function is used.Moreover,to further reduce the energy consumption of each sensor node and improve the efficiency of resource utilization,a DETS is adopted to regulate the frequency of data transmission.For the addressed MSNSSs,our purpose is to construct the local outlier-resistant filter under the effects of the measurement outliers and the DETS;the local upper bound(UB)on the filtering error covariance(FEC)is derived by solving the difference equations and minimized by designing proper filter gains.Furthermore,according to the local filters and their UBs,a DFF algorithm is presented in terms of the inverse covariance intersection fusion rule.As such,the proposed DFF algorithm has the advantages of reducing the frequency of data transmission and the impact of measurement outliers,thereby improving the estimation performance.Moreover,the uniform boundedness of the filtering error is discussed and a corresponding sufficient condition is presented.Finally,the validity of the developed algorithm is checked using a simulation example.

This paper investigates the anomaly-resistant decentralized state estimation(SE) problem for a class of wide-area power systems which are divided into several non-overlapping areas connected through transmission lines. Two classes of measurements(i.e., local measurements and edge measurements) are obtained, respectively, from the individual area and the transmission lines. A decentralized state estimator, whose performance is resistant against measurement with anomalies, is designed based on the minimum error entropy with fiducial points(MEEF) criterion. Specifically, 1) An augmented model, which incorporates the local prediction and local measurement, is developed by resorting to the unscented transformation approach and the statistical linearization approach;2) Using the augmented model, an MEEF-based cost function is designed that reflects the local prediction errors of the state and the measurement;and 3) The local estimate is first obtained by minimizing the MEEF-based cost function through a fixed-point iteration and then updated by using the edge measuring information. Finally, simulation experiments with three scenarios are carried out on the IEEE 14-bus system to illustrate the validity of the proposed anomaly-resistant decentralized SE scheme.

Effect of BaO and MgO on structure and properties of aluminate slag with various CaO/Al_(2)0_(3)ratios

To provide theoretical guidance for performance stability control of low-reactive mold fluxes,the effects of BaO and MgO on the structure and properties of aluminate slag with various CaO/Al_(2)0_(3)(C/A)ratios were investigated using the Fourier transform infrared spectrometer,Raman spectroscope,hemispherical melting point instrument,rotational viscometer and X-ray diffractometer.The results indicated that with BaO and MgO addition,the structure polymerization was first weakened and then enhanced at C/A of 1.1,and the transition contents corresponded to 8 wt.%BaO and 2 wt.%MgO,respectively,while the structure polymerization decreased continuously at C/A of 1.3.Since the viscosity change was well consistent with the structure evolution,the polymerization degree played a more prominent role in the slag viscosity than superheat degree when the melting temperature difference was within 40℃.The break temperature decreased initially and then increased with augment of BaO and MgO at C/A of 1.1,while it manifested a decrease trend with BaO addition,and it decreased obviously but then turned to increase with MgO increment at C/A of 1.3.The crystallization phase and crystallization ratio kept stable with BaO increment,while the crystallization ratio rose greatly with MgO promoting LiAlO_(2)precipitation at C/A of 1.1.The crystal types of all experimental slags were mainly Ca_(12)Al_(14)O_(33)and CaF_(2)at C/A of 1.3,and the precipitation of crystalline phase BaAl2O4 demonstrated a rising trend,while that of Ca_(12)Al_(14)O_(33)gradually declined with BaO augment.

Photocatalytic in situ H2O2 production and activation for enhanced ciprofloxacin degradation over CeO2-Co3O4/g-C3N4:key role of CeO2

