A novel structure of ultra-high-loading small moleculesencapsulated ZIF-8 colloid particles

The safety of nanoparticle-based drug delivery systems(DDSs)for cancer treatment is still a challenge,restricted by the intrinsic cytotoxicity of drug carriers and leakage of loaded drug.Here,we propose a novel nanocarrier’s cytotoxicity avoidance strategy by synthesizing an encapsulation core–shell structure of zeolitic imidazolate framework-8(ZIF-8)-based colloid particles(CPs)with an amorphous ZIF-8 skin.This encapsulation structure achieves an ultra-high loading rate(LR)of 90%(i.e.,9 mg doxorubicin(DOX)per 1 mg ZIF-8)for DOX and the protection of DOX from leaking.Notably,to deliver unit-dose drug,this ultra-high LR of 90%significantly reduces the usage of ZIF-8 to 1.2%(2 orders of magnitude)compared to that of DOX@ZIF-8 with a 10%LR,in which cytotoxicity of ZIF-8 could well below the safety limit and then be relatively ignored.Safety,drug delivery efficacy,scale-up ability,and universality of this encapsulation structure have been further verified.Our findings suggest the great potential of this ZIF-8-based encapsulation core–shell structure in the field of drug delivery.

In order to reduce or eliminate the adverse effects of calcium impurity,the interaction of Ca and rare earth Y co-doping on the preparation of WC-Co cemented carbides was investigated.X-ray diffraction,scanning electron microscopy and transmission electron microscopy were employed to analyze the phase compositions,particle sizes and morphologies of them,respectively.The results show that the second type of"yttrium barrier effect"is observed and the adverse influence of Ca-rich phase on tungsten powder can be eliminated by it.The flexural strength and fracture toughness of YG6-0.01%Ca+0.09%Y alloy are 2199.5 MPa and 11.49 MPa·m^(1/2),showing improvement of 6.5% and 2.7% compared to YG6 alloy,respectively.The evolutions of Ca-rich phase and Y-rich phase at the every alloy preparation stage of WO_(3),W,WC and cemented carbides are obtained.Furthermore,the strengthening mechanism of WC-Co cemented carbide with co-addition of Ca and Y is proposed.

Dear editor,Patients with clear-cell renal cell carcinoma(ccRCC)potentially have a high recurrence rate of more than 40%[1].Patients receive radical or partial nephrectomy,sometimes assisted by targeted therapies or/and immune checkpoint inhibitors(ICIs),to improve overall survival(OS)[2].Large-cohort studies and large-scale human tissue sequencing have provided a series of molecular subtyping methods for precision medicine-based ccRCC risk stratification and therapeutic regimens,such as ClearCode34,the prognostic risk predictor[3],and seven subsets for ICI and angiogenesis blockade outcomes[4].However,these subtyping results are all based on human genomes without including the host microbiota,which may be non-negligible genome components.

Most existing treatments for origami-folding simulations have focused on regular-shaped configurations.This article aims to introduce a general strategy for simulating and analyzing the deformation process of irregular shapes by means of computational capabilities nowadays.To better simulate origami deformation with folding orders,the concept of plane follow-up is introduced to achieve automated computer simulation of complex folding patterns,thereby avoiding intersection and penetration between planes.Based on the evaluation criteria such as the lowest storage energy with tightening and the fastest pace from tightening to unfolding,the optimal crease distribution patterns for four irregular(‘N’-,‘T’-,‘O’-,and‘P’-shaped)origami configurations are then presented under five candidates.When the dimensions of the origami are fixed,it is discovered that simpler folding patterns lead to faster deformation of the origami configuration.When the folding complexity is fixed,higher strain energy results in more rapid origami expansion.

To explore the response law of non-lethal large-size kinetic energy projectiles to blunt attack on skin tissue,and to evaluate the skin injury characteristics of the attacked personnel and the use safety of kinetic energy projectiles.Based on the LS-DYNA simulation software,a three-layer skin simulation model and a Flash-Ball rubber bullet model are established,and the force-time and deformation-time biomechanical corridors of the Flash-Ball rubber bullet impacting human skin tissue are obtained.The corridor curve and the energy transfer and diffusion are analyzed and compared.The safety evaluation of the damage caused by the rubber bullet shooting a human body at different distances is carried out using the empirical formula of the penetration limit.Finally,the safe shooting distance is obtained.The results show that the model used in the simulation has a good correlation with the experimental data,its biomechanical corridor characteristics are different from those of conventional vehicle impact and smallsize projectile response characteristics.The energy transfer and action time of medium and low-speed impact may cause greater damage.The fat layer is the largest energy absorption unit.The minimum safe shooting distance to ensure skin tissue from penetrating damage is 15.8 m,and the limit specific kinetic energy of skin damage is 7.88 J/cm^(2).This study can be extended to the study of biomechanical response law and safety evaluation under the impact of the same type of large kinetic energy projectile,which provides an important theoretical reference for the police to use large kinetic energy projectiles to conduct safe shooting in peacekeeping operations.

