Comparative Transcriptome Analysis Reveals Different Host Cell Responses to Acute and Persistent Foot-and-Mouth Disease Virus Infection

Foot-and-mouth disease virus(FMDV) rapidly causes cytopathic effects in susceptible cells. Incomplete viral clearance during the acute infection leads to persistent infection. The relationship between host gene expression and the persistent infection remains unclear. In this study, we analyzed the transcriptome profiles of BHK-21 cells acutely and persistently infected with FMDV to identify differences in gene expression. GO and KEGG enrichment analyses showed that the 8,378 differentially expressed genes were significantly enriched in categories including metabolism, biosynthesis, ribosome function, and endocytosis. In persistently infected BHK-21 cells, ribosome-and translation-related genes were significantly down-regulated. There were more differentially expressed immune-related genes during persistent infection than during acute infection. Two hundr.d and seventy-four genes were differentially expressed in both acutely and persistently infected BHK-21 cells. Among these genes, heat shock protein family B member 1(Hspb1) knockdown significantly inhibited FMDV replication. Our research provides a basis for further research to understand the mechanisms of persistent FMDV infection including the genes involved in FMDV replication.

A 12-year-old boy was seen with a discharging ulcer over the thoracic spine on July 5, 2002. The boy was apparently well 4 days before, when his mother accidentally noticed a small boil on the middle of the spine, surrounded by erythema and edema. The swelling increased until it burst 1 day prior to the examination, emitting a purulent discharge. The patient had fever with profuse sweating. There was no history of cough or expectoration, nausea, and/or vomiting. There was a history of recurrent chest pain, which was a consequence of pulmonary pathology, as the boy had been taking treatment for a recurrent chest infection for the past 4 years. The parents had also observed a loss of appetite and weight. The child was never vaccinated with bacillus Calmette-Gué rin (BCG) . His grandfather had succumbed to pulmonary tuberculosis. A history of tuberculosis was elicited in a family friend, as well as in the next-door neighbors. Examination of the skin surface on the back revealed the presence of a 1.5 cm × 1.5 cm ulcer. It had a purulent discharge. The ulcer was angry looking and had everted, undermined, irregular borders. It was tender to the touch and surrounded by erythema and edema. The ulcer was located at the dorso-lumbar spine (Fig. 1a,b). Smear(s) were prepared from the purulent discharge and were stained for the demonstration of acid-fast bacilli (AFB) with Ziehl-Neelsen stain; they were negative on three occasions. culture result of the purulent discharge showed no growth after 48 h of incubation at 37 ° C aerobically. Also, hematoxylin and eosin-stained sections prepared from the representative margin of the ulcer were studied to illustrate its microscopic pathology. It was characterized by a bizarre collection of epitheliold cells and partial foreign body giant cell formation (Fig. 2a,b). Objective: To assess the effect of using 2 throat swabs vs 1 on rapid detectio n of group A streptococcus by the STREP A OIA MAX (hereafter, OIAMAX) test. Meth ods: Childr.n aged 5 to 18 years with acute pharyngitis were randomizedto 1 of 2 study groups. In group 1, one throat swab was obtained, streaked first on sheep blood agar, and then used for OIA MAX testing. In group 2, two throat swabs wer e obtained simultaneously. One swab was streaked first on sheep blood agar and t hen joined with the other swab for OIA MAX testing. In both groups, the pledgets in the collection-transport tube were incubated in Todd-Hewitt broth. A posit ive group A streptococcus culture either by sheep blood agar or Todd-Hewitt bro th was confirmed by a latex agglutination test. Results: Three hundr.d sixtythre e patients were enrolled, 177 in group 1 and 186 in group 2. cultures were posit ive for group A streptococcus in 154 (42.4%) of 363 patients. The sensitivity a nd specificity of OIA MAX testing were 94.7%and 100.0%, respectively, in group 1, and 92.4%and 96.3%, respectively, in group 2. There was no statistical dif ference between the sensitivity, the specificity, and the predictive values of t he OIA MAX test performed with 1 swab compared with those performed with 2 swabs (P>.10). There was no association between OIA MAX test sensitivity and the seve rity of pharyngitis as measured by the modified Centor criterion (history of fev er, absence of cough, presence of pharyngeal or tonsillar exudates, and presence of cervical lymphadenopathy) scores. Conclusions: The OIA MAX test yielded comp arable sensitivity and specificity in both study groups. The use of 2 throat swa bs instead of 1 swab did not increase the sensitivity of the OIA MAX test. The p erformance of the OIA MAX test did not depend on the severity of pharyngitis.