The abused ciprofloxacin antibiotics have caused significant environmental damage.Although oxidative degradation of ciprofloxacin exhibits promising efficacy,it often entails excessive energy consumption.Hence,it is necessary to explore an effective and ecologically sustainable degradation strategy.Herein,we demonstrated that g-C3N4decorated with the coordinated CeO2and Co3O4（CeO2-Co3O4/CN） exhibited effective ciprofloxacin photodegradation via in situ H2O2production and activation mechanism.Results indicate that the introduced CeO2enhances the transference of photogenerated electrons to O2by adjusting the oxygen vacancy of photocatalyst,thereby increasing the generation of superoxide radicals,which in turn generate H2O2,resulting in a 22.4-fold increase in H2O2generation over g-C3N4.Moreover,the in situ H2O2generation facilitated by CeO2is confirmed to be essential for ciprofloxacin degradation via CeO2-Co3O4/CN,as it provides enough oxidant for Co3O4to activate into hydroxyl radicals for the pollutants degradation.Ultimately,CeO2-Co3O4/CN achieves a ciprofloxacin degradation ratio of 97.7% within 80 min.This study introduces a novel approach that combines H2O2generation and activation,offering an innovative perspective for achieving clean and efficient purification of antibiotic-contaminated water.

Reversed charge transfer induced by nickel in Fe-Ni/Mo_(2)C@nitrogen-doped carbon nanobox for promoted reversible oxygen electrocatalysis

The interaction between metal and support is critical in oxygen catalysis as it governs the charge transfer between these two entities,influences the electronic structures of the supported metal,affects the adsorption energies of reaction intermediates,and ultimately impacts the catalytic performance.In this study,we discovered a unique charge transfer reversal phenomenon in a metal/carbon nanohybrid system.Specifically,electrons were transferred from the metal-based species to N-doped carbon,while the carbon support reciprocally donated electrons to the metal domain upon the introduction of nickel.This led to the exceptional electrocatalytic performances of the resulting Ni-Fe/Mo_(2)C@nitrogen-doped carbon catalyst,with a half-wave potential of 0.91 V towards oxygen reduction reaction(ORR)and a low overpotential of 290 m V at 10 mA cm^(-2)towards oxygen evolution reaction(OER)under alkaline conditions.Additionally,the Fe-Ni/Mo_(2)C@carbon heterojunction catalyst demonstrated high specific capacity(794 mA h g_(Zn)~(-1))and excellent cycling stability(200 h)in a Zn-air battery.Theoretical calculations revealed that Mo_(2)C effectively inhibited charge transfer from Fe to the support,while secondary doping of Ni induced a charge transfer reversal,resulting in electron accumulation in the Fe-Ni alloy region.This local electronic structure modulation significantly reduced energy barriers in the oxygen catalysis process,enhancing the catalytic efficiency of both ORR and OER.Consequently,our findings underscore the potential of manipulating charge transfer reversal between the metal and support as a promising strategy for developing highly-active and durable bi-functional oxygen electrodes.

GRK2 inhibits Flt-1^(+)macrophage infiltration and its proangiogenic properties in rheumatoid arthritis

Rheumatoid arthritis(RA)is an autoimmune disease with a complex etiology.Monocyte-derived macrophages(MDMs)infiltration are associated with RA severity.We have reported the deletion of G-protein-coupled receptor kinase 2(GRK2)reprograms macrophages toward an anti-inflammatory phenotype by recovering G-protein-coupled receptor signaling.However,as more GRK2-interacting proteins were discovered,the GRK2 interactome mechanisms in RA have been understudied.Thus,in the collagen-induced arthritis mouse model,we performed genetic GRK2 deletion using GRK2^(f/f)Lyz2-Cre^(+/−)mice.Synovial inflammation and M1 polarization were improved in GRK2^(f/f)Lyz2-Cre^(+/−)mice.Supporting experiments with RNA-seq and dual-luciferase reporter assays identified peroxisome proliferator-activated receptorγ(PPARγ)as a new GRK2-interacting protein.We further confirmed that fms-related tyrosine kinase 1(Flt-1),which promoted macrophage migration to induce angiogenesis,was inhibited by GRK2-PPARγsignaling.Mechanistically,excess GRK2 membrane recruitment in CIA MDMs reduced the activation of PPARγligand-binding domain and enhanced Flt-1 transcription.Furthermore,the treatment of mice with GRK2 activity inhibitor resulted in significantly diminished CIA pathology,Flt-1^(+)macrophages induced-synovial inflammation,and angiogenesis.Altogether,we anticipate to facilitate the elucidation of previously unappreciated details of GRK2-specific intracellular signaling.Targeting GRK2 activity is a viable strategy to inhibit MDMs infiltration,affording a distinct way to control joint inflammation and angiogenesis of RA.
