The ubiquitous lipid-derived molecules N-acylethanolamines(NAEs)have multiple immune functions in mammals,but their roles and mechanisms in plant defense response during changing environment remain largely unclear.Here,we found that exogenous NAE18:0 and NAE18:2 promoted defense against the necrotrophic pathogen Botrytis cinerea but suppressed defense to the hemi-biotrophic pathogen Pseudomonas syringae pv.tomato(Pst)DC3000 in tomato.The knocking-down and overexpression function analysis of the pathogen-responsive NAE synthetic gene PHOSPHOLIPASE Dγ(PLDγ)and hydrolytic gene FATTY ACID AMID HYDROLASE 1(FAAH1)revealed that the NAE pathway is crucial for plant defense response.Using exogenous applications and SA-abolished NahG plants,we unveiled the antagonistic relationship between NAE and SA in plant defense response.Elevated CO_(2) and temperature significantly changed the NAE pathway in response to pathogens,while inhibition of the NAE pathway led to the alternation of environment-mediated defense variations against Pst DC3000 in tomato,indicating that NAE pathway is associated with plant defense variations in response to elevated CO_(2) and temperature.The results herein reveal a new function of NAE in plant defense,and its involvement in environment-mediated defense variation in tomato.These findings shed light on the NAE-based plant defense,which may have relevance to crop disease management in future changing climate.

Soil pollution by diesel fuels is a worldwide environmental problem,but little research has been carried out into on-site techniques for remediation of soil polluted by waste solvents.This study compared chemical oxidation and soil washing methods for their efficiency and environmental and economic impacts.Soil was spiked with 0#diesel to simulate an actual pollution level of about 1260 mg/kg total petroleum hydrocarbon(TPH).Fenton-like oxidation eliminated 90.4%of the TPH with a Fe2+׃H2O2 ratio of 1:10 in 5 d compared with 25.8%removal by the activated persulfate method under the same conditions.In washing tests,sodium dodecylbenzenesulfonate and Tween 80 were both unsuitable for TPH washing,while ultrapure water removed 36.1%of TPH in 75 min.Only the Fenton-like oxidation technique met remediation goals based on the screening values of the Guideline for Risk Assessment of Contaminated Sites.The environmental impact and economic assessment of techniques demonstrated the superiority of water washing for dealing with low-degree TPH contamination.

Circular RNA(circRNA)is a special type of non-coding RNA that participates in diverse biological processes in both animals and plants.Five years ago,we developed a comprehensive plant circRNA database(PlantcircBase),which has attracted much attention from the plant circRNA community.Here,we report an updated PlantcircBase(v.7.0),which contains 171,118 circRNAs from 21 plant species.Over 31,000 of the circRNAs have full-length sequences constructed based on analysis of 749 bulk RNA sequencing(RNAseq)datasets downloaded from the public domain and Nanopore long-read sequencing results of rice RNAs newly generated in this study.A plant multiple conservation score(PMCS),based on the conservation of both sequence and expression profiles,was calculated for each circRNA to quantify and compare the conservation of all circRNAs.A new parameter,plant circRNA confidence level(PCCL),is introduced to measure the identity reliability of each circRNA based on experimental validation results and the number of references that support the circRNA.All this information and other details of circRNAs can be browsed,searched,and downloaded from PlantcircBase 7.0,which also provides online bioinformatics tools for visualization and sequence alignment.PlantcircBase 7.0 is publicly and freely accessible at http://***/plantcircbase/.

研究目的:开发多态性高的基因组简单重复序列(SSR)标记,为杨梅遗传多样性、遗传图谱构建及品种鉴定相关研究奠定基础。创新要点:从杨梅全基因组鸟枪法测序数据中新开发了107个SSR标记,并应用于品种间遗传多样性分析及品系鉴定。研究方法:应用生物信息学软件从基因组测序数据中搜索SSR位点并设计相应引物,添加通用的M13尾巴和荧光标识筛选多态性高的SSR标记。重要结论:开发的107个基因组SSR标记可以广泛应用于杨梅种质资源和遗传育种研究,鉴定出三个杨梅新品系。