Matrix-assisted laser desorption/ionization(MALDI)mass spectrometry(MS)plays an indispensable role in analyzing protein covalent structures.The reliable identification of amino acid residues and modifications relies on the mass accuracy,which is highly dependent on calibration.However,the accuracy provided by the currently available calibrants still needs further improvement in terms of compatibility with multiple tandem MS modes or ion polarity modes,calibratable range,and minimizing suppression of and interference with analyte signals.Here aiming at developing a versatile calibrant to solve these problem,we designed a synthetic peptide format of calibrant R_x(GDP_n)_m(referred to as“Gly-Asp-Pro,GDP”)according to the chemical natures of amino acids and polypeptide fragmentation rules in tandem MS.With four types of amino acid residues selected and arranged through rational designs,a GDP peptide produces highly regulated fragments that give rise to evenly spaced signals in each tandem MS mode and is compatible with both positive and negative ion modes.In internal calibration,its regulated fragmentation pattern minimizes interference with analyte signals,and using a single peptide as the input minimizes suppression of the analyte signals.As demonstrated by analyses of proteins including monoclonal antibody and Aβ-42,these features allowed significant increase of the mass accuracy and precision,which improved sequence coverage and sequence resolution in sequence analyses(including de novo sequencing).This rational design strategy may also inspire further development of synthetic calibrants that benefit structural analysis of biomolecules.

OBJECTIVE To evalu ate the role of resting magnetocardiography in identifying seve re coronary artery stenosis in patients with suspected coronary artery disease.METHODS A total of 513 patients with angina symptoms were included and divided into two groups based on the extent of coronary artery disease determined by angiography:the non-severe coronary stenusis group(analysis were performed to investigate the clinical utility and performance of the models,respectively.RESULTS In the severe coronary stenosis group,QR_MCTDd,S_MDp,and TT_(MA)C_(50) were significantly higher than those in the non-severe coronary stenosis group(10,46±10.66 vs,5.11±6.07,P analysis showed that the c ombine d model provided greater net benefit compared to the magnetocardingraphy model.CONCLUSIONS The novel quantitative MFM parameters

Persulfate is considered a convenient and efficient remediation agent for organic contaminated soil.However,the potential risk of sulfur into the soil remediation by persulfate remains ignored.In this study,glass bottles with different persulfate dosages and groundwater tables were set up to simulate persulfate remediation of organic pollutants(aniline).The results found sulfate to be the main end-product(83.0%–99.5%)of persulfate remediation after10 days.Moreover,H_(2)S accounted for 93.4%–99.4%of sulfur reduction end-products,suggesting that H_(2)S was the final fate of sulfur.H_(2)S was released rapidly after one to three days at a maximum concentration of 33.0 ppm,which is sufficient to make a person uncomfortable.According to the fitted curve results,H_(2)S concentration decreased to a safe concentration(0.15 ppm)after 20–85 days.Meanwhile,the maximum concentration of methanethiol reached 0.6 ppm.These results indicated that secondary pollution from persulfate remediation could release harmful gases over a long time.Therefore,persulfate should be used more carefully as a remediation agent for soil contamination.

Effect of Central Multiphase Microstructure of Thick Plates on Work Hardening and Crack Propagation

The effect of multiphase microstructure in the center segregation zones(CSZ)of high-strength steel thick plates on work hardening characteristics and crack initiation/propagation were studied.Microstructure was correlated with deformation damage behavior by using Taylor factor(TF)gradients,kernel average misorientation(KAM),and geometric necessary dislocation(GND)density.The results show that the segregation leads to a mixed structure of ferrite(soft phase)and martensite/bainite(hard phase)in the center of the thick plate.Compared to the 1/4 thickness(1/4 T)region,grain refinement occurred in the CSZ,with KAM values increasing from 0.24 to 0.49 and a decrease in the proportion of high-angle grain boundaries(>15°)from 77.4 to 51.8%.In the process of deformation,due to the influence of grain refinement and adjacent martensite/bainite,ferrite grains were divided into structural units with different TF values.The higher KAM value and GND density at the interface between soft/hard phases resulted in severe work hardening.In addition,the presence of micron-sized inclusions in the CSZ caused local strain concentration and non-continuous deformation of the matrix,which induced crack initiation.The fracture surface showed a mixed mode of brittle cleavage fracture and ductile fracture.