OBJECTIVE: To estimate whether the rates of recovery of group B streptococci from combined vaginal and perianal cultures and combined vaginal and rectal cultures are equivalent. METHODS: We performed a prospective cohort study of vaginal-perianal versus vaginal-rectal culture for group B streptococci. Two hundr.d pregnant women in the third trimester were enrolled. Three specimens were collected from each patient in the following order: lower third of the vagina, perianal skin, and rectum. Each specimen was cultured serially in selective broth media, then on sheep’s blood agar. Suspicious colonies were confirmed by latex agglutination. culture positivity rates from the combined sites of vagina and perianal skin were compared with vagina and rectum. Laboratory personnel were blinded to the collection site of each individual swab. RESULTS: Of the 200 subjects, 71 (36%) had a positive culture from at least 1 site. Vaginal culture was positive in 55 patients (28%), compared with 48 patients (24%) with positive perianal cultures and 50 patients (25%) with positive rectal cultures. Results of combined vaginal and perianal cultures were positive in 68 patients (34%); results of combined vaginal and rectal cultures were positive in 67 patients (34%) (P =1.0). CONCLUSION: The group B streptococci detection rate from vaginal-perianal specimens is not significantly different from the detection rate from vaginal-rectal specimens. Therefore, pregnant women do not need to be subjected to the discomfort of collection of a rectal specimen.

Fungal diversity notes is one of the important journal series of fungal taxonomy that provide detailed descriptions and illustrations of new fungal taxa,as well as providing new information of fungal taxa *** article is the 11th contribution to the fungal diversity notes series,in which 126 taxa distributed in two phyla,six classes,24 orders and 55 families are described and *** in this study were mainly collected from Italy by Erio Camporesi and also collected from China,India and Thailand,as well as in some other European,North american and South american *** described in the present study include two new families,12 new genera,82 new species,five new combinations and 25 new records on new hosts and new geographical distributions as well as sexual-asexual *** two new families are Eriomycetaceae(Dothideomycetes,family incertae sedis)and Fasciatisporaceae(Xylariales,Sordariomycetes).The twelve new genera comprise Bhagirathimyces(Phaeosphaeriaceae),Camporesiomyces(Tubeufiaceae),Eriocamporesia(Cryphonectriaceae),Eriomyces(Eriomycetaceae),Neomonodictys(Pleurotheciaceae),Paraloratospora(Phaeosphaeriaceae),Paramonodictys(Parabambusicolaceae),Pseudoconlarium(Diaporthomycetidae,genus incertae sedis),Pseudomurilentithecium(Lentitheciaceae),Setoapiospora(Muyocopronaceae),Srinivasanomyces(Vibrisseaceae)and Xenoanthostomella(Xylariales,genera incertae sedis).The 82 new species comprise Acremonium chiangraiense,Adustochaete nivea,Angustimassarina camporesii,Bhagirathimyces himalayensis,Brunneoclavispora camporesii,Camarosporidiella camporesii,Camporesiomyces mali,Camposporium appendiculatum,Camposporium multiseptatum,Camposporium septatum,Canalisporium aquaticium,Clonostachys eriocamporesiana,Clonostachys eriocamporesii,Colletotrichum hederiicola,Coniochaeta vineae,Conioscypha verrucosa,Cortinarius ainsworthii,Cortinarius aurae,Cortinarius britannicus,Cortinarius heatherae,Cortinarius scoticus,Cortinarius subsaniosus,Cytospora fusispor