Piriformospora indica, a root-colonizing endophytic fungus of Sebacinales, promotes plant growth and confers resistance against biotic and abiotic stresses. In order to confirm the influence of P. indica on growth, proline, malondialdehyde（MDA）, chlorophyll, and cadmium（Cd） amounts in Nicotiana tabacum under Cd stress, hydroponics, pot and field trials were conducted. The results showed that P. indica can store Cd in plant roots and reduce leaf Cd content, reduce the concentration of MDA, and increase the proline and chlorophyll content and the activities of catalase, peroxidase, and superoxide dismutase under hydroponic Cd stress. RT-PCR analysis showed that the relative expression level of genes Gsh2, Ta PCS1, oas1, GPX, and Hsp70 in colonized plants was 4.3, 1.4, 2.9, 1.7, and 6.9fold higher than in un-colonized plants respectively. Cd exposure significantly reduced un-colonized plants＇ agronomic traits compared to P. indica-colonized ones. Our results suggested that P. indica can sequester Cd in roots, so that much less cadmium was transported to leaves, and the increased concentrations of antioxidant enzymes, pigments and proline contents, as well as the higher expression of stress-related phytochelatin biosynthesis genes in P. indica-inoculated plants, may also serve to protect N. tabacum plants against oxidative damage, enhancing Cd tolerance.

Characterization and quantification of two acylated flavonol glycosides from Camellia sinensis and their antibacterial effect on oral pathogens

Viralinfection usually induces remodeling of host cellular membranes and organelles,especially endoplasmic reticulum（ER） for generating membrane packed viral factory,in which viruses replicate and transcribe its hereditary nucleic acids,translate into proteins,and then assemble proteins and nucleic acids into virion.During the infection of bymoviruses,a kind of membranous bodies（MBs）are generated in host cells,which have thought to be as ER aggregates（Hibino et al.,1981）.So far,studies on bymovirus-induced MBs usually focused on their viral components（Schenk et al.,1993;Sun et al.,2014）,but less on its membrane origination and structure.In this study,MBs induced by Wheat yellow mosaic virus（WYMV） were intensively investigated with several transmission electron microscopy（TEM）-based methods,including cytological observation,component analysis by immunogold labeling and structural analysis by electron tomography（ET）.WYMV infection induced at least two morphologies of MB,including the lamella dominated morphology（lamella-MB） looked like sprawling cirrus,and the tubule dominated morphology（tubule-MB） looked like latticed network.MBs were verified composing of ER as revealed by immuno-gold labeling by antibody against endoplasmic reticulum（ER） retention signal as well as by detailed observation of MB construction modules as double layer membrane.By immuno-gold labeling,both two MB morphologies（lamella-MB and tubule-MB） had same components in viral derived protein and membrane origination（from ER）.Structural analysis by ET reconstruction revealed the organization of ER in MBs.Lamella-MBs were composed of cesER like structures arranged irregularly whereas tubule-MBs were composed of tubER like structures arranged regularly.This study provided insights into the structural details in how Bymovirus utilizing host membrane system.

Acylated flavonol glycosides(AFGs)are a new group of flavonoids with multiple pharmacological functions.In the present work,two AFGs were isolated with a combined chromatography technology from Camellia sinensis and identified as quercetin tetraglycosides with one p-coumaryl moiety(F1)and kaempferol tetraglycosides with one p-coumaryl moiety(F2)by ultra-performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS)and nuclear magnetic resonance(NMR).F1 and F2 with high purity and quantity can be obtained through relatively simple steps,which is of great significance in industrial production and application.The quantification of two AFGs in different tea cultivars and different season samples was conducted using the high-performance liquid chromatography(HPLC)method.Among fifty tested tea cultivars,the contents of F1 and F2 were extremely high in'Zhenghe Dabaicha'and'Wannong 95',but these were undetectable in some tea cultivars.Relatively high contents of AFGs were found in autumn harvested tea samples which might be related to sunlight irradiation.Further in vitro experiments showed that F1 and F2 monomers were effective on inhibiting growth and biofilm development of Porphyromonas gingivalis and Fusobacterium nucleatum.This antibacterial activity was proven to involve the chelation of iron.These results provide useful information on AFGsrich tea resources and new insights of AFGs into effective agents against oral diseases.

The reclamation of tidal flats has been one of the important approaches to replenish the arable lands in the coastal areas; pollution status of reclaimed soils has received wide attention recently, especially for the study of heavy metals due to the relative high pollutant concentrations in wetlands. To understand the impact of land use change on heavy metal and arsenic(As) geochemistry by the reclamation of wetlands for agriculture, surface soils and soil profiles were collected from the agricultural land reclaimed in the 1990 s and the intertidal flat wetland at Dongtan on Chongming Island in the Yangtze River Estuary, China. The soil samples were analyzed for total concentrations and chemical speciation of chromium(Cr), zinc(Zn), copper(Cu), lead(Pb), nickel(Ni), cadmium(Cd) and As using inductively coupled plasma mass spectrometry(ICP-MS). Results showed that soil properties(salinity, total organic carbon and grain-size distribution) and the concentrations of heavy metals and As in the soils differed under the different land use types. The conversion of wetland to forest had caused obvious losses of all the measured heavy metals. In paddy field and dryland with frequent cultivation, the concentrations of Cr, Zn, Cu, Ni and As were higher when compared to forest land which was disturbed rarely by human activities. Speciation analysis showed that Cr, Zn, Cu, Ni and As were predominated by the immobile residual fraction, while Pb and Cd showed relatively higher mobility. In general, metal(except Ni) and As mobility decreased in the following order: wetland> dryland ≥ paddy field > forest land, which suggested that the reclaimed soils had lower metal and As mobility than the intertidal flat wetland. The results of this study contribute to a better understanding of the effects of land use on heavy metals and As in the reclaimed soils of the study area and other similar coastal areas.

Characterization of the temperature and oxygen concentration field in a decentralized solid‑waste incinerator for villages and towns

Decentralized solid-waste incinerators(DSWIs)have certain advantages for waste disposal from villages and towns.However,the incineration condition is always affected by the distribution of temperature and oxygen concentration,which causes difficulties in operation and maintenance.In this study,the temperature and oxygen concentration distribution of DSWI were characterized using different air flow rates and bottom ash volumes.The results showed that the adjustment of air flow has no significant influence on the heating process of the DSWI,while the retention of bottom ash did affect the temperature and oxygen concentration fields in the furnace.When the air flow rate was increased without the retention of bottom ash,99%of the furnace volume temperature was observed between 780℃ and 800℃.However,once the bottom ash was retained,the whole furnace temperature was steadily maintained between 800℃ and 850℃.When the air flow rate was increased without bottom ash,the highest furnace volume percentage of oxygen concentrations higher than 3%maxed out at 11%volume,while it could reach 100%when bottom ash remained.The distribution of the temperature and oxygen concentration in the DSWI characterized by this research provides strong support for the operation and management of such systems.

Occurrence, distribution and risk assessment of antibiotics at various aquaculture stages in typical aquaculture areas surrounding the Yellow Sea

The pollution of antibiotics commonly existed throughout the entire aquaculture process,but the residues of antibiotics at different aquaculture stages have rarely been studied.This study investigated the occurrence,distribution and risk assessment of antibiotics at different aquaculture stages (the non-aquaculture stage,the early aquaculture stage,the middle aquaculture stage,and the late aquaculture stage) in two typical marine aquaculture areas(Mahegang River and Dingzi Bay) surrounding the Yellow Sea.Fluoroquinolones and tetracyclines were commonly used antibiotics in the aquaculture of these areas with high detection frequencies (17%to 83%).Compared among four aquaculture stages,the highest concentration of antibiotics (9032.08 ng/L) in aquaculture ponds was detected at the late aquaculture stage.And the antibiotic pollution level of natural water was directly related to the aquaculture stages.Similarly,at the aquaculture stages,the detection frequency of antibiotics in sediments was higher than that at the non-aquaculture stage.Based on the correlation analysis,the concentration of main antibiotics in water showed a positive correlation with total nitrogen (p<0.05) and chlorophyll a (p<0.01),while it showed a negative correlation with salinity (p<0.01) in coastal water of the Dingzi Bay.According to the risk assessment,with the development of aquaculture stages,the selection pressure of fluoroquinolones and tetracyclines on resistant bacteria had increased.And the ecological risks caused by sulfonamides and tetracyclines to aquatic organisms had also increased markedly.Overall,this study may provide a reference for formulating regulatory policies regarding antibiotic use at different aquaculture stages.
